低聚古罗糖醛酸数均分子量测定方法的研究

摘    要:分别采用3,5-二硝基水杨酸(DNS)比色法和高效凝胶渗透色谱法(HPGPC)测定了低聚古罗糖醛酸的数均分子量。结果表明:DNS比色法可直接测定低聚古罗糖醛酸的数均分子量,以麦芽糖和古罗糖醛酸标准三糖分别作为标准品,其测定结果相近,该方法准确度高,重复性好;HPGPC法简便易行,可通过测定低聚古罗糖醛酸的重均分子量(Mw)和分子量分布宽度D间接计算数均分子量,该法测定结果与前者结果相比有较大差别,可能与2种方法使用的标准品不同有关。

Abstract:The DNS and HPGPC methods were used to determine the number average molecular weight(M_n) of Oligoguluronic acid in this study.Both maltose and guluronate trimer were used in the DNS method as the standard samples,and their results were similar.The DNS method can determine the number average molecular weight of Oligoguluronic acid directly with high accuracy and good repeatability.Although the HPGPC method is simple and convenient,it can only determine M_n indirectly by calculating with M_w and molecular weight distribution width,and its results exhibit remarkable differences with that deter mined by the DNS method,which may be due to the difference of the standard samples.
