克隆编码红笛鲷(Lutjanus sanguineus)RAG1蛋白(recombination activating protein1)活性核心区的基因序列,并与pET-28a(+)载体连接,构建原核表达载体pET-28a-RAG1,将其转入大肠杆菌BL21(DE3)菌株,利用IPTG进行诱导表达。为提高融合蛋白的表达效率,运用传统的实验方法对诱导条件进行优化。SDS-PAGE分析表明,在37℃条件下,利用0.1 mmol/L IPTG诱导8 h后,RAG1重组融合蛋白的表达量最大,相对分子质量与预测值相符,该蛋白主要以包涵体形式高效表达,利用His Trap HP亲和柱使其得到进一步纯化;Western blot分析显示,该融合蛋白可与鼠抗His-tag单克隆抗体发生特异性结合,说明表达蛋白为目的蛋白。  相似文献

理论解析以非农就业程度表征的农户生计转型和以人均农村居民点用地面积度量的农村土地利用特征的相互关系,应用GIS技术和空间自相关分析方法研究中国县域尺度农户生计转型的存在性、拐点值及其主导下的农村居民点用地整治适宜区域,为规范推进农村居民点用地整治与重构乡村地域的可持续性提供决策参考。结果表明,农户非农就业程度与人均农村居民点用地面积存在倒U型规律;资源禀赋、经济发展阶段、地形等外部因子影响农户生计与农村土地利用的相互关系;控制资源禀赋、地形等外部因子作用的农户非农就业程度拐点为38.36%,结合人均农村居民点用地规模,可将中国县域划分为适宜程度依次递减的优先整治区、优化调整区、适度挖潜区与限制整治区等4 种类型区;最后,提出了切实反映农户生计特征的区域农村居民点用地整治政策建议。  相似文献

“Stone runs” is the Falklands vernacular term for openwork boulder accumulations, which include extensive blockstreams like the famous Darwin “stone-river” and associated features such as stone stripes. Since the early 20th century, they have been interpreted as the product of a suite of periglacial processes, including frost-wedging, gelifluction, frost heave, frost-sorting and snowmelt runoff. Following a literature review, the results of recent field investigations of the valley-floor blockstreams of East Falkland are presented. Access to the internal structure of these forms provides evidence for the existence of a three-fold profile, with clear vertical size gradation presenting striking similarities with an inverted weathering profile. Micromorphological analyses, SEM, XRD, thin sections and grain-size analyses lead to the hypothesis of an alternative model of stone run formation. It is suggested that the material forming the stone run profile lato sensu (including the superficial pavement) is not of periglacial origin, but derives directly from the stripping and accumulation downslope of a regolith, possibly Tertiary in age and formed under subtropical or temperate conditions. The valley-floor stone runs should, therefore, be considered as complex polygenetic landforms that may have formed according to a six-stage scenario, including in situ chemical weathering, regolith stripping by mass movements, soil formation, further regolith stripping, downslope accumulation and matrix washing-out (all phases possibly achieved by the Early Quaternary). Periglacial reworking of the stone run material would have operated at a “final” stage, i.e. during Quaternary cold stages, with boulder bioweathering and limonite-staining operating during the temperate intervals including the present one. The suggested antiquity of the Falklands blockstreams is in accordance with Caine's pioneer interpretation of Tasmania blockfields and with recent analyses and cosmogenic datings of blockfields from Scandinavia and North America.  相似文献

The Algodones Dune Field of southeastern California shows a complex and spatially diverse dune-field pattern that is superimposed upon a series of topographic lineations. Analysis of dune-field pattern parameters (dune crest length, crest orientation, dune spacing and defect density) derived from aerial images indicates that the dune-field pattern represents two constructional generations. Prominent compound crescentic dunes formed during the first constructional generation. A younger generation consists of a variety of simple crescentic dunes, linear dunes and zibars. Statistical differences in the pattern parameters between the dune groups within the second generation are resolved through consideration of the boundary conditions under which the dune pattern evolved, and provide explanations for: (1) diversity of dune types, (2) range in implied constructional times, (3) range in crest orientations, and (4) the anomalous nature of the population of linear dunes. The boundary conditions that have modified pattern development include orographic effects, grain size, vegetation, areal extent and antecedent conditions. Topographic lineations in the Algodones range from the Western Ramp, which defines the field margin, to subtle features masked by the pattern of dunes. Imaging of the Western Ramp using Ground Penetrating Radar shows high-angle cross-strata migrating perpendicular to the lineation trend. The most plausible hypothesis for the origin of the lineations is as dune ridges sequentially shed from adjacent Lake Cahuilla, which is the source of Algodones sands. The overall geomorphic complexity of the Algodones originates from the emplacement of the dune ridges during stages of Lake Cahuilla, the two generations of dune-field construction, and the controls exerted by boundary conditions.  相似文献

