壳寡糖对绝经后骨质疏松症模型大鼠骨质量作用的研究

摘    要:探讨壳寡糖(Chitooligosaccharide COS)对绝经后骨质疏松症模型大鼠骨质量的影响,研究壳寡糖对绝经后骨质疏松症模型大鼠骨组织微观结构的保护作用,为壳寡糖在预防和治疗绝经后骨质疏松症的广泛应用提供理论依据。通过切除3月龄雌性大鼠两侧卵巢建立绝经后骨质疏松症动物模型,并灌服不同剂量的壳寡糖。收集骨组织标本,分别进行骨密度、骨矿元素含量、骨组织生物力学、骨组织形态学、骨组织形态计量学的测定。结果显示:中、低剂量壳寡糖能够显著提高股骨干骺的骨密度,增加Ca,Mg元素在股骨、胫骨中的含量,保护胫骨远端的微观结构,提高骨小梁体积百分比和宽度,显著降低骨小梁的分离度以及平均矿化率;中剂量壳寡糖可显著提高股骨最大载荷和最大弯曲力。说明低分子量的壳寡糖能显著改善绝经后骨质疏松症大鼠的高转换状态。

Abstract:Purpose: The present article intents to study the effect of chitooligosaccharide on the bone quality of animal models in ovariectomized rats and the protective function on the microarchitecture of bone tissue.Methods: From the bone mineral density,the content of mineral element,biomechanics,bone morphology,histomorphometry,we fed different concentrations of chitooligosaccharide to three-month old ovariectomized rats and study its function.Results: The results demonstrate that middle and low concentrations of chitooligosaccharide can significantly increase the bone mineral density of the of femur epiphysis.Corresponding to the changes,the contents of Ca and Mg were increased in the femur and tibiae.In the study of biomechanics,we found out that middle concentrations of chitooligosaccharide can increase the breaking force and ultimate stress.The microarchitecrure of femur was well protected in the middle and low concentration chitooloigosaccharide groups.Conclusion: The low molecular weight chitooligosaccharide can significantly increase the bone mineral density of the femur and improve the biomechanical properties.So chitooligosaccharide can diverse-the main character of postmenopausal osteoporosis the high rate of bone remodelling turnover.
