Copper (Cu) is an essential element for biological systems, however, when present in excess, is toxic. Metallothioneins can play an important role in Cu homeostasis and detoxification. Moreover, Cu can catalyse the production of toxic hydroxyl radicals that cause lipid peroxidation but defence systems in the cells can limit the oxidative damage. The present study was performed to investigate the effect of three Cu concentrations (0.5, 2.5 and 25 μg l−1) on the response of antioxidant enzyme activities (superoxide dismutase (SOD), catalase (CAT), selenium-dependent glutathion peroxidase and total glutathion peroxidase), total proteins, metallothioneins (MT), malondialdehyde (MDA) concentrations in the gills of the clam, Ruditapes decussatus. The activity of antioxidant enzymes and total proteins, MT and MDA concentrations were measured in the gills of the clams after 1, 3, 7, 14, 21 and 28 days of Cu exposure. Results indicate that Cu only induces an imbalance in the oxygen metabolism during the first week of Cu exposure due to a decrease in mitochondrial SOD and CAT, selenium-dependent and total glutathion peroxidase activities. Cu also causes lipid peroxidation, measured by the MDA formation, that was Cu dependent. In the gills of clams exposed to 25 μg Cu/l, the excess of Cu triggers the induction of MT synthesis after 3 days of exposure.  相似文献

本文在实验室条件下通过静态水质染毒方式在生理生化和转录水平上探究了我国重要经济鱼种鲤鱼(Cyprinus carpioio)对多氯联苯Aroclor 1242的氧化应激:鲤鱼肝脏组织抗氧化酶[超氧化物歧化酶(SuperoxideDismutase,SOD)和过氧化氢酶(Catalase,CAT)]活性、脂质过氧化产物丙二醛(Malonydialdehyde,MDA)含量、相关抗氧化酶编码基因(Cu/Zn-SOD、Mn-SOD、CAT)、应激蛋白HSP70、转录因子Nrf2和芳烃受体AhR2基因表达。结果表明:Aroclor 1242胁迫下,鲤鱼肝脏组织抗氧化酶活性表现出低浓度促进高浓度抑制的趋势;肝脏组织丙二醛含量显著升高,其中390μg/L暴露组鲤鱼肝脏组织MDA含量在第6d高于对照组92.1%(P0.01),表明肝脏组织发生脂质过氧化,膜系统受到损伤;鲤鱼肝脏组织中Cu/Zn-SOD、Mn-SOD、CAT、HSP70、Nrf2和AhR2编码基因表达出现不同程度下调。本文从酶学和基因转录水平上揭示了Aroclor1242胁迫下鲤鱼肝脏组织细胞的氧化应激。  相似文献

鲢鱼抗氧化肽对冷藏鲅鱼肉脂质氧化的影响   总被引:3,自引:0,他引:3  
研究鲢鱼抗氧化肽对冷藏鲅鱼肉脂质氧化的抑制效果和保鲜效果.将不同浓度的鲢鱼抗氧化肽(SAP)添加到鲅鱼肉中,测定了该鱼肉冷藏期间(2 ℃,12 d)的脂质水解产生的游离脂肪酸(FFA)的含量、二级氧化产物的量(TBA-i值)、初级氧化产物的量(过氧化值:POV)、产生的荧光物质的量及多烯指数(PI)等变化.结果表明,不同浓度的SAP对冷藏期间鱼肉的TBA-i、FFA、FR(荧光比值),POV的值都有较强的抑制作用,并呈现一定的量效关系.贮藏结束时,添加浓度为1.0% SAP与添加PG的鱼肉的PI值之间差异不显著.这表明SAP 主要通过抑制脂质氧化的初级氧化产物、二级氧化产物的生成及多烯不饱和脂肪酸的氧化降解而发挥作用.  相似文献

