A recent development in algal pigment analysis by high-performance liquid chromatography (HPLC) is the application of automation. An optimization of a complete sampling and analysis protocol applied specifically in automation has not yet been performed. In this paper we show that automation can only be successful if the various methodological aspects of the sampling and analysis protocol are considered in coherence. We introduce an optimized protocol that involves freeze-drying of the sample, subsequent extraction in 90% acetone and the application of water-packing during analysis. The method was evaluated on both natural plankton populations and a broad spectrum of microalgal cultures: Thalassiosira weisflogii (Bacillariophyceae), Emiliania huxleyi (Prymnesiophyceae), Phaeocystis globosa and Phaeocystis antarctica (Prymnesiophyceae) and Pyramimonas sp. (Prasinophyceae). Whereas pigment extracts were unstable in methanol, with recorded chlorophyll a losses from 10% to 60% per day, pigment degradation rates in acetone were generally less than 1% over 18 h storage in the autosampler (4 °C). In addition, it was found that the extraction efficiency of acetone significantly increased upon freeze-drying prior to extraction. Increases as high as 50–60% were measured in P. antarctica. The application of water-packing of the sample during injection resulted in improved peak shape and peak separation, without diluting the pigment concentrations. Automation is especially beneficial for application in the field, when mixed algal assemblages and low biomass put a high demand on the sensitivity as well as reproducibility of the method. 相似文献

The extensive intertidal flats along Eighty-mile Beach in North-western Australia appear to be monotonous and homogeneous and seem ideally suited to study tidal zonation in macrozoo-benthic communities and their possible correlates with characteristics of the sediment. In October 1999, we sampled benthic invertebrates and sediments at a total of 895 sampling stations distributed over six different locations, each location separated by 15 km of unsampled foreshore along Eighty-mile Beach. To test for the presence or absence of patterns of tidal zonation (distinct height-related zones of specific sediment grain sizes or zoobenthic taxonomic groups) or patchiness (distinct patches of specific sediment grain sizes or zoobenthic taxonomic groups not related to tidal height) each location was divided into three along-shore sections and each section (transect) was examined at two or three tidal heights. Zonation was observed for sediment grain sizes. Sediments were coarser at the highest intertidal level and finer towards the low water line. Benthic assemblages also differed among tidal heights, but in terms of species-composition the differences were not consistent among the locations. Each location supported a unique collection of benthic invertebrates. Therefore the hypothesis of the presence of distinct zones of specific species or zoobenthic taxonomic groups was rejected; the presence of benthic patches was confirmed. The distribution of sediments and the composition of benthic assemblages were surprisingly poorly correlated compared to those reported in 12 previous quantitative studies around the world. One possible explanation might be that super-cyclone Vance, which hit the study-area only six months before this study, contributed to this poor correlation. It is becoming more apparent that commonly used statistical methods (e.g. analysis of variance and regression) are not the
best methods for estimating limiting relationships or stressor effects. A major challenge of estimating the effects associated
with a measured subset of limiting factors is to account for the effects of unmeasured factors in an ecologically realistic
matter. We used quantile regression to elucidate multiple stressor effects on end-of-season biomass data from two salt marsh
sites in coastal Louisiana collected for 18 yr. Stressor effects evaluated based on available data were flooding, salinity
air temperature, cloud cover, precipitation deficit, grazing by muskrat, and surface water nitrogen and phosphorus. Precipitation
deficit combined with surface water nitrogen provided the best two-parameter model to explain variation in the peak biomass
with different slopes and intercepts for the two study sites. Precipitation deficit, cloud cover, and temperature were significantly
correlated with each other. Surface water nitrogen was significantly correlated with surface water phosphorus and muskrat
density. The site with the larger duration of flooding showed reduced peak biomass, when cloud cover and surface water nitrogen
were optimal. Variation in the relatively low salinity occurring in our study area did not explain any of the variation inSpartina alterniflora biomass. 相似文献

Low-low satellite-to-satellite tracking: a comparison between analytical linear orbit perturbation theory and numerical integration 总被引:1,自引:0,他引:1

