Our goal was to study the involvement of cytochrome P450 genes in estrogen metabolism and the extent to which the potentially carcinogenic 4-hydroxyestradiol metabolite is formed by channel catfish (Ictalurus punctatus; CC). Estradiol metabolism and ethoxyresorufin-O-deethylase (EROD) activity were assessed in several tissues from fish collected from three variably contaminated sites in the Mississippi River Delta, from laboratory control fish, and from fish exposed to 20 mg/kg benzo(a)pyrene (BaP) i.p. for 4 days. Liver EROD activity was induced by BaP, but Delta fish EROD activities were not statistically higher than activities in control fish. Gill microsomal EROD activity was also induced by BaP, but activities were 8- to 77-fold lower than those from liver. The predominant estrogen metabolites formed by CC liver, gill and gonad microsomes were 2-hydroxyestradiol and estrone as detected by GC/MS. Liver and gill 2-hydroxyestradiol formation was induced in BaP-dosed fish. The trends in hydroxyestradiol formation in field collected fish were more variable. In all fish liver microsomes there was more 2- compared to 4-hydroxyestradiol formed. However, BaP-treatment increased the 4:2 hydroxyestradiol ratio from 0.04 in control fish to 0.2 in BaP-exposed fish, suggesting that BaP induces the formation of the potentially genotoxic estrogen metabolite. No detectable 4-hydroxyestradiol was produced by gill and gonad microsomes. These results will ultimately help in determining which fish P450 genes are susceptible to environmental contamination and may be involved in estrogen genotoxicity.  相似文献

Marine geology of the Medriff Corridor, Mediterranean Ridge   总被引:1,自引:0,他引:1  
Abstract Thirty-one piston and/or gravity cores not exceeding 10 m in length were raised in selected areas of a 300 km-long transect (Medriff Corridor) crossing the Mediterranean Ridge, an accretionary complex subject to continent/continent collision, containing an important evaporitic body (Messinian evaporites), in order to ground-truth the geological make-up. Core location, very accurate with reference to a complex bottom configuration, was preceded by swathe mapping, seismic profiling and side-scan sonar investigations. Most sediment cores have a pelagic facies, with biogenic marls as dominant lithology, and sapropels and tephras as minor, isochronous lithologies. A combination of isochronous lithologies and calcareous plankton biochronology permits high resolution stratigraphic correlations. Pelagic facies sediments are Middle Pleistocene to Holocene in age. Two cores associated with mounds located along thrusts contain a matrix-supported mud breccia of deep provenance, Burdigalian-Langhian in age, similar to that characteristic of the Mediterranean Ridge diapiric belt (Cita et al. 1995 ). Three new brine-filled anoxic basins (Urania, l'Atalante and Discovery) were discovered. The brines originated from submarine dissolution of Messinian evaporites and are different in the various basins. The sedimentary record strongly differs from basin to basin. These brine lakes are very young (35 000 years or less). A drastic change in sedimentation rate recorded in the Discovery Basin suggests that basin collapse was sudden and followed by progressive development of basin anoxia. Some cores were analyzed with a prototype multisensor for P-wave velocity, magnetic susceptibility and density. Sapropels show up as abrupt decreases in P-wave velocity and density, and tephra as sudden increases in magnetic susceptibility. Mud breccia displays P-wave velocities greater than pelagic marls, with peaks related to lithic clasts. Anoxic sediments have high P-wave velocities; peaks are associated with gypsum crystals.  相似文献

Methoxychlor (MXC) has been shown to possess estrogenic activity in mammals and fish. Although MXC does not appear to appreciably bind the mammalian estrogen receptor, its demethylated metabolites have been shown to be significantly more potent agonists and are believed responsible for estrogenic effects in mammals following exposure to this pesticide. To determine whether catfish were capable of MXC demethylotion, and, hence, activation to a more estrogenic compound, in vitro biotransformation studies were carried out using hepatic microsomes from mature male channel catfish. Hepatic microsomes catalyzed the NADPH-dependent formation of monodemethylated (mono-MXC) and bisdemethylated (bis-MXC) metabolites of MXC. Treatment with mono-MXC at 40% of the MXC dose in catfish significantly induced serum vitellogenin (Vg) levels compared to MXC. Estrogen receptor binding studies in catfish liver cytosol showed that a racemic mixture of the mono-MXC had approximately 43 times the affinity for the receptor than MXC, but was still over 1000-fold less potent that 17β-estradiol. These results demonstrate that catfish are capable of biochemically activating MXC to a more potent hepatic estrogen receptor agonist.  相似文献
