The active lifetime of extracellular enzymes is a critical determinant of the effectiveness of enzyme production as a means for heterotrophic marine microbes to obtain organic substrates. Here, we report lifetimes of three classes of extracellular enzyme in Arctic seawater. We also investigated the relative importance of photochemical processes and particle-associated processes in inactivating extracellular enzymes. Enzyme inactivation in filtered seawater was slow, with apparent half-lives of enzyme activities on the order of hundreds of hours. The presence of particles (including cells) did not significantly change inactivation rates, suggesting that the long half-lives observed in filtered seawater were realistic for enzymes in unfiltered seawater. Phosphatase and leucine aminopeptidase were susceptible to photoinactivation, but only under high intensity UV-B and UV-C illumination; there was no evidence for increased inactivation rates under natural illumination at our study site in Ny Ålesund, Svalbard. Comparison of inactivation rates of commercially-obtained enzymes from non-marine sources with the extracellular enzymes naturally present in Arctic seawater suggests that the natural enzymes contain structural features that confer longer lifetimes, consistent with observations reported by others from a range of field sites that cell-free enzymes can contribute a substantial fraction of total hydrolytic activity in the water column.  相似文献

The interaction between a gaining stream and a water-table aquifer is studied at an outwash plain. The aquifer is hydraulically well connected to the stream. Pumping tests were carried out in 1997 and 1998 in two wells 60 m from the stream, screening different depths of the aquifer. Drawdown was measured on both sides of the stream. Hydraulic head, drawdown, and stream depletion data were analyzed using numerical flow models. Similar models were fitted to each of two different data sets: Model A was fitted to steady-state hydraulic head and streamflow gain data not influenced by pumping; and model B was fitted to drawdown data measured during the 1998 pumping test. Each calibrated model closely fits its calibration data; however, predictions were biased if model A was used to predict the calibration data of model B, and vice versa. To further test the models, they were used to predict streamflow depletion during the two pumping tests as well as the drawdown during the 1997 test. Neither of these data were used for calibration. Model A predicted the measured depletions fairly accurately during both tests, whereas the predicted drawdowns in 1997 were significantly larger than actually measured. Contrary to this, the 1997 drawdowns predicted by model B were nearly unbiased; the predicted depletions deviate significantly from the measured depletions in 1997, but they compare well with the observations in 1998. Thus, although field work and analyses were extensive and done carefully to develop a ground water flow model that could predict both drawdown and streamflow depletion, the model predictions are biased. Analyses indicate that the deviations between model and data may be because of error in the models' representations of either the release of water from storage or of the hydrology in the riparian zone.  相似文献

Groundwater models need to account for detailed but generally unknown spatial variability (heterogeneity) of the hydrogeologic model inputs. To address this problem we replace the large, m-dimensional stochastic vector β that reflects both small and large scales of heterogeneity in the inputs by a lumped or smoothed m-dimensional approximation γθ, where γ is an interpolation matrix and θ is a stochastic vector of parameters. Vector θ has small enough dimension to allow its estimation with the available data. The consequence of the replacement is that model function f(γθ) written in terms of the approximate inputs is in error with respect to the same model function written in terms of β, f(β), which is assumed to be nearly exact. The difference f(β) − f(γθ), termed model error, is spatially correlated, generates prediction biases, and causes standard confidence and prediction intervals to be too small. Model error is accounted for in the weighted nonlinear regression methodology developed to estimate θ and assess model uncertainties by incorporating the second-moment matrix of the model errors into the weight matrix. Techniques developed by statisticians to analyze classical nonlinear regression methods are extended to analyze the revised method. The analysis develops analytical expressions for bias terms reflecting the interaction of model nonlinearity and model error, for correction factors needed to adjust the sizes of confidence and prediction intervals for this interaction, and for correction factors needed to adjust the sizes of confidence and prediction intervals for possible use of a diagonal weight matrix in place of the correct one. If terms expressing the degree of intrinsic nonlinearity for f(β) and f(γθ) are small, then most of the biases are small and the correction factors are reduced in magnitude. Biases, correction factors, and confidence and prediction intervals were obtained for a test problem for which model error is large to test robustness of the methodology. Numerical results conform with the theoretical analysis.  相似文献

