Differential expression of genes is crucial to growth and development of fish. To select the appropriate genes for gene normalization during Cynoglossus semilaevis early developmental process, eight candidate reference genes (ACTB, B2M, EF1A, GADPH, RPL7, TUBA, UBCE and 18S) were tested for their adequacy by using quantitative real-time PCR. The results showed that the expression of all the examined genes exhibited tissue dependent variations in the mature C. semilaevis. EF1A was listed as the most stable reference among the 14 tissues by RefFinder. Furthermore, the recommended comprehensive ranking of the stability determined by RefFinder showed that 18S was the most stable gene during the early developmental stages (from oosphere to 90 days old) in this study. However, when divided the Ct value data of the above mentioned early developmental stages into two separate periods (embryo and post-hatching periods), TUBA and 18S represented the most stable references of these two developmental periods, respectively. Consequently, the reference gene should be carefully and accurately chosen even for studies of the same species at various developmental processes. The relevant data may help in selecting appropriate reference genes for mRNA expression analysis, and is of great value in the studies of fish growth and development. 相似文献

为检测10个半滑舌鳎(Cynoglossus semilaevis)家系MHCⅡB(Cyse-DAB)基因的多态性水平、MHCⅡ类B位点数目以及平衡选择的作用,作者利用多聚酶链式反应(PCR)和直接测序的方法对10个半滑舌鳎家系MHCⅡB基因位点遗传变异和平衡选择进行了研究。用特异性引物和PCR扩增的半滑舌鳎MHCⅡB基因片段大约397 bp,包含一部分第一外显子,全部第一内含子和全部第二外显子。10个半滑舌鳎家系中,每家系选取5个体,每个体5个克隆序列分析发现60个不同序列,代表60个等位基因,其中有28个是新发现的,已提交到Gen Bank。同源分析表明60个等位基因相似性为89.36%。共50个个体中,有6个存在5个不同等位基因,表明在半滑舌鳎至少存在3个座位。有9个家系的MHCⅡB序列多肽结合区(PBR)的非同义替换(dN)显著高于同义替换(dS)。 相似文献

Stable carbon and nitrogen isotopes were used to assess site fidelity of Solea solea and Solea senegalensis juveniles, to investigate food web interactions and to determine the dominant nutrient pathways in two nursery areas in the Tagus estuary, Portugal. Samples of water from the main sources and from the nursery areas and respective saltmarsh creeks were collected for isotope analysis, as well as sediment, benthic microalgae, saltmarsh halophytes, S. solea, S. senegalensis and its main prey, Nereis diversicolor, Scrobicularia plana and Corophium spp. While site fidelity was high in 0-group juveniles, it was lower for 1-group juveniles, possibly due to an increase in mobility and energy demands with increasing size. Analysis of the food web revealed a complex net of relations. Particulate organic matter from the freshwater sources, from each nursery's waters and saltmarsh creeks presented similar isotopic composition. Sediment isotopic composition and saltmarsh halophytes also did not differentiate the two areas. All components of the food web from the benthic microalgae upwards were isotopically different between the nursery areas. These components were always more enriched in δ13C and δ15N at the lower nursery area than at the nursery located upstream, appearing as if there were two parallel trophic chains with little trophic interaction between each other. A mixture of carbon and nitrogen sources is probably being incorporated into the food web. The lower nursery area is more dependent upon an isotopically enriched energy pathway, composed of marine particulate organic matter, marine benthic microalgae and detritus of the C4 saltmarsh halophyte Spartina maritima. The two nursery areas present a different level of dependence upon the freshwater and marine energy pathways, due to hydrological features, which should be taken into account for S. solea and S. senegalensis fisheries and habitat management. 相似文献

Studies were conducted to determine the cause of the acute mortality of half-smooth tongue sole Cynoglossus semilaevis Günther juveniles in a fish farm in Jimo, Shandong Province, China, in June 2006. Gross signs of the diseased tongue sole included several petechiae and ecchymoses on the body and fin necrosis and hemorrhagic lesion at the base of the fin. Bacteria were isolated from kidney, liver and hemorrhagic lesions of the diseased tongue sole. Among 14 strains, SJ060621 was proved to be highly virulent to juvenile tongue sole with LD50 value of 〈1.0×10^5 colony forming units (CFU)mL^-1, while the remaining 13 were avirulent. Among the 16 antibiotics tested, SJ060621 was sensitive to gentamicin and nitrofurantoin. It was identified as Listonella anguillantm with conventional plate and tube tests in combination with API 20E analysis. 16S rRNA gene and partial HSP60 gene sequenceing analysis revealed that the strain was highly homologous with L. anguillarum. Examination of the infected musculature by electron microscopy indicated numerous bacteria and lots of macrophages containing phagocytosed bacteria. Histopathological investigations revealed severe necrotic degenerative changes in the infected organs. By comparison with the general features of metamorphic soles (e.g. vertical and lateral extension, metamorphic grade and diagnostic mineral parageneses, deformation and dominant rock types), it is inferred that the amphibolites, metagabbros and hornblendites of the Wadi Um Ghalaga–Wadi Haimur area in the southern part of the Eastern Desert of Egypt represent the metamorphic sole of the Wadi Haimur ophiolite belt. The overlying ultramafic rocks represent overthrusted mantle peridotite. Mineral compositions and thermobarometric studies indicate that the rocks of the metamorphic sole record metamorphic conditions typical of such an environment. The highest P – T conditions ( c . 700 °C and 6.5–8.5 kbar) are preserved in clinopyroxene amphibolites and garnet amphibolites from the top of the metamorphic sole, which is exposed in the southern part of the study area. The massive amphibolites and metagabbros further north (Wadi Haimur) represent the basal parts of the sole and show the lowest P – T conditions (450–620 °C and 4.7–7.8 kbar). The sole is the product of dynamothermal metamorphism associated with the tectonic displacement of ultramafic rocks. Heat was derived mainly from the hot overlying mantle peridotites, and an inverted P – T gradient was caused by dynamic shearing during ophiolite emplacement. Sm/Nd dating of whole-rock–metamorphic mineral pairs yields similar ages of c . 630 Ma for clinopyroxene and hornblende, which is interpreted as a lower age limit for ophiolite formation and an upper age limit for metamorphism. A younger Sm/Nd age for a garnet-bearing rock ( c . 590 Ma) is interpreted as reflecting a meaningful cooling age close to the metamorphic peak. Hornblende K/Ar ages in the range 570–550 Ma may reflect thermal events during late orogenic granite magmatism. 相似文献
