Pure fraction (92%–95%) of phagocytes (FP) and a mixture of amoebocytes (62%) and morula cells (38%)-FPMC- of the holothurianEupentacta fraudatrix' (Holothuroidea, Dendrochirota) were obtained by using ficoll-verographine step gradient. Basal production of reactive oxygen species (ROS) in FP quantified
by using reduction of nitroblue tetrazolium (NBT) was more than twice that in FPMC. Thermostable toxin ofYersinia pseudotuberculosis (TST) at different concentrations (0.2; 0.5; 2.5 μg/ml, but not 0.1 μg/ml) stimulated NBT reduction in FPMC after 24 h incubation.
In FP, TST at concentrations of 0.1 and 0.2 μg/ml inhibited and at concentrations of 0.5 and 2.5 μg/ml stimulated NBT reduction
after 24 h incubation. Maximal effect was observed in FP and FPMC at TST concentrations of 0.5 and 0.2 μg/ml, respectively.
Addition of catalase (0.7 μg/ml) to the cells treated with TST (2.5 μg/ml) was followed by a decrease in NBT reduction compared
to that under toxin treatment alone. TST stimulated superoxide dismutase activity in concentration-dependent manner (maximum
at 0.5 μg/ml concentration in FP) after 24 h treatment, and this stimulation was prevented by a commercial catalase. Plant
lectin concanavalin A stimulated NBT reduction more than 5-fold in FPMC compared to the control. With addition of TST, lectin
stimulated ROS to lesser extent than that with lectin alone. When catalase, TST, and lectin were added into the FPMC simultaneously,
ROS increase was similar to that under lectin treatment alone. On the whole, data obtained indicated that ROS generation in
holothurian coelomocytes especially occurs in both stimulated and not stimulated phagocytes, and that changes in ROS production
by these cells may be one of the mechanisms of antibacterial protection of holothurians.

A model based on that of Kishi et al. (2001) has been extended to 15 compartments including silicon and carbon cycles. This model was applied to Station A7 off
Hokkaido, Japan, in the Northwestern Pacific. The model successfully simulated the observations of: 1. a spring bloom of diatoms;
2. large seasonal variations of nitrate and silicate concentrations in the surface water; and 3. large inter-annual variations
in chlorophyll-a. It also reproduced the observed features of the seasonal variations of carbon dioxide partial pressure (pCO2)—a peak in pCO2 in winter resulting from deep winter convection, a rapid decrease in pCO2 as a result of the spring bloom, and an almost constant pCO2 from summer through fall (when the effect of increasing temperature cancels the effect of biological production). A comparison
of cases with and without silicate limitation shows that including silicate limitation in the model results in: 1. decreased
production by diatoms during summer; and 2. a transition in the dominant phytoplankton species, from diatoms to other species
that do not take up silicate. Both of these phenomena are observed at Station A7, and our results support the hypothesis that
they are caused by silicate limitation of diatom growth.

