Many classes of environmental contaminants affect the reproductive function of animals through interactions with the endocrine system. The primary components affected by endocrine active compounds (EACs) are the steroid receptors and the enzymes responsible for steroidogenesis. This study sought to develop an in vitro model for assessing EAC effects in sea turtles by examining their ability to alter cytochrome P450 aromatase (CYP19) activity. Aromatase is the enzyme responsible for the conversion of testosterone to estradiol. This enzyme is critical in the sexual differentiation of reptiles which demonstrate temperature-dependent sex determination. An immortal testis cell line GST-TS from a green sea turtle was grown in culture at 30 degrees C in RPMI 1640 media. The cells were exposed to three known aromatase inducers; dexamethasone (Dex), 8Br-cyclic AMP, or human chronic gonadotropin (HCG) and one aromatase inhibitor 4-androstenol-dione (4-OHA). In addition, the GST-TS cells were exposed to 0.1-30 microM atrazine and 3-100 microM 4,4'-DDE. The inducing compounds that have been shown to increase aromatase activity in other systems failed to induce aromatase activity in the GST-TS cells, yet exposure to the inhibiting compound, 4-OHA, did result in a significant reduction. Atrazine (0.1, 1.0 and 10 microM) significantly induced aromatase activity following a 24 h exposure, and 4,4'-DDE inhibited the activity but only at cytotoxic concentrations (100 microM). Based on these results, this in vitro model can be useful in examining the endocrine effects of EACs in sea turtles. 相似文献

Modelling and experimental study of coupled exchange of p,p′‐DDE and colloids in a flume simulated stream–streambed system 下载免费PDF全文
Stream–subsurface exchange plays a significant role in the fate and transport of contaminants in streams. It has been modelled explicitly by considering fundamental processes such as hydraulic exchange, colloid filtration, and contaminant interactions with streambed sediments and colloids. The models have been successfully applied to simulate the transport of inorganic metals and nutrients. In this study, laboratory experiments were conducted in a recirculating flume to investigate the exchange of a hydrophobic organic contaminant, p,p′‐dichloro‐diphenyl‐dichloroethane (DDE), between a stream and a quartz sand bed. A previously developed process‐based multiphase exchange model was modified by accounting for the p,p′‐DDE kinetic adsorption to and desorption from the bed sediments/colloids and was applied to interpret the experimental results. Model input parameters were obtained by conducting independent small‐scale batch experiments. Results indicate that the immobilization of p,p′‐DDE in the quartz sand bed can occur under representative natural stream conditions. The observed p,p′‐DDE exchange was successfully simulated by the process‐based model. The model sensitivity analysis results show that the exchange of p,p′‐DDE can be sensitive to either the sediment sorption/desorption parameters or colloidal parameters depending on the experimental conditions tested. For the experimental conditions employed here, the effect of colloids on contaminant transport is expected to be minimal, and the stream–subsurface exchange of p,p′‐DDE is dominated by the interaction of p,p′‐DDE with bed sediment. The work presented here contributes to a better mechanistic understanding of the complex transport process that hydrophobic organic contaminants undergo in natural streams and to the development of reliable, predictive models for the assessment of impacted streams. Reinforced concrete structure may exhibit significant inelastic hysteretic behavior when subject to strong earthquake excitation. To investigate such an inelastic behavior, in this study, a new system identification technique is applied by using the deteriorating distributed element (DDE) model to simulate the hysteretic behavior of a degrading structure. Through the advanced signal processing technique, the multiple singular spectrum analysis (SSA) and the nonlinear SSA, the recorded inelastic restoring force of a deteriorating structure can be decomposed into several independent additive components in its sequentially degrading order and with decreasing weight. With each decomposed hysteresis loop, the model parameters of the DDE model are identified. The evolutionary properties of the progressive stiffness degradation behavior of reinforced concrete structure can be observed from the identified model parameters. Finally, comparison on the physical properties of the identified DDE model with respect to the seismic response data of the deteriorating structure is also discussed. The result shows that the proposed identification technique can have a good estimation on the seismic behavior of the degrading structure. Copyright © 2012 John Wiley & Sons, Ltd. 相似文献

Sediments are depositories of toxic substances such as organochlorine pesticides and there is a global need for their removal in contaminated environments. Studies that combine contaminated sediments and phytoremediation are relatively recent and their number has been increasing. This work aimed to investigate whether salt marsh plants (sea club-rush Scirpus maritimus, sea rush Juncus maritimus and sea purslane Halimione portulacoides) can favor DDT and metabolites remediation in estuarine environment. For this purpose the levels of DDT, DDE and DDD were compared in vegetated and non-vegetated sediments from an estuary in the North of Portugal (in-situ study) and from another in the South of Portugal (ex-situ study). Results obtained both in the in-situ study, involving S. maritimus and J. maritimus, and in the ex-situ study, involving H. portulacoides, indicated that these plants did not have a significant role in DDTs removal and/or degradation. Therefore, it seems that the tested plants cannot influence levels and distribution of DDTs in estuarine areas. 相似文献

Study of the influence of different organic pollutants on Cu accumulation by Halimione portulacoides

The influence of each of four organic pollutants selected from among those commonly found in coastal areas, 1,1-dichloro-2,2-bis(p-chlorophenyl) ethylene (DDE), monobutyltin (MBT), Triton X-100 and polycyclic aromatic hydrocarbons (PAHs), on Cu accumulation by Halimione portulacoides was investigated. Experiments were carried out in a laboratory setting, either in hydroponics (sediment elutriate) or in a salt marsh sediment (Cávado River, NW Portugal) soaked in elutriate. Groups of H. portulacoides were exposed to media for 6 days spiked with 10 mg/L Cu(II) and with one of the selected pollutants, at an environmentally realistic concentration. DDE and MBT did not cause any major change on Cu accumulation by H. portulacoides, whereas PAHs slightly increased accumulation only in hydroponics i.e. in the absence of sediment. On the other hand, the non-ionic surfactant Triton X-100 markedly favoured Cu accumulation on plant roots both in the presence and absence of sediment. The addition of DDE, MBT and Triton X-100 also favoured Cu solubility from sediments. Therefore, the simultaneous presence of pollutants from different nature (inorganic and organic) in the estuarine environment may result in a composition of water column, pore water, sediment or biota different of that expected considering the effect of each individual pollutant. 相似文献