The aim of this study was to compare the effects of waterborne copper(Cu) and cadmium(Cd) on survival, anti-oxidative response, lipid peroxidation and metal accumulation in abalone Haliotis discus hannai. Experimental animals(initial weight: 7.49 g ± 0.01 g) were exposed to graded concentrations of waterborne Cu(0.02, 0.04, 0.06, 0.08 mg L-1) or Cd(0.025, 0.05, 0.25, 0.5mg L-1) for 28 days, respectively. Activities of the anti-oxidative enzymes(catalase, CAT; superoxide dismutase, SOD; glutathione peroxidases, GPx; glutathione S-transferase, GST), contents of the reduced glutathione(GSH) and malondiadehyde(MDA) in the hepatopancreas, and metal accumulation in hepatopancreas and muscles were analyzed after 0, 1, 3, 6, 10, 15, 21, 28 days of metal exposure, respectively. Results showed that 0.04 mg L-1, 0.06 mg L-1 and 0.08 mg L-1 Cu caused 100% death of abalone on the 21 st, 10 th and 6th day, respectively. However, no dead abalone was found during the 28-day waterborne Cd exposure at all experimental concentrations. Generally, activities of SOD and GST in hepatopancreas under all Cu concentrations followed a decrease trend as the exposure time prolonged. However, these activities were firstly increased and then decreased to the control level and increased again during Cd exposure. Activities of CAT in all Cu exposure treatments were higher than those in the control. These activities were firstly increased and then decreased to the control level and increased again during Cd exposure. Contents of MDA in hepatopancreas in all Cu treatments significantly increased first and then decreased to the control level. However, the MDA contents in hepatopancreas were not significantly changed during the 28-day Cd exposure. The metals accumulation in both hepatopancreas and muscles of abalone significantly increased with the increase of waterborne metals concentration and exposure time. These results indicated that H. discus hannai has a positive anti-oxidative defense against Cu or Cd. In conclusion, anti-oxidative mechanism in abalone to resist waterborne Cu did not follow the same pattern as that for waterborne Cd.  相似文献

Mussels are used as sentinel organisms and bioindicators to evaluate the toxic effects of chemical pollutants in marine organisms, especially heavy metals, representing an important tool for biomonitoring environmental pollution in coastal areas. Antioxidant defence enzymes play an important role in cellular antioxidant defence systems and protect from oxidative damage by reactive oxygen species (ROS). Indigenous mussels Mytilus galloprovincialis of the Saronikos Gulf of Greece were used for monitoring heavy metal pollution in three polluted sites in the area and in one unpolluted site. Seasonal variations of the activity of antioxidant defence enzymes, superoxide dismutase (SOD) and catalase (CAT), as well as lipid peroxidation (LP) were measured as biomarkers in a period of three years in relation to concentrations of trace metals in their gills and mantle and compared to mussels from an unpolluted sampling site. SOD activity increased at least 2 fold at the polluted sites when compared to the control site (the high activity was recorded in the spring time). CAT activity was increased 2-3 times at the polluted sites, with high activity in the winter and spring time, compared to the control site. LP concentration was twice higher at the polluted sites, following the same seasonal pattern. Trace metals contents in mussels collected at polluted sites were 3-4 fold higher compared to the control site and showed moderate variations along the months, with a winter maximum followed by a summer pre-spawning minimum matching the seasonal trends of temperature and salinity. Our results showed that metal pollution in the Elefsis Bay (the most polluted coastal area) causes relatively medium levels of oxidative stress in tissues of mussels due to cellular oxy-radical generation. This study, which is the first in the area, showed that seasonal variations of the activity of antioxidant defence enzymes and LP concentrations in mussels can be used as potential biomarkers of toxicity for long-term monitoring in marine coastal ecosystems.  相似文献

There is a lack of information on monitoring neurotoxicity in offshore commercial species. To help fill this gap, we sampled hake (Merluccius merluccius) and Norway lobster (Nephrops norvegicus) in fishing grounds off the coast of l’Ametlla de Mar (NW Mediterranean) in June 2005 at a depth of 100 m and 400 m. Additionally, at 400 m depth, two other fish species, Micromesistius poutassou and Phycis blennoides were included. Neurotoxicity markers such as Colinesterases (ChEs), namely acethyl- (AChE), butyryl- (BChE), propionyl- (PrChE) and carboxilesterase (CbE) were measured in muscle. Lipid peroxidation (LP), a marker of oxidative damage, was also included. The results are discussed in relation to the animal’s sex, size and fishing depth. A comparison of esterases and LP levels between muscle and liver of hake and between muscle and hepatopancreas of Norway Lobster was made.