The larval shells of Paphies ventricosa, reared to settlement in the hatchery, are similar in longitudinal profile to shells of some other mactracean larvae; the umbo‐stage larval shells are nearly round with the anterior margin less broadly curved than the posterior margin, the anterior end is longer than the posterior end, and the umbo is broadly rounded. Straight‐hinge larvae had total lengths up to 149 μm, and total heights up to 126 μm, and umbo larvae had total lengths up to 320 μm, and total heights up to 282 μm. The larvae settled in about 22 days at a size of about 270 μm. The larval hinge structure bears some similarities to those of other mactracean larvae.  相似文献

An attempt is made to delineate the relative performances and credentials of GFS, FNL, and NCMRWF global analyses/forecast products as initial and boundary conditions (IBCs) to the WRF-ARW model in the simulation of four Bay of Bengal tropical cyclones (TCs). The results suggest that FNL could simulate horizontal advection of vorticity maxima at 850 hPa; hence, the tracks are more realistic with least errors as compared to GFS and NCMRWF. The mean landfall errors for 24-, 48-, and 72-hour forecasts are 73, 41, and 72 km, respectively. The TC intensity is well captured by NCMRWF IBCs, as it could predict 850 hPa vorticity maxima. The 24-hour accumulated rainfall is well simulated with FNL, and equitable threat score is more than 0.2 up to 100 mm with minimum bias.  相似文献

Routine monitoring of the quality status of water bodies demands the best cost‐benefit relation and sample‐size reduction is therefore welcomed. However, great caution is needed because such reduction affects the accuracy and variation of the results. In the present study we tested the influence of sample size (number of replicate samples) on reference condition values and within‐sample ecological quality ratio (EQR) variability of six commonly used ecological indices (taxa richness, Shannon–Wiener diversity, AMBI, Medocc index, Bentix and M‐AMBI). Analysis of soft‐bottom benthic invertebrate data from Slovenian coastal waters showed that sample size influenced the reference condition values of richness/diversity indices (taxa richness and Shannon–Wiener diversity) but not of the sensitivity/tolerance indices (AMBI, Medocc index, Bentix). Increased sample size decreased the within‐sample EQR variability and, concomitantly, increased the accuracy of site ecological status classification for all indices. The size of EQR variability differed depending on the index used. EQR variability of M‐AMBI, an index composed of metrics with different within‐sample EQR variability, was statistically the same as that of the metric with the lowest within‐sample EQR variability. Whether this is a common principle for multimetric indices remains to be confirmed. Based on these results, the use of at least three replicates is suggested to obtain reliable measures of reference condition and EQR for the assessment of ecological status. This level of replication is particularly necessary in areas with high diversity and environmental patchiness, and when richness/diversity measures and indices that include these measures are used.  相似文献

Grow-out studies of the scallop Pecten sulcicostatus, endemic to the South African coast, contribute to an investigation of the suitability of this species for commercial culture. Hatchery-reared juvenile scallops were placed in a suspended culture system at 5 m depth in Saldanha Bay on the west coast of South Africa. Scallops of 78 days old and ranging in size from 4.5 mm to 11.0 mm shell height (mean 6.9 mm), as measured on 2 February 2010, were deployed in Saldanha Bay on 9 February 2010. Subsequent growth was assessed monthly through increments in shell height in relation to changing environmental conditions as determined through continuous measures of temperature and chlorophyll a. Upon termination of the experiment on 15 February 2011, scallops ranged in size from 42.1 mm to 48.7 mm (mean 45.1 mm), representing an increment in shell height of 38.2 mm over one year. The mean growth rate of 0.10 mm day?1 (mean specific growth rate of 0.0046 day?1) compares favourably with other commercially cultured species and exceeds previous estimates of growth of naturally occurring populations of P. sulcicostatus. Scallop growth was poorly correlated with either temperature or chlorophyll a concentration, but scallop mortality was closely aligned to the temperature regime of Saldanha Bay, exhibiting high mortalities during mid-summer.  相似文献
