日本鬼鲉不同组织的8 种同工酶谱

采用垂直聚丙烯酰胺平板电泳技术,对象山港野生日本鬼鲉(Inimicus japonicus)的9种组织器官的8种同工酶进行研究,并分析了其酶谱表型。结果表明,琥珀酸脱氢酶(Succinatedehydrogenase,简称SuDH)在所有组织中均存在,组织间差异较小;乳酸脱氢酶(Lactic dehydrogenase,简称LDH)、苹果酸脱氢酶((Malate dehydrogenase,简称MDH)、苹果酸酶(Malic enzyme,简称ME)、酯酶(Esterase,简称EST)、醇脱氢酶(Alcoholdehydrogenase,简称ADH)、过氧化物酶(Peroxidase,简称POD)、超氧化物歧化酶(Superoxide dismutase,简称SOD)则具有明显的组织特异性,且这些酶在表型、分布和活性上均表现出高度的组织特异性。     

雨生红球藻八氢番茄红素脱氢酶pds基因上游序列的分离和分析

在某些蓝藻、绿藻和高等植物中,八氢番茄红素脱氢酶是β-胡萝卜素合成过程中的一个关键酶之一,应用巢式PCR方法从雨生红球藻中克隆了八氢番茄红素脱氢酶基因约1kb的5’上游序列。通过生物信息学方法进行序列分析,发现pds基因上游序列中包含了一些可能的顺式元件,如ABRE调控元件、C-repeat/DRE调控元件等。同时,构建了由pds-启动子控制报告基因的重组载体,并转化到雨生红球藻细胞中,进行了报告基因瞬间表达的检测。结果表明,克隆的pds启动子区域具有启动子活性,可以驱使报告基因进行瞬间表达。     

一株海洋细菌铁吸收调节蛋白(Fur)基因的克隆和表达

对海洋细菌来说铁是一种必需元素,对铁吸收与利用的调控在细菌生长和防止铁毒性中起重要作用,在革兰氏阴性细菌中,铁的代谢由铁吸收调节蛋白(Fur) 来调控.橙色交替单胞菌是1种具有广泛抑菌谱的有益菌,本研究克隆表达了橙色交替单胞菌的fur基因,并分析了其相关特征.该基因序列中包含447bp的开放阅读框,编码148个氨基酸残基的蛋白,推导的相对分子量为16.637 kD,与GenBank报道的革兰氏阴性菌的相应序列同源性很高,其中氨基酸序列的中段从G47位至D104位,为高度保守区域.在限铁和富铁培养条件下铁载体分泌量的结果分析表明,fur基因可在大肠杆菌中大量表达,为进一步研究Fur蛋白的结构与功能打下基础.     

整合多源异构的城市规划数据进行认知交流的研究

针对整个城市规划过程的参与者对规划数据的不同种类、不同层次认知需求,本文利用本体思想对包括GIS数据、规划文档、规划图件等多源异构的规划数据进行关系重构,设计本体模型要素的数据语义关系,定义数据间的关联关系,实现基于本体方法的规划领域语义表达算法,方便了各类规划参与者对规划方案认识的交流:同时根据用户特征分类,进行本体关系的逻辑推理,过滤冗余信息,进行多源异构规划数据的可视化。     

基于交通轨迹数据的三维动态噪声地图

噪声信息具有空间传播和随距离衰减变化的特征,采用三维动态可视化形式表达噪声地图可以体现该特征。本文针对噪声的空间分布与强度变化提出一种噪声地图表达的方法,基于Bertin符号参量分析,以具有高度可变的方柱并配以深浅变化的颜色作为噪声符号,动态传递噪声信息。为进行多尺度地图展示,在不同地图等级下设有相应细节层次的符号尺寸和定位排列方式。噪声地图以Web端Mapbox为展示平台,基于交通噪声模型和GPS轨迹数据,对上海黄浦区某区域一时间段内的噪声情况进行模拟表达,实现噪声数据的立体动态化显示。三维动态的表达扩展了噪声数据的表现维度,多维度、多参量的立体动态符号不仅可准确刻画噪声的分布,而且从视觉观感上提高了受众的兴趣度和专注度,这为研究噪声的热点分布、传递趋势等空间规律提供了可视化参考。     

雄性半滑舌鳎雄激素受体基因的克隆与表达分析

利用RT-PCR方法从半滑舌鳎(Cynoglossus semilaevis)中克隆了雄激素受体(Androgen receptor,AR)基因cDNA的部分序列(434 bp),经BLAST比对与狼鲈(Dicentrarchus labrax)、花溪(Kryptolebias marmoratus)、三斑海猪鱼(Halichoeres trimaculatus)和三刺鱼(Gasterosteus aculeatus)的雄激素受体同源性分别为84%,82%,82%和81%.组织表达分析表明,AR基因在半滑舌鳎的性腺、肝、胃、脾、肾、头肾、肠、鳃、心、脑和肌肉这11种组织中均有表达,表达量有所差异.肾中表达量最丰富,鳃中最少.根据其他硬骨鱼类表达模式,推测半滑舌鳎可能只含有1种AR.

Matrix expression of thermal radiative characteristics for an open complex

The directionality of thermal radiance of a homogeneous isothermal non-black plane surface is totally decided by its directional emissivity, which depends on the complex dielectric constant and roughness of surface. It can be expressed by This paper proves that it is necessary to express emissivity by a matrix when a target becomes an inhomogeneous non-isothermal open complex with complicated inner geometric structure. The matrix describes the inner radiative interaction among components accurately and also expresses its thermal radiative directionality and structural characteristics completely. Advantages of matrix expression are as follows: first, the physical mechanics of effective emissivity of an open complex is described in a simple and perfect way; second, it becomes easy to understand the principle and method to retrieve components temperature from multi-angle thermal remotely sensed data; and third, the differences of directionalities between an open complex and a homogeneous isothermal non-black plane body are expressed by just using an effective emissivity matrix instead of an emissivity vector. Formula in classic physics is only the special case of matrix expression; therefore, the matrix is a universal unconditional expression to describe the directionality of thermal radiance.

Introduction Based on the elastic theory of hard inclusion model proposed by Dobrovolskii (1991), we developed a rheologic inclusion model to study the spatial-temporal variation of earthquake pre-cursor by using the bulk-strain field resulted from rheologic inclusion model (SONG et al, 2000). Sex expression and growth rates in natural populations of the desert soil crustal mossSyntrichia caninervis

The low elevation Mojave Desert cryptobiotic crust is dominated by the mossSyntrichia caninervis. In 16 populations of this moss, stem height and population were significantly associated with sex expression, with longer stems expressing sex more frequently and producing more perichaetia. The above-ground age of stems ranged up to 10 years, with a mean growth rate across populations of 0.36 mm year⁻¹. The overall stem sex ratio, as a proportion of the total number of stems, was 0.30♀: 0♂: 0.70 non-expressing. The probability of expressing sex in a given season was 0.09. The absence of sexual reproduction (no sporophytes) and likely absence of the male sex from the area indicate that the non-expressing plants are female individuals. The slow growth rates, low rates of sex expression, absence of male plants, and absence of sexual reproduction in this crustal species may help explain why re-establishment of mosses on desert soils can take decades.
