大鼠口服古糖酯后尿液中糖胺聚糖的变化及其对一水草酸钙晶体抑制活性的影响

摘    要:通过对尿液总糖胺聚糖的含量测定,尿液原型古糖酯的检测以及尿液对一水草酸钙晶体生长和聚集抑制率的变化,研究大鼠一次性口服古糖酯(200m g/kg)后,三天内尿液糖胺聚糖的变化,以及尿液对一水草酸钙晶体生长和聚集抑制率的变化。结果表明,大鼠口服古糖酯后,尿总糖胺聚糖含量在第一天和第二天显著升高,第三天恢复到原有水平。与之对应,尿原型古糖酯在第一、二天可以测到,第三天消失,尿液对一水草酸钙晶体聚集抑制活性显著增强,但对晶体的生长抑制没有显著增强作用。该实验部分提供了古糖酯的代谢结果,并为古糖酯作为尿路结石抑制剂提供了有力的实验证据。

The Alteration of Urinary Glycosaminoglcyans and Urinary Inhibitory Activity to Calcium Oxalate Monohydrate after PGSS Oral Administration
Abstract:To study whether unchanged Propyleneglucol Guluronate Sodium Sulfate (PGSS) presents in urine after oral administration, together with alteration of the total urinary glycosaminoglycans (GAGs) and inhibitory activities of urine on growth and agglomeration to calcium oxalate monohydrate (COM), Sprague Dawley (SD) rats were administrated orally with 200mg/kg PGSS, 24h urines were collected before ig and three successive days thereafter with metabolic cages. The results showed that total urinary GAGs significantly increased on the first two days (p<0.01, p<0.05 respectively) after ig, with the peak value on the first day. On the third day, the GAGs secretion came back to normality (p>0.05). In consistent with total GAGs secretion, the unchanged PGSS was detectable on the first two days after ig, and absent on the third day. In terms of the inhibitory activities, the inhibitory activity of agglomeration significantly enhanced after ig of PGSS (p<0.05). But the enhance of inhibitory activity of growth was not found in this study (p>0.05). So we conclude that the current results offer strong evidence of the metabolic insight of PGSS after ig, and reveal that PGSS is a great promising compound for urolithiasis prevention and treatment.
