Most anti-tumour agents are known to be carcinogenic, mutagenic, teratogenic, embryotoxic or fetotoxic. Only little is known about the environmental impact of pharmaceuticals. Unmetabolized active substances are excreted and will show up in municipal wastewater. Therefore, we examined the biodegradability of the widely used anti-tumour agent 5-fluorouracil (5-FU), also as an example for assessing the impact of hospital effluent on the biodegradation of a test compound. The biodegradability of the structurally similar anti-tumour agents cytarabine and gemcitabine was also examined. Test systems used were the closed bottle test (OECD 301 D) and the modified Zahn-Wellens test (OECD 302 B). 5-FU was not biodegradable in the closed bottle test (CBT) nor in the Zahn-Wellens test (ZWT). At the highest concentration of 5-FU, with hospital effluent an inhibition of the degradation of substances present in hospital effluent was observed, probably as a result of a synergistic effect by 5-FU with antibiotics present in hospital sewage. Gemcitabine was biodegraded 42% in the CBT. The prolongation of the test period to 40 days only improved the result to 45%. In the ZWT, the biodegradation of gemcitabine was 50%. Cytarabine was partially biodegraded in the CBT (50%), but only after an adaptation period of 20 days. After a test prolongation to 40 days, the degree of biodegradation was 80%. This contribution attempts to determine whether, and to what degree, environmental samples of resuspended particulate matter and sediments exert a toxicological impact. Further, an attempt is made also to screen the toxic level of potentially hazardous sites, based upon established sediment toxicity criteria. Therefore, a rapid, cost-effective and highly sensitive biotest (bioluminescence assay, based upon marine bacteria) has been applied on: biological fluid extracts; bottom sediments; and sediment trap samples. Samples were taken either from the Bay of Banyuls-sur-Mer (northwestern Mediterranean, France) or Thermaikos Gulf (northeastern Mediterranean, Greece). Biological fluid extracts and sediment trap samples corresponded to periods of resuspension events, or preceding and following such events.The results have revealed that the sampling strategy and biotest implemented in this study might be a useful tool for screening the toxicity of resuspended matter and sediments. Resuspension events appear to be able to exert an influence on the chemical forms of the micro-pollutants; thus on their bioavailability and toxicity. Nevertheless, based upon chemical analysis combined with the bioassay, the toxic level of the sediment samples could be determined, with the level at potentially hazardous sites being characterised. 相似文献

4 种重金属离子对中华鳖(Trionyx sinensis)稚鳖的急性致毒效应 总被引:1,自引:0,他引:1
以5日龄中华鳖稚鳖为实验动物,采用静水停食实验法,在水温(27.4±1.3)℃条件下,开展了Hg2+、Cr6+、Cu2+、Zn2+对中华鳖稚鳖的急性毒性和加和等毒性强度联合毒性试验。结果表明,4种重金属离子对中华鳖稚鳖均呈现以蓄积为主导的急性毒发效应,Cr6+、Zn2+对中华鳖稚鳖的急性致毒高峰期明显滞后于Hg2+、Cu2+;4种重金属离子毒性大小依次为Hg2+、Cu2+、Cr6+、Zn2+,其对中华鳖稚鳖96h的半致死质量浓度分别为5.98、16.42、28.90和91.88mg/L;所构建的中华鳖稚鳖累积死亡概率与重金属质量浓度和实验时间之间的数学模型,以及半致死时间-质量浓度回归方程,可作为侦查和分析重金属离子排放时间和致中华鳖稚鳖大量死亡时间的重要计算工具;Hg2+、Cr6+、Cu2+、Zn2+离子两两组合在加和等毒性强度下对中华鳖稚鳖96h联合急性毒性所呈现的致毒特征与离子种类及其毒性强度匹配情形密切相关。 相似文献

Fate and effect of copper and cadmium on the degradation of chitin were studied in sediment microcosms. Artificial sediment was spiked with metals, and metal partitioning between sediment components, overlying water and bacteria was described. Most of the added metals were bound to sediment particles. Remarkable metal enrichments were found on Fe-oxide, Mn-oxide, and chitin. In sediments containing the nominal concentration of 100 mg/kg Cu, the chitin-degrading bacteria Cytophaga johnsonae showed a significant decrease in N-acetylglucoseaminidase activity. Toxicity of copper was attributed to free ions in the overlying water. The bacteria were more sensitive to copper than to cadmium. Inhibitory effects of cadmium were primarily caused by sediment-associated cadmium. 相似文献

