Response surface methodology was used to optimize a medium for a red-pigmented marine bacterium S-9801 strain (Flavobocterium sp.). In the first optimization step the influence of yeast extract, peptone, glucose and sodium chloride on pigment production was evaluated using a fractional factorial design. Pigment production was positively influenced by glucose and sodium chloride while other components had no significant effect. In the second step the path of steepest ascent was used to approach the optimal region of the medium composition. In the third step the optimal concentration of glucose and sodium chloride was determined by a central composite design and response analysis. The optimized medium allowed pigment production (A 535-650) to be increased from 0.137 to 0.559, being 320% higher than the original medium. 相似文献

This paper concentrates on the practices and possibilities of doing poststructural cultural geographies of nature. The paper begins with an overview of the ways in which nature is being reconceptualised in social thought. It charts a middle territory between strong social constructivism and realist approaches, arguing for a relational understanding of nature and culture, and the extension of agency to non-humans--organic and artificial. These 'more than human worlds' (<citeref rid="b51">Whatmore, 'Hybrid geographies'</citeref>, Sage, London, 2002) pose considerable challenges for researchers. They open spaces for new subjects and new subjectivities, decentre human agency, and challenge more conventional methodological approaches that rely on a separation between subject and object. The paper considers the sorts of methodological shifts required for relational analysis, arguing for a multilayered, multivalent, embodied and situated approach. The practices of situatedness are emphasised in the Australian context, especially in relation to cross-cultural and postcolonial understandings. To collect time-use data in Third World rural areas, researchers should use broad-focus studies that provide adequate coverage throughout the year and employ direct observation of activities. Various methods used by geographers to obtain time expenditure information are examined in relation to these criteria; certain weaknesses are discussed. An alternative method developed by Allen Johnson, an anthropologist, has particular advantages for geographers because it facilitates mapping spatial patterns of time use, calculating average distances traveled to activities, and computing time spent on movement. 相似文献

Ninety mainstream quantitative methods used in 14 major geographical journals are analyzed for four selected years: 1956, 1966, 1976, and 1986. Mainstream methods are used in 53% of all articles in 1986, as opposed to 3% in 1956. Both the intensity and variety of methods used also increased, particularly in physical and techniques articles compared to human articles. Major methods that dominate the entire period include bivariate correlation and regression, multiple regression, factor analyses, and chi-square. The use of nonparametric methods is increasing, although their frequency of occurrence is still low. Spatial autocorrelation and advanced multiple regression appear to be strong newcomers to the discipline. 相似文献