Permeable Reactive Barrier (PRB) is an emplacement of inert material (s) in the subsurface, designed to intercept a contaminated plume, provides a preferential flow path through the reactive media, and transforms the contaminant into environmentally acceptable forms to attain concentration remediation goals at the discharge of the barrier. The phenomena, which help in remediation within PRB, are adsorption/sorption, precipitation, oxidation/reduction and biodegradation. Various materials like zero-valent iron, zero-valent bi-metals, natural zeolites, organic carbon, fly ash, zeolites, limestone, activated alumina, apatites, etc. have been tried by many researchers to remove organic and inorganic contaminants. In USA, Canada, and many European countries commercial full-scale and pilot scale PRBs are successfully working. The design and installation of full scale PRBs needs laboratory treatability and dynamic flow column experiments? The concept of PRB is being applied to treat contaminated surface water in the Katedan industrial area, Hyderabad, India. National Geophysical Research Institute (NGRI), Hyderabad, India, conducted systematic studies in collaboration with Norwegian Geotechnical Institute (NGI), Norway, to develop PRB technique to decontaminate the surface water pollution due to industrial effluent. A site assessment study in the Katedan Industrial Area, were carried out and water, soil and sediment from the lakes of the area were found to be polluted with high concentrations of heavy metals like As, Pb, Cr, Cd, Ni, etc. Adsorption studies at NGRI with synthetic samples and in-situ industrial effluent using different reactive media for removing contaminants like arsenic, chromium, cadmium, copper, nickel, lead and zinc have been carried out and yielded satisfactory results. This contribution aims to report the reflections we had with the scientific community during two international workshops on reference materials for stable isotopes in Davos (2002) and Nice (2003). After evaluating the isotopic homogeneity of some existing reference materials, based on either certificates, literature data or specific inter-laboratory rounds, we confirm these as primary reference materials or propose new ones relative to which stable isotope compositions should be reported. We propose DSM-3 for Mg, NIST SRM 915a for Ca, L-SVEC for Li and NBS28 for Si. Cadmium does not yet have a well identified delta zero material, although three commercial mono-elemental Cd solutions have yielded the same isotopic composition relative to one another. In order to scale the linearity of any mass spectrometer, some secondary reference materials are also proposed: Cambridge-1 solution for Mg, the "Münster-Cd" and JEPPIM Cd solutions for Cd and the "Big Batch" silicate for Si. The team from Nancy propose to prepare a mixed spike solution for Li isotopes. Well-characterised natural samples such as ocean or continental waters, diatoms, sponges, rocks and minerals are needed to validate the entire analytical procedure, particularly to take into account the effect of sample mineralisation and of chemical manipulations for elemental separation prior to analysis. 相似文献

We give some results obtained for the Couplex test cases proposed by the ANDRA. In this paper our aim is twofold. Firstly, to compute the release of nuclides out of the repository by concentrating on the 3D near field (Couplex 2). The simulation of the transport phenomena takes into account the dissolution of the glass containers and congruent emissions of the radio-nuclides including filiation chains and some simplified chemistry. Secondly, it is to use the near field computations in order to simulate the nuclide migrations in a 2D far field (Couplex 3). Coupling in between the two simulations takes into consideration the periodicity of the disposal modules and the geometry of the repository described in Couplex 1. The mixed finite element and discontinuous Galerkin methods are used to solve the convection–diffusion equations. In order to handle the nonlinear precipitation/dissolution term, we developed a new iterative technique that combines Picard and Newton–Raphson methods. 相似文献

Modelling contaminant transfer with biological/chemical/radioactive processes needs appropriate numerical methods able to reproduce sharp concentration fronts. In this work, we develop a new Eulerian–Lagrangian Localized Adjoint Method (ELLAM) for solving the reactive transport equation with non-constant coefficients. To avoid interpolation (leading to errors), we use a moving grid to define the solution and test functions. The method is used to simulate first the infiltration of solute into a column of unsaturated porous medium and second the multispecies transport. The developed ELLAM gives accurate results without non-physical oscillations or numerical diffusion, even when using large time steps. To cite this article: A. Younes, C. R. 探讨活性氧(reactive oxygen species,ROS)在温石棉激活线粒体通路诱导A549凋亡中的作用。采用不同浓度温石棉粉尘染毒人肺上皮细胞(A549)后,MTT法检测细胞存活率,流式细胞术检测ROS抑制剂N-乙酰半胱氨酸(N-acetyl-L-cysteine,NAC)预处理前后的细胞凋亡率、胞内ROS水平及线粒体膜电位,Western Blot法检测各组细胞半胱天冬氨酸蛋白酶3(caspase-3)、p53蛋白表达水平。实验结果显示,温石棉可降低A549细胞存活率,呈现时间-剂量效应关系;NAC可显著减少胞内ROS水平、阻止线粒体膜电位下降,并下调caspase-3、p53蛋白的表达水平,抑制部分细胞凋亡的发生。表明ROS可通过诱导p53蛋白过表达,激活线粒体凋亡通路,诱导A549细胞发生凋亡。 相似文献
