We have previously demonstrated that medaka CYP3A is associated with metabolism of several endobiotics including steroids and bile acids. In this study, we demonstrate that medaka CYP3A catalysis exhibits unusual kinetic behaviors consistent with allosteric interaction which cannot be described by hyperbolic kinetic models. Using 7-benzyloxy-4-(trifluoromethyl)-coumarin (BFC) and nonylphenol as CYP3A substrates, we describe both homotropic and heterotropic cooperative interactions. Given the role of CYP3A in maintaining the homeostatic balance for numerous endobiotics, enzymatic activation/inhibition by endocrine disruptors (EDCs) represents a putative (non-genomic) mechanism for endocrine disruption. 相似文献

In this paper, we present a system capable of simultaneously estimating the position of an autonomous underwater vehicle (AUV) and the positions of stationary range-only beacons. Notably, our system does not require beacon positions a priori, and our system performs well even when range measurements are severely degraded by noise and outliers. We present a powerful outlier rejection method that can identify groups of range measurements that are consistent with each other, and a method for initializing beacon positions in an extended Kalman filter (EKF). We have successfully applied our algorithms to real-world data and have demonstrated a simultaneous localization and mapping (SLAM) system whose navigation performance is comparable to that of systems that assume known beacon locations 相似文献

At least five Middle to Late Pleistocene advances of the northern Cordilleran Ice Sheet are preserved at Silver Creek, on the northeastern edge of the St Elias Mountains in southwest Yukon, Canada. Silver Creek is located 100 km up‐ice of the Marine Isotope Stage (MIS) 2 McConnell glacial limit of the St Elias lobe. This site contains ~3 km of nearly continuous lateral exposure of glacial and non‐glacial sediments, including multiple tills separated by thick gravel, loess and tilted lake beds. Infrared‐stimulated luminescence (IRSL) and AMS radiocarbon dating constrain the glacial deposits to MIS 2, 4, either MIS 6 or mid‐MIS 7, and two older Middle Pleistocene advances. This chronology and the tilt of the lake beds suggest Pleistocene uplift rates of up to 1.9 mm a?1 along the Denali Fault since MIS 7. The non‐glacial sediment consists of sand, gravel, loess and organic beds from MIS 7, MIS 3 and the early Holocene. The MIS 3 deposits date to between 30–36 14C ka BP, making Silver Creek one of the few well‐constrained MIS 3‐aged sites in Yukon. This confirms that ice receded close to modern limits in MIS 3. Pollen and macrofossil analyses show that a meadow‐tundra to steppe‐tundra mosaic with abundant herbs and forbs and few shrubs or trees, dominated the environment at this time. The stratigraphy at Silver Creek provides a palaeoclimatic record since at least MIS 8 and comprises the oldest direct record of Pleistocene glaciation in southwest Yukon. 相似文献

Stream water temperature plays a significant role in aquatic ecosystems where it controls many important biological and physical processes. Reliable estimates of water temperature at the daily time step are critical in managing water resources. We developed a parsimonious piecewise Bayesian model for estimating daily stream water temperatures that account for temporal autocorrelation and both linear and nonlinear relationships with air temperature and discharge. The model was tested at 8 climatically different basins of the USA and at 34 sites within the mountainous Boise River Basin (Idaho, USA). The results show that the proposed model is robust with an average root mean square error of 1.25 °C and Nash–Sutcliffe coefficient of 0.92 over a 2‐year period. Our approach can be used to predict historic daily stream water temperatures in any location using observed daily stream temperature and regional air temperature data. 相似文献

We propose a numerical method that couples a cohesive zone model (CZM) and a finite element‐based continuum damage mechanics (CDM) model. The CZM represents a mode II macro‐fracture, and CDM finite elements (FE) represent the damage zone of the CZM. The coupled CZM/CDM model can capture the flow of energy that takes place between the bulk material that forms the matrix and the macroscopic fracture surfaces. The CDM model, which does not account for micro‐crack interaction, is calibrated against triaxial compression tests performed on Bakken shale, so as to reproduce the stress/strain curve before the failure peak. Based on a comparison with Kachanov's micro‐mechanical model, we confirm that the critical micro‐crack density value equal to 0.3 reflects the point at which crack interaction cannot be neglected. The CZM is assigned a pure mode II cohesive law that accounts for the dependence of the shear strength and energy release rate on confining pressure. The cohesive shear strength of the CZM is calibrated by calculating the shear stress necessary to reach a CDM damage of 0.3 during a direct shear test. We find that the shear cohesive strength of the CZM depends linearly on the confining pressure. Triaxial compression tests are simulated, in which the shale sample is modeled as an FE CDM continuum that contains a predefined thin cohesive zone representing the idealized shear fracture plane. The shear energy release rate of the CZM is fitted in order to match to the post‐peak stress/strain curves obtained during experimental tests performed on Bakken shale. We find that the energy release rate depends linearly on the shear cohesive strength. We then use the calibrated shale rheology to simulate the propagation of a meter‐scale mode II fracture. Under low confining pressure, the macroscopic crack (CZM) and its damaged zone (CDM) propagate simultaneously (i.e., during the same loading increments). Under high confining pressure, the fracture propagates in slip‐friction, that is, the debonding of the cohesive zone alternates with the propagation of continuum damage. The computational method is applicable to a range of geological injection problems including hydraulic fracturing and fluid storage and should be further enhanced by the addition of mode I and mixed mode (I+II+III) propagation. This paper studies the risk of collision between asteroids and Earth. It focuses on uncertainty in the human consequences of asteroid collisions, with emphasis on the possibility of global catastrophe to human civilization. A detailed survey of the asteroid risk literature shows that while human consequences are recognized as a major point of uncertainty, the studies focus mainly on physical and environmental dimensions of the risk. Some potential human consequences are omitted entirely, such as the possibility of asteroid explosions inadvertently causing nuclear war. Other human consequences are modeled with varying degrees of detail. Direct medical effects are relatively well-characterized, while human consequences of global environmental effects are more uncertain. The latter are evaluated mainly in terms of a global catastrophe threshold, but such a threshold is deeply uncertain and may not even exist. To handle threshold uncertainty in asteroid policy, this paper adapts the concept of policy boundaries from literature on anthropogenic global environmental change (i.e., planetary boundaries). The paper proposes policy boundaries of 100 m asteroid diameter for global environmental effects and 1 m for inadvertent nuclear war. Other policy implications include a more aggressive asteroid risk mitigation policy and measures to avoid inadvertent nuclear war. The paper argues that for rare events like large asteroid collisions, the absence of robust data means that a wide range of possible human consequences should be considered. This implies humility for risk analysis and erring on the side of caution in policy. 相似文献