Low-low satellite-to-satellite tracking (ll-SST) range-rate observations have been predicted by two methods: one based on a linear perturbation theory in combination with the Hill equations, and one based on solving the equations of motion of two low-flying satellites by numerical integration. The two methods produce almost equivalent Fourier spectra of the range-rate observations after properly taking into account a few resonant terms. For a typical GRACE-type configuration, where the two satellites trail each other at a distance of 300 km at an altitude of 460 km, and in the presence of the EGM96 gravity field model, complete to spherical harmonic degree and order 70, the agreement between the Fourier spectra is about 1 mm/s compared to a root-mean-square (RMS) value of more than 220 mm/s for the range-rate signal. The discrepancy of 1 mm/s can be reduced significantly when not taking into account perturbations caused by the J2 term. Excluding the J2 term, the agreement between the two methods improves to 0.4 mm/s compared to a RMS value of 6 mm/s for the range-rate signal. These values are 0.01 and 2.3 mm/s when ignoring the spectrum for frequencies below two cycles per orbital revolution, reducing the discrepancy even further to about 0.5% of the signal. The selected linear perturbation theory is thus capable of modeling gravity field induced range-rate observations with very high precision for a large part of the spectrum. 相似文献

Basins within the African sector of Gondwana contain a Late Palaeozoic to Early Mesozoic Gondwana sequence unconformably
overlying Precambrian basement in the interior and mid-Palaeozoic strata along the palaeo-Pacific margin. Small sea-board
Pacific basins form an exception in having a Carboniferous to Early Permian fill overlying Devonian metasediments and intrusives.
The Late Palaeozoic geographic and tectonic changes in the region followed four well-defined consecutive events which can
also be traced outside the study area. During the Late Devonian to Early Carboniferous period (up to 330 Ma) accretion of
microplates along the Patagonian margin of Gondwana resulted in the evolution of the Pacific basins. Thermal uplift of the
Gondwana crust and extensive erosion causing a break in the stratigraphic record characterised the period between 300 and
330 Ma. At the end of this period the Gondwana Ice Sheet was well established over the uplands. The period 260–300 Ma evidenced
the release of the Gondwana heat and thermal subsidence caused widespread basin formation. Late Carboniferous transpressive
strike-slip basins (e.g. Sierra Australes/Colorado, Karoo-Falklands, Ellsworth-Central Transantarctic Mountains) in which
thick glacial deposits accumulated, formed inboard of the palaeo-Pacific margin. In the continental interior the formation
of Zambesi-type rift and extensional strike-slip basins were controlled by large mega-shear systems, whereas rare intracratonic
thermal subsidence basins formed locally. In the Late Permian the tectonic regime changed to compressional largely due to
northwest-directed subduction along the palaeo-Pacific margin. The orogenic cycle between 240 and 260 Ma resulted in the formation
of the Gondwana fold belt and overall north–south crustal shortening with strike-slip motions and regional uplift within the
interior. The Gondwana fold belt developed along a probable weak crustal zone wedged in between the cratons and an overthickened
marginal crustal belt subject to dextral transpressive motions. Associated with the orogenic cycle was the formation of mega-shear
systems one of which (Falklands-East Africa-Tethys shear) split the supercontinent in the Permo-Triassic into a West and an
East Gondwana. By a slight clockwise rotation of East Gondwana a supradetachment basin formed along the Tethyan margin and
northward displacement of Madagascar, West Falkland and the Gondwana fold belt occurred relative to a southward motion of
Africa.

Under apartheid, tourism was essentially anti-developmental in focus and tourism research largely centred on the recreation
of white South Africans. Since the 1994 democratic transition tourism has become recognised as an increasingly important sector
for South Africa's economy and for achieving the government's goals for reconstruction and development. In this paper a review
and critical analysis is presented of the changing directions and foci of tourism scholarship in South Africa. It is shown
that South African scholars are beginning to respond to the challenges posed by tourism as a developmental focus.

Recent hydro‐climatological trends and variability characteristics were investigated for the Lake Naivasha basin with the aim of understanding the changes in water balance components and their evolution over the past 50 years. Using a Bayesian change point analysis and modified Mann–Kendall tests, time series of annual mean, maximum, minimum, and seasonal precipitation and flow, as well as annual mean lake volumes, were analysed for the period 1960–2010 to uncover possible abrupt shifts and gradual trends. Double cumulative curve analysis was used to investigate the changes in hydrological response attributable to either human influence or climatic variability. The results indicate a significant decline in lake volumes at a mean rate of 9.35 × 106 m3 year?1. Most of the river gauging stations showed no evidence of trends in the annual mean and maximum flows as well as seasonal flows. Annual minimum flows, however, showed abrupt shifts and significant (upward/downward) trends at the main outlet stations. Precipitation in the basin showed no evidence of abrupt shifts, but a few stations showed gradual decline. The observed changes in precipitation could not explain the decline in both minimum flows and lake volumes. The findings show no evidence of any impact of climate change for the Lake Naivasha basin over the past 50 years. This implies that other factors, such as changes in land cover and infrastructure development, have been responsible for the observed changes in streamflow and lake volumes.