AChE was dominant in muscle and CbE in hepatopancreas. No differences between fish species were seen for AChE. However, N. norvegicus, presented lower levels of ChEs and LP. A size-dependence in ChEs was seen for M. merluccius, with larger animals showing significantly lower activities (p < 0.05). Sex-dependence was seen in N. norvegicus for most esterases, except AChE, with males displaying higher activities (p < 0.05). A sampling-depth effect was also seen in the crustacea, with animals from 100 m generally presenting lower esterase activities and higher LP levels.  相似文献

用5个级别的人工复合重金属污水处理红树植物木榄Bruguiera gymnorrhiza幼苗,2个月后对其叶片和根系中的保护酶SOD、POD和CAT活性变化以及对膜脂质过氧化作用的影响进行分析。结果显示,当污染级别不大于5倍污水时,根系中SOD活性随污染程度的增加而增加,污染程度再升级其活性迅速下降但仍高于对照组水平;叶片中SOD活性在1倍污水时与对照组相比有所下降,随后的变化与在根系中的变化相似。POD活性在根系和叶片中的变化相对一致,均随污染级别的增加而升高;但在叶片中的变化幅度大于根系中的变化幅度。CAT活性在叶片中不受污染程度的影响,保持平稳,但在高程度污染下活性有下降趋势;而根系中CAT活性在10倍污水时达到最大值,污染程度再升级其活性下降。叶片中膜脂质过氧化作用随污染程度的增加而加剧,MDA的积累增加;但根系中MDA含量在5倍污水时最少,只为对照组的21.98%,其后随着污染程度的增加其MDA含量大幅度积累,膜脂质过氧化作用加剧。从整个试验结果分析认为,木榄在遭受重金属胁迫时具有优先保护根系以增强植物的抗重金属能力。  相似文献

This study is part of a project aiming to validate the use of the intertidal shanny Lipophorys pholis as a sentinel species in pollution monitoring in NW European marine ecosystems. To this end, a characterisation of acethylcholin (AChE), butyrylcholin (BChE) and propionylcholin (PrChE) esterases in L. pholis muscle was performed and the results indicated that AChE was predominant. Furthermore, the use of eserine sulphate and BW284c51 (0.64–800 μM), and iso-OMPA (0.08–16 mM), confirmed the measurement of true cholinesterases (ChEs) as well as the presence of pseudocholinesterases. The field application of these markers to L. pholis, sampled in seven locations along the Portuguese coast, revealed that fish were likely to be affected by neurotoxic compounds. This was indicated by the significant depletion of AChE (p < 0.05) in animals collected at urban and industrialised sites, compared with those from reference locations. The inclusion of a marker of effect, measured as lipid peroxidation levels in muscle tissue, also revealed the existence of site differences. Overall, the study further validates the utility of L. pholis in pollution monitoring studies.  相似文献

Juvenile grey mullet (Chelon labrosus) were fed either a basal (4.4 mg Cu/kg dry wt) or high-Cu diet (2400 mg Cu metal/kg dry wt) for 10 weeks to assess the relationship between growth and Cu-induced oxidative damage. No mortalities were observed, but growth rate and food intake were reduced by 43 and 29%, respectively, in the high-Cu group. This was not only attributed to peroxidative damage of the food which reduced food intake (palatability), but also to direct effects of hepatic lipid peroxidation. Hepatic α-tocopherol concentrations were 63% lower, while malondialdehyde (MDA) increased by 304% in mullet fed a high-Cu diet for 67d. Cu-exposure reduced apparent α-tocopherol retention by 420%. Hepatic Fe and Zn levels fell, while Cu increased (p < 0.05). The results show that oral Cu exposure induces oxidative stress in grey mullet. Quantification of lipid peroxidation (rancidity) in the food, reflecting palatability, is essential to differentiate the direct toxic effects of Cu on growth from the secondary influences of reduced food intake.  相似文献
