将嗜热菌Geobacillus sp. ZH1的超氧化物歧化酶(supseroxide dismutase,SOD)基因插入表达载体pET-32α(+),在Escherichia coli BL21(DE3)中进行表达,并利用亲和层析纯化重组超氧化物歧化酶.将制备的脱辅SOD进行Mn2+和Fe2+金属重构后,得到的Mn2+重构SOD的比活力达668U/mg,Fe2+重构Fe-SOD没有活性,说明ZH1菌株的超氧化物歧化酶为Mn-SOD.凝胶过滤及SDS-PAGE分析显示,Mn2+重构SOD为同聚二聚体,亚基分子量为71.7 kDa.这些研究结果为进一步研究该酶的生化特性及酶的定向进化研究奠定了良好的基础.  相似文献

The Indian crust, generally regarded as a stable continental lithosphere, experienced significant tectono-thermal reconstitution during the Phanerozoic. The earliest Phanerozoic tectonic process, which grossly changed the geological and geophysical character of the Precambrian crust, was during the Jurassic when this crustal block broke up from the Gondwana Supercontinent. There were two earlier abortive attempts to fragment the supercontinent in the Palaeozoic. Different types of geological processes were associated with these aborted events. The first was the intrusion of anorogenic alkali granites during the Early Palaeozoic (at 500±50 Ma), while the second was linked with formation of the Gondwana rift basins during Late Palaeozoic. The tectonic history of the Indian Shield subsequent to its separation from the Gondwanaland at around 165 Ma is a complex account of its northward journey, which was culminated with its collision with the northern continental blocks producing the mighty Himalayas in the process. Considerable reconstitution of the Indian Shield took place due to magma underplating when this lithospheric block passed over the four mantle plumes. While the underplating events grossly changed the geophysical character of the Indian Shield in isolated patches, the propagation of the underplated materials was assisted by the deep crustal fractures (geomorphologically expressed as lineaments), which formed during the break-up of the Gondwanaland. Several of these deep fractures evolved through the reactivation of the pre-existing (Precambrian) tectonic grains, while some others developed as new fractures in response to either the extensional stresses generated during the supercontinental break-up or the plume-lithosphere reactions. Significant geomorphological changes occurred in peninsular India subsequent to the continental collision. Most of these changes were brought about by the movements along the lineaments, which fragmented the Indian Shield into a number of rigid crustal blocks. The present day seismic behaviour of the Indian Shield is a reflection of movements of the rigid crustal blocks relative to each other. An interesting feature of the Phanerozoic geological history of the Indian Shield is the evolution of a number of sedimentary basins under different tectono-thermal regimes.  相似文献

全球化是推动乡村地方重组的重要因素,但国家政策深刻影响乡村地方参与全球化的过程与景观。立足已有研究,围绕“全球化-国家乡村政策-乡村地方重组”之间的关联,通过梳理全球化与中国乡村政策演化的互动关系,分析乡村政策的地方实施过程,总结乡村地方工业化、休闲旅游化和农业现代化等典型重组类型及其面临的不确定性。认为早期农村改革耦合全球化引致的城镇化进程,解决了农村温饱和人口“过密化”问题,推动部分乡村工业化,参与国际分工;持续的深化改革与地方动员,加快了先进技术和生产要素的应用,促进了农业规模化与专业化,并嵌入国际市场;乡村旅游的兴起,促进部分乡村复兴和国际元素的进入。农业收益的增长与稳定、老年农民有序退出以及新业态的就业容量将是乡村振兴的潜在制约因素。  相似文献

A thermostable superoxide dismutase (SOD) from the inshore thermophile Thermus sp. JM1 was purified to homogeneity by steps of fractional ammonium sulfate precipitation, DEAE-Sepharose chromatography and Phenyl-Sepharose chromatography. The specific activity of the purified native enzyme was 1 656 U/mg. A sod gene from this strain was cloned and overexpressed in Escherichia coli (E. coli). The prepared apo-enzyme of the purified recombinant SOD (rSOD) was reconstituted with either Fe or Mn by means of incubation with appropriate metal salts. As a result, only Mn 2+ - reconstituted rSOD (Mn-rSOD) exhibited the specific activity of 1 598 U/mg. SOD from Thermus sp. JM1 was Mn-SOD, judging by the specific activities analysis of Fe or Mn reconstituted rSODs and the insensitivity of the native SOD to both cyanide and H 2 O 2 . Both the native SOD and Mn- rSOD were determined to be homotetramers with monomeric molecular mass of 26 kDa and 27.5 kDa, respectively. They had high thermostability at 50 ° C and 60 ° C, and showed striking stability across a wide pH span from 4.0 to 11.0.  相似文献

