Diagnosis of iridovirus in large yellow croaker by PCR   总被引:1,自引:0,他引:1  
A rapid and sensitive PCR-based method for the detection of the large yellow croaker iridovirus(LYCIV) is described,which involves the amplification of a 295 bp fragment of the LYCIV ATPase gene from DNA isolated from naturally infected fish spleen.Sequencing of LYCIV ATPase gene fragment showed it shared 100% nucleotide sequence homology with the corresponding region of the ATPase gene of red sea bream iridovirus(RSIV) and sea bass iridovirus(SBIV),suggesting that LYCIV was homologous with RSIV and SBIV at least in part of the gemone.The specificity and sensitivity of the PCR procedure were tested on the iridovirus-infected fishes,the expected fragment was detected from spleen DNA samples of infected fishes,whereas no fragments were amplified from healthy fish spleen DNA,white spot syndrome baculoviruses(WSBV) DNA and pseudorabies virus(PRV) DNA.Detection limit of this method was 10-7 ng positive plasmid DNA containing target sequence,equal to about 100 virions.In the infected experiment,first positive detection(1/4) appeared at Day 3 post-infection,all fish(4/4) tested positive at Day 7,however obvious symptoms were observed at Day 8,so LYCIV infection could be detected prior to the appearance of obvious symptoms.These results indicate that this PCR method could be used for early,rapid and specific detection of LYCIV infection.  相似文献

本文对黄海测区空气微生物作了一次测定 ,结果表明空气中海洋性细菌、真菌的检出率分别为 52 .4 %和 4 7.6 %。陆源性细菌、真菌的检出率分别为80 .0 %和 6 0 .0 %。陆源性微生物出现机率大于海洋性的。空气微生物以细菌为主。测区平均的海洋性空气细菌、真菌、总菌量及真菌 /总菌 %分别为 592 .6 ,32 9.1 ,92 1 .7CFU·m-3 及 35.4。平均的陆源性空气细菌、真菌、总菌量及真菌 /总菌 %分别为 6 89.1、377.9、1 0 6 7.0CFU·m-3 及 35.7。文章分析了空气微生物的时空分布状态。指出测区空气微生物状态反映了海 -气、海 -陆、人和自然的相互作用。  相似文献

High concentrations of arsenic and humic substances in groundwater from the southwestern coastal plain of Taiwan were well known for their probable relationships with black-foot disease. In order to realize the relationships between the concentrations of humic substances and arsenic in this area, 24 well water samples were analyzed. After filtered through 0.45 μm glass fiber membrane in the field, samples were kept in the dark at 4℃ and then separated into six fractions with varying range of molecular weight (〈0.5, 0.5-1, 1-5, 5-10, 10-50 and 〉50 k Da) by ultrafiltration apparatus (Molecular/Por Stirred Cell system) in the lab. Concentrations of humic substances were measured by fluorescence spectrometer (HITACHI F-2000, ex=370, em=445) and arsenic by FIAS-AA (Perkin Elmer AAnalyst 100, FIAS-400). On average, only 6.2% of the total arsenic in water existed in the fraction of 〉0.5 k Da. and the others were complexed with humic substances (including humic acid and fulvic acid). The results demonstrated a distinct positive correlation between the concentrations of humic substances and arsenic.  相似文献

Low molecular weight organic acids (LMWOAs) are important components of root exudates. They play an important role in immobilizing and remobilizing contaminants in rhizospheric soil. Effects of four LMWOAs on the sorption and desorption behavior of p-chlorophenol by yellow earth was studied in batch mode. The results showed that the previous application of LMWOAs to enhanced adsorption capacity of p-chlorophenol of the soil in the order of maleic acid〉malic acid〉oxalic acid〉citric acid. However, when LMWOAs were applied to soil where p-chlorophenol had been previously adsorbed, substantial p-chlorophenol was desorbed from soil by oxalic acid, whereas citric acid, malic acid and maleic acid didn't desorb as much p-chlorophenol from soil as deionized water.  相似文献

Photodegradation of chrysene, benzo (a) pyrene and benzo (g, h, i) perylene in natural water of the Yellow River was studied using simulation sunlight. The effects of particulates on the photodegradation were explored. Several results arose from this study. (1) The photodegradation of PAHs can be fitted with first-order kinetics when no particulate exists in water system, and the first-order constant increases with decreasing of initial concentration of PAHs. The photodegradation rates of the three PAHs are related to their molecule absorption spectrum. (2) The existence of loess exerts two kinds of effects on the photodegradation of PAHs, including the inner filter effects and the photosensitizing effects of humic substance in loess. These two contrary effects lead to the difference of net effects among different contents of loess. When the loess contents are 0.1 and 5.0 g/L, the existence of loess stimulates the photodegradation of chrysene, benzo (a) pyrene. When the loess content is 5.0 g/L, the existence of loess stimulates the photodegradation of benzo (g, h, i) perylene. In addition, the photodegradation of PAHs can be fitted with the second-order kinetics when there is loess in the water system. (3) The soluble humic substances in loess can accelerate the photodegradation of PAHs while the in soluble part cannot. 对于一般树状分布河网,上一级河道总是汇入至下一级河道,位于河道下游断面的流量总是由其上游汇集而至。但对于平原河网地区,特别是人工控制建筑物众多,又受潮汐影响的地区(如太湖流域),河网错综复杂,水流方向不定,要跟踪某个断面的水流去向,或某个河段的水体、断面流量是从哪里汇集而来的,非常困难。但这样的问题在生产实践中往往对其很感兴趣。例如从常熟枢纽引长江水流进入望虞河后,流向何处?河网各断面流量中或各河段水体中常熟枢纽引江水量占多少比重?对这些问题的研究可以估计常熟枢纽引江的效果和影响范围。因此,平原河网区域来水组成方法在生产实践中具有重要意义。重点介绍了平原河网区域来水组成原理及其在太湖流域的应用。  相似文献
