Formation and early development of tetraspores of Polysiphonia urceolata (Rhodomelaceae, Rhodophyta)
Polysiphonia urceolata is one type of potential commercial red seaweeds used for breeding and cultivation, because of its significant biochemical
and biomedical application. However, the information of breeding and seedling incubation for cultivation is limited, especially
the early development. In this study, tetrasporohyte and gametophyte of P. urceolata were taken as the study materials in Huiquan Bay, Qingdao, China. The cleaned and sterilized tetrasporophytes and gametophytes
were pre-cultured in sterilized seawater, then nurtured at 18°C, 25 μmol photons m−2 s−1 in 12:12 h (light:dark) photoperiod. Continuous observation under microscope showed that the early development consists of
bipolar division stage and seedling stage. In the division stage, tetraspores germinate into bipolar sporelings that further
differentiate into a colorless rhizoidal portion and a lightly pigmented upright shoot. The lightly pigmented rhizoidal cell
develops to a rhizoid and the larger pigmented cell transforms to an erect axis. In the seedling stage, several quasi-protuberances
appear on the erect axis and form juvenile seedlings. The results demonstrate the culture of P. urceolata from tetraspores under laboratory conditions.

The effect of dietary phosphorus on the growth and body components of juvenile Synechogobius hasta was determined. Different percentages of dietary phosphorus (0.63, 0.77, 0.93, 1.06, 1.22 and 1.36) were tested by feeding the fish (body weight, 15.81 g ± 0.32 g; 20 individuals each group; 3 groups each percentage) at a surplus of 5%–10% above satiation for 35 d. Microsatellite markers have become one kind of the most important molecular tools used in various researches. A large number of microsatellite markers are required for the whole genome survey in the fields of molecular ecology, quantitative genetics and genomics. Therefore, it is extremely necessary to select several versatile, low-cost, efficient and time- and labor-saving methods to develop a large panel of microsatellite markers. In this study, we used Zhikong scallop (Chlamys farreri) as the target species to compare the efficiency of the five methods derived from three strategies for microsatellite marker development. The results showed that the strategy of constructing small insert genomic DNA library resulted in poor efficiency, while the microsatellite-enriched strategy highly improved the isolation efficiency. Although the mining public database strategy is time- and cost-saving, it is difficult to obtain a large number of microsatellite markers, mainly due to the limited sequence data of non-model species deposited in public databases. Based on the results in this study, we recommend two methods, microsatellite-enriched library construction method and FIASCO-colony hybridization method, for large-scale microsatellite marker development. Both methods were derived from the mi-crosatellite-enriched strategy. 研究β-1,3/1,6-D-glucan及不同投喂方式对中国明对虾(Fenneropenaeus chinensis)幼体的存活、生长、发育、非特异性免疫及抗副溶血弧菌能力的影响.将β-glucan以0.25%的剂量添加到微黏合饲料中,以不同的投喂方式投喂对虾幼体,即D0(一直投喂基础饲料);D1(一直投喂含0.25%β-葡聚糖饲料);3D1-3D0(3 d投喂含0.25%β-葡聚糖的饲料-3 d投喂基础饲料);1D1-3D0(1 d投喂含0.25%β-葡聚糖的饲料-3 d投喂基础饲料),共4个处理,每个处理随机设置3个重复,对虾幼体养殖于可控温的半开放系统中,每个养殖桶中盛有50 L盐度为30~32消毒海水,养殖密度为100尾/L.实验结果表明,在孵化后14 d,投喂D1饲料组在总存活率,变态指数,体长,特定生长率方面均高于其它各组,虽然有时差异不显著.在免疫指标方面,在9和14 dph分别进行了酚氧化酶(PO)、碱性磷酸酶(ALP)、超氧化物歧化酶(SOD)、酸性磷酸酶(ACP)活力的测定,结果表明,投喂D1饲料组的4个免疫指标均高于其它3组,虽然有时差异不显著.在14 dph的攻毒实验结果表明,对照组的累积死亡率显著高于D1组(P<0.05),而1D1-3D0和3D1-3D0的累积死亡率则与其它各组均不显著.实验结果表明,间隔投喂方式并不适合于对虾幼体,在中国明对虾幼体饲料中使用β-葡聚糖时,建议添加量为0.25%,并采用一直投喂的方式. 相似文献

Banana is one of the main economic agrotypes in Zhangzhou, Fujian Province. The multi-temporal ENVISAT ASAR data with different
polarization are used to classify the banana fields in this paper. Principal component analysis (PCA) was applied for six
pairs of ASAR dual-polarization data. For its large leaves, banana has high backscatter. So the value of banana fields is
high and shows very bright in the 1st component, which makes it much easier for banana fields extraction. Dual-polarization
data provide more information, and the VV and VH backscatter of banana show different characters with other land covers. Based
on the analysis of the radar signature of banana fields and other land covers and the 1st component, banana fields are classified
using object-oriented classifier. Compared to the field survey data and ASTER data, the accuracy of banana fields in the study
area is 83.5%. It shows that the principal component analysis provides the useful information in SAR images analysis and makes
the extraction of banana fields easier.