着重论述了蛇绿岩地幔橄榄岩中豆荚状铬铁矿的成因,并对现今盛行的岩石/熔体反应成矿说提出了质疑。世界含铬铁矿的地幔橄榄岩均显示上部偏基性、下部偏酸性的垂直熔融分带,与蛇绿岩堆晶岩中上部偏酸性、下部偏基性的岩浆分异垂直层序恰恰相反。豆荚状铬铁矿与熔融剖面上部的纯橄岩或纯橄岩-方辉辉橄岩杂岩带紧密伴生。豆荚状铬铁矿是原始地幔岩高度熔融再造的产物,高铬型铬铁矿与PPG型蛇绿岩伴生,形成于岛弧或弧前盆地环境;高铝型铬铁矿与PTG型蛇绿岩伴生,形成于扩张脊(MOR)或弧后盆地环境。玻安岩(boninite)与高铬型豆荚状铬铁矿无成因关系,铬铁矿(或富铬矿浆)的形成反而为boninite提供了其形成所需的残余地幔;高铝型铬铁矿不是地幔橄榄岩/拉斑玄武质熔体反应形成的,而是富铬矿浆与基性熔体发生再平衡的产物。豆荚状铬铁矿中超高压矿物包体的出现为其地幔深部成因提供了佐证,而boninite形成于浅部较低压的条件;豆荚状铬铁矿中富集强相容元素IPGE(Os、Ir、Ru)合金,boninite富集不相容元素PPGE (Pt、Pd)硫(砷)化物, 而亏损IPGE,显示其形成较晚。因此,boninite与铬铁矿无生因关系,两者均受岛弧(或弧前盆地)环境的制约而在空间上相伴产出。  相似文献

A sequence of at least three Al2SiO5-bearing mineral assemblages are preserved in successively overprinted ductile shear zones in the Willimantic window, Connecticut. The ductile deformation, localized at and near the boundary between the Putnam-Nashoba terrane and underlying Avalon terrane is characterized by a network of anastomozing shear zones that outline metre-scale tectonic blocks of migmatitic Kfs + Sil + Gt + Bi + Pg + Qtz + Ilm + Ru gneiss. These assemblages record Acadian or older metamorphic conditions of 6 kbar, 700d? C. Mylonitic gneisses in shear zones that define block margins were formed by reconstitution and recrystallization of the migmatitic gneiss. The reconstituted rocks exhibit relict Ky + St + Grt (+Pl + Bt + Qtz + Rt + Ilm) assemblages and require a minimum pressure for the Ky-Str grade metamorphism of 8.5 kbar. Kyanite in block margins is widely replaced by sillimanite, and locally by andalusite, during a period of post-Alleghanian ductile deformation. The interiors of blocks do not record this sequence of polymorphs. The pattern of reconstitution is accounted for by localization of strain along block margins within a regionally extensive terrane-bounding fault zone. Strain provided the activation energy for recrystallization and retrograde mineral reactions. The P-T conditions of post-Alleghanian ductile deformation evolved from 600d? C and 6 kbar to 550d? C and 3 kbar. The occurrence of Ky + Str-bearing assemblages, overprinting Acadian Kfs + Sil-bearing assemblages and subsequently overprinted by Alleghanian sillimanite- and andalusite-bearing assemblages, along with reset hornblende 40Ar/39 Ar mineral ages from Mississippian to Permian, requires a prograde Alleghanian metamorphism of rocks previously metamorphosed during the Acadian. Thus, mineral assemblages from gneisses in the Willimantic fault zone retain evidence of a protracted tectonothermal evolution that included high-grade Acadian orogenesis, tectonic loading resulting from Alleghanian collision of Avalon with North America, and tectonic exhumation in Permo-Triassic time. The c.3-kbar pressure decrease between prograde and retrograde Alleghanian metamorphic conditions corresponds to 10 km of crust that must have been tectonically excised from the base of the Putnam-Nashoba terrane cover sequence following Alleghanian orogenesis in south-eastern New England.  相似文献

简要叙述了现行航空摄影测量的作业模式和关键技术要求,分析了直接利用航摄像片外方位元素恢复立体模型所得到的目标点物空间位置精度以及模型点的上下视差。经对带有双频动态GPS数据的1∶2 500~1∶60 000各种摄影比例尺的覆盖多种地形航摄像片的试验表明,由摄影测量加密所获取的影像外方位元素可直接用于4D产品生产中的影像定向,而由POS系统提供的影像外方位元素还难以直接用于摄影测量测图。   相似文献