During December 1982, a novel Fabry-Perot interferometer—a Doppler Imaging System (DIS)— was used at Kiruna Geophysical Institute (KGI), Sweden (67.8°N, 21.2°E) to complement a series of coordinated observations of global thermospheric dynamics utilizing a number of conventional ground-based Fabry-Perot interferometers and the NASA Dynamics Explorer satellite. The DIS is an interferometer with two unique attributes : it has a luminosity or étendue more than one hundred times that of the conventional Fabry-Perot interferometer, and it is also capable of deducing a two-dimensional velocity field of a suitable line-emitting areal source by independently measuring the Doppler shift at a large number of points within the field of view. On 17 December 1982, a very large geomagnetic Storm Sudden Commencement (08.05 U.T.) preceded a major geomagnetic disturbance. During this disturbance, Northern Scandinavia was influenced by a strong eastward auroral electrojet for an extended period (10–19 U.T.). The DIS was able to observe the dynamical response of the upper thermosphere to this event in conjunction with a second Fabry-Perot interferometer (FPI) at KGI. Westward thermospheric winds of about 900 m s?1 occurred during the disturbance and, at the peak of the disturbance, the combined DIS and FPI observations indicate that the thermospheric flow was quite chaotic. Fluctuations of the order of ± 150 ms?1, associated with spatial scales of the order of 100 or 200 km occurred within the mean westward flow inside the 800 km diameter region observed from Kiruna.  相似文献

A fixed link (tunnel and bridge, in total 16 km) was constructed between Sweden and Denmark during 1995-2000. As part of the work, approximately 16 million tonnes of seabed materials (limestone and clay till) were dredged, and about 0.6 million tonnes of these were spilled in the water. Modelling of the spreading and sedimentation of the spilled sediments took place as part of the environmental monitoring of the construction activities. In order to verify the results of the numerical modelling of sediment spreading and sedimentation, a new method with the purpose of distinguishing between the spilled sediments and the naturally occurring sediments was developed. Because the spilled sediments tend to accumulate at the seabed in areas with natural sediments of the same size, it is difficult to separate these based purely on the physical properties. The new method is based on the geo-chemical differences between the natural sediment in the area and the spill. The basic propertiesused are the higher content of calcium carbonate material in the spill as compared to the natural sediments and the higher Ca/Sr ratio in the spill compared to shell fragments dominating the natural calcium carbonate deposition in the area. The reason for these differences is that carbonate derived from recent shell debris can be discriminated from Danien limestone, which is the material in which the majority of the dredging took place, on the basis of the Ca/Sr ratio being 488 in Danien Limestone and 237 in shell debris. The geochemical recognition of the origin of the sediments proved useful in separating the spilled from the naturally occurring sediments. Without this separation, validation of the modelling of accumulation of spilled sediments would not have been possible. The method has general validity and can be used in many situations where the origin of a given sediment is sought.  相似文献

We analyze the optimal design of a pumping test for estimating hydrogeologic parameters that are subsequently used to predict stream depletion caused by groundwater pumping in a leaky aquifer. A global optimization method is used to identify the test’s optimal duration and the number and locations of observation wells. The objective is to minimize predictive uncertainty (variance) of the estimated stream depletion, which depends on the sensitivities of depletion and drawdown to relevant hydrogeologic parameters. The sensitivities are computed analytically from the solutions of Zlotnik and Tartakovsky [Zlotnik, V.A., Tartakovsky, D.M., 2008. Stream depletion by groundwater pumping in leaky aquifers. ASCE Journal of Hydrologic Engineering 13, 43–50] and the results are presented in a dimensionless form, facilitating their use for planning of pumping test at a variety of sites with similar hydrogeological settings. We show that stream depletion is generally very sensitive to aquitard’s leakage coefficient and stream-bed’s conductance. The optimal number of observation wells is two, their optimal locations are one close to the stream and the other close to the pumping well. We also provide guidelines on the test’s optimal duration and demonstrate that under certain conditions estimation of aquitard’s leakage coefficient and stream-bed’s conductance requires unrealistic test duration and/or signal-to-noise ratio.  相似文献
