鸡EX-FABP基因遗传变异及与肌内脂肪含量的相关分析

摘    要:利用PCR-SSCP结合DNA测序方法,对贵妃鸡、怀乡鸡及二者的杂交F1代共计360个个体EX-FABP基因遗传特征及品种差异进行研究,发现2773T>C,2867 C>T和2913 G>A三个位点在贵妃鸡中表现为单态,在怀乡鸡和杂交F1群体中表现为多态,这3个位点有望作为贵妃鸡、怀乡鸡乃致其他品种鸡种质资源鉴定的参考依据。对比分析3个群体胸肌肌内脂肪含量(IMF)表明,怀乡鸡和杂交鸡母鸡的IMF显著高于贵妃鸡;而3个群体公鸡间的IMF差异不显著;除贵妃鸡外,同一品种内公鸡的IMF均小于母鸡(P<0.05)。多态位点与鸡胸肌肌内脂肪含量的相关性分析结果表明,在2773位点,TC基因型个体的IMF值大于TT基因型;在2913位点,AA基因型个体的IMF值大于GG基因型,但没有达到显著水平(P>0.05)。

Abstract:In the present study,Ex-FABP genetic polymorphism in a sample of 360 chicken individuals from Princess chicken,Huaixiang chicken and their F1 population were investigated.The screening of the coding regions with their intron-exon boundaries and the proximal flanking regions were performed by using a PCR–SSCP strategy.Following sequence analysis revealed 34 novel single nucleotide polymorphisms(SNPs) of chicken Ex-FABP gene,of which three SNPs including 2773T > C,2867 C > T and 2913 G > A were only found in Huaixiang chicken and the F1 chicken.IMF of breast muscle in Huaixiang hens and F1 was significantly higher than that in Princess hens.But there were no significant difference among the cocks.For every breed,intramuscular fat of hen breast muscle was significantly higher than that of cocks except for the Princess chicken(P <0.05).Then the relationship between genotypes and IMF was analyzed by the Least Squire Method.The results showed that IMF of the individuals with genotype TC was higher than those of the TT genotype at loci 2773,and individuals with genotype AA had higher values than those of the GG genotype at loci 2913.But the difference between the genotypes had not attained to significant level(P <0.05).
