The state-of-the-art monitoring of drinking water hygiene is based on the cultivation and enumeration of indicator bacteria. Despite its proven reliability, this approach has the disadvantages of being (a) relatively slow and (b) limited to a small number of indicator bacteria. Ideally, alternative methods would be less time-consuming while providing information about a larger set of hygienically relevant microorganisms including viruses. In this paper, we present insights into the design of a modular concentration and detection system for bacteria, bacteriophages and viruses. Following further validation, this or similar techniques have the potential to extend and speed up the monitoring of raw and drinking water hygiene in the future. The system consists of different modules for the concentration of microorganisms, an amplification and detection unit that includes a module for the differentiation between live and dead microorganisms, and an automated system for decision support and self-diagnosis. The ongoing testing under controlled laboratory conditions and real-life conditions in the water supply industry yields further system improvements. Moreover, the increased sensitivity and broader range of microbiological parameters emphasize the need for a reconsideration of the currently used criteria for the assessment of (drinking) water hygiene.  相似文献

A major substratum for vent organisms on Juan de Fuca and Explorer Ridges of the northeast Pacific is the polymetallic sulphide chimney. The deposition processes and subsequent growth of such chimneys provide a dynamic and extreme habitat that changes rapidly. During initial venting stages, colonization must await stabilization of the chimney structure. As the porous anhydrite shell accumulates sulphide minerals, protection from direct hot water contact increases for organisms colonizing the outer walls while sufficient vent water still diffuses for the support of chemosynthesis. Vestimentiferan worms are found on small sulphide mounds but the active growth and collapse of anhydrite spires is a major source of disruption and mortality. High temperature venting and spire growth appear to attract alvinellid polychaetes which may be implicated in the process of strengthening and sealing the anhydrite spires. Fauna on the growing chimney is subject to changes in fluid flow patterns that can make the structure uninhabitable. Large chimneys are inhabited by more species indicating that a diversification of habitat results from variations in mineralization and fluid availability. Chimney fauna has become specialized in exploiting an unpredictable and unstable habitat in an extreme example of the control of biotic development by the physical environment.  相似文献

Ocean drilling has revealed the existence of vast microbial populations in the deep subseafloor, but to date little is known about their metabolic activities. To better understand the biogeochemical processes in the deep biosphere, we investigate the stable carbon isotope chemistry of acetate and other carbon-bearing metabolites in sediment pore-waters. Acetate is a key metabolite in the cycling of carbon in anoxic sediments. Its stable carbon isotopic composition provides information on the metabolic processes dominating acetate turnover in situ. This study reports our findings for a methane-rich site at the northern Cascadia Margin (NE Pacific) where Expedition 311 of the Integrated Ocean Drilling Program (IODP) sampled the upper 190 m of sediment. At Site U1329, δ13C values of acetate span a wide range from −46.0‰ to −11.0‰ vs. VPDB and change systematically with sediment depth. In contrast, δ13C values of both the bulk dissolved organic carbon (DOC) (−21.6 ± 1.3‰ vs. VPDB) and the low-molecular-weight compound lactate (−20.9 ± 1.8‰ vs. VPDB) show little variability. These species are interpreted to represent the carbon isotopic composition of fermentation products. Relative to DOC, acetate is up to 23.1‰ depleted and up to 9.1‰ enriched in 13C. Broadly, 13C-depletions of acetate relative to DOC indicate flux of carbon from acetogenesis into the acetate pool while 13C-enrichments of pore-water acetate relative to DOC suggest consumption of acetate by acetoclastic methanogenesis. Isotopic relationships between acetate and lactate or DOC provide new information on the carbon flow and the presence and activity of specific functional microbial communities in distinct biogeochemical horizons of the sediment. In particular, they suggest that acetogenic CO2-reduction can coexist with methanogenic CO2-reduction, a notion contrary to the hypothesis that hydrogen levels are controlled by the thermodynamically most favorable electron-accepting process. Further, the isotopic relationship suggests a relative increase in acetate flow to acetoclastic methanogenesis with depth although its contribution to total methanogenesis is probably small. Our study demonstrates how the stable carbon isotope biogeochemistry of acetate can be used to identify pathways of microbial carbon turnover in subsurface environments. Our observations also raise new questions regarding the factors controlling acetate turnover in marine sediments.  相似文献

Despite a long history of related research, quantifying and verifying recharge is still a major challenge. The combination and comparison of conceptually different methods has been recommended as a strategy for evaluating recharge estimates. In this article, recharge estimates from water-table fluctuation (WTF) methods are combined with and compared to the results of the spatially and temporally discretized soil-water-balance model PROMET (processes of radiation, mass and energy transfer). As PROMET and WTF methods rely on different measurable variables, a comparison of these two contrasting techniques allows improved assessment of the plausibility of recharge estimates. An enhanced approach to WTF methods is presented. The approach assumes that in the case of no recharge, there exists a maximum possible potential decline for any given groundwater level. The primary conclusion is that WTF methods are excellent for determining the plausibility of spatially distributed regional-groundwater-recharge estimation approaches and for detecting inconsistencies in available models. Recharge estimates derived from WTF approaches alone are, however, not suitable for regional-scale recharge estimation due to (1) their strong dependency on local data, applicability of which is limited to only very specific conditions, and (2) their sensitivity to influences other than recharge.  相似文献

The monoaromatic and total aromatic hydrocarbon fractions of two pairs of undegraded and moderately biodegraded crude oils from the Santa Maria basin (California) and the Vienna basin (Austria), all dominated by unresolved complex mixtures, were studied regarding their composition and toxicity towards the feeding rate of the marine mussel Mytilus edulis. Total aromatic and monoaromatic hydrocarbon fractions from sulphur-rich Monterey Formation crude oils were slightly more toxic than the fractions isolated from sulphur-lean Vienna basin oils. The ecotoxicity tests did not show any significant differences in toxicity of aromatic compounds from undegraded or in-reservoir biodegraded crude oils from the same oilfield although some differences in composition were observed. Organic sulphur compounds are suspected to cause the slightly higher toxicity of the aromatic hydrocarbon fractions from the Monterey oils.  相似文献