The Conductivity Test — Determining the Membrane-toxic Properties of Substances A new biotest system is introduced which is specific in terms of the measured effect but not in terms of the applied organisms. The system is able to detect the effect of membrane-toxic compounds e.g. detergents, organic solvents, and radical formers. Released into the environment, these substances damage biological membranes and therefore cause ion leakage into a surrounding medium. Ion leakage is taken as a measure for membrane damage and is determined as an increase of the electrical conductivity in the medium (conductivity test). The results presented for the tensides benzalkonium chloride, LAS (linear alkylbenzenesulfonate), and APG (alkylpolyglycoside) demonstrate the capability of the conductivity test to distinguish between the differing toxicological potential of these membrane-toxic substances. In contrast to most of the conventional biotests, the endpoint of the conductivity test is independent of a specific organism. This independence is demonstrated by the results obtained with benzalkonium chloride treatment of Elodea canadensis, Lemna minor, and Salvinia natans. The conductivity test represents a versatile, easy to handle biotest system for the assessment of ecotoxicological effects caused by membrane-toxic substances. 相似文献

The OECD soil, which is standardised for use in the sediment toxicity with the midge Chironomus riparius revealed inhibition of the nematode Caenorhabditis elegans in body growth length and egg production of 41.0% and 84%, respectively. 18 formulated sediments were tested to optimise the sediment composition for growth and reproduction of C. elegans. Their components were chosen to simulate native sediments, whereby different quantities and different clay minerals were tested. The mixture that we found to be optimal in our experiments consisted of 5% sphagnum peat, 70% calcitic sand, 0.5% dolomite limestone, 4.5% iron(III) oxide, and a clay combination of 1.5% chlorite and 18.5% aluminium(III) oxide. When applying this mixture to C. riparius, the new formulated sediment improved also growth in comparison to the OECD sediment. 相似文献

Hydrochemical data are presented for arsenic (As) in the mine waters of 34 gold and base-metal mining localities in seven
countries of south-east Asia, Africa and Latin America, encompassing contrasting climatic settings and at least eight discrete
styles of primary mineralization. Peak dissolved As concentrations at these sites range from 0.005–72 mg/l, with the United
States Environmental Protection Agency (US-EPA) potable water threshold of 50 μg/l exceeded in 25 cases. Arsenate (As5+) constitutes the dominant species at over 80% of sites. Very high dissolved As concentrations (>1 mg/l) show no systematic
between-site relationship with mine water pH/Eh regime. Important determinants of mine water As fluxes include iron hydrochemistry,
the presence of cyanic ore processing effluents, site geology, the paragenetic sequence that follows sulphide oxidation, climate
and mine management. Human toxicological impacts of As contamination have been recognized at only one case-study site, with
a further four considered to warrant more detailed risk assessment.

Crude oils produced in the North West shelf of Western Australia are highly volatile, a characteristic not shared by most of the Northern Hemisphere crude oils on which internationally accepted toxicity test protocols were developed. Because of this volatility and some other factors, the LC50 and EC50 values obtained from acute toxicity tests will be significantly affected by the changes of toxicant concentration in test solutions during the period of exposure. To address these issues all steps of a standard protocol for crude oil toxicity testing have been revised. A systematic study has been performed on factors which affect petroleum hydrocarbon solubilisation in aqueous systems during test solution preparations. The influence of mixing time, agitation energy and volume/interface ratio on a hydrocarbon concentration in a water-soluble fraction (WSF) was studied for heavy, medium and light crude oils. A study of the sensitivity of marine unicellular algae to WSF of crude oils was conducted with Isochrysis sp., Nannochloropsis-like sp. and Nitzchia closterium. Total concentrations of hydrocarbons dissolved in test solutions were estimated by UV-spectrometry and GC/FID chemical analyses. When the toxicant concentration decreased during the exposure period, the EC50 values derived from initial or final concentrations either underestimate or overestimate toxicity, respectively. Therefore, weighted average concentrations (WAC) calculated for the whole test period were recommended for expressing hydrocarbon concentrations in test solutions of crude oils. Toxicity indices calculated from WAC of total hydrocarbons for different crude oils can be compared regardless of the rates of hydrocarbon loss. 相似文献
