从山东近海海域沉积物的微生物筛选中获得了一株高产胞外黑色素的菌株QJNY82,通过对该菌株菌落形态特征观察、生理生化检验及16S rDNA序列测定,初步确定该菌株为发光杆菌属细菌.通过对菌株QJNY82的胞外产物进行红外光谱分析,得到该细菌的胞外产物为黑色素,纯化后的黑色素的红外光谱图与标准黑色素(SIGMA)的红外光谱图一致,且该细菌所产黑色素的理化性质与标准黑色素具有一致性.该海洋菌株具有产黑色素速度快,产量高且不需要L-酪氨酸诱导的特点,而且经L-酪氨酸诱导后,可显著提高菌株QJNY82胞外黑色素的产量,在L-酪氨酸的含量为0.75 g/dm3、盐度为30、pH值为8.0、温度为28℃的条件下,菌株黑色素产量是未加L-酪氨酸的3.5倍,其产量达1.325g/dm3.鉴于菌株QJNY82具有产黑色素的特性,将成为发光杆菌属的一个新的菌种资源. 相似文献

Characterization of calcium deposition induced by Synechocystis sp. PCC6803 in BG11 culture medium 总被引:1,自引:0,他引:1
Calcium carbonate (CaCO3) crystals in their preferred orientation were obtained in BG11 culture media inoculated with Synechocystis sp. PCC6803 (inoculated BG11). In this study, the features of calcium carbonate deposition were investigated. Inoculated BGll in different calcium ion concentrations was used for the experimental group, while the BGll culture medium was used for the control group. The surface morphologies of the calcium carbonate deposits in the experimental and control groups were determined by scanning and transmission electron microscopy. The deposits were analyzed by electronic probe micro-analysis, Fourier transform infrared spectrum, X-ray diffraction, thermal gravimetric analysis and differential scanning calorimetry. The results show that the surfaces of the crystals in the experimental group were hexahedral in a scaly pattern. The particle sizes were micrometer-sized and larger than those in the control group. The deposits of the control group contained calcium (Ca), carbon (C), oxygen (O), phosphorus (P), iron (Fe), copper (Cu), zinc (Zn), and other elements. The deposits in the experimental group contained Ca, C, and O only. The deposits of both groups contained calcite. The thermal decomposition temperature of the deposits in the control group was lower than those in the experimental group. It showed that the CaCO3 deposits of the experimental group had higher thermal stability than those of the control group. This may be due to the secondary metabolites produced by the algae cells, which affect the carbonate crystal structure and result in a close-packed structure. The algae cells that remained after thermal weight loss were heavier in higher calcium concentrations in BGll culture media. There may be more calcium- containing crystals inside and outside of these cells. These results shall be beneficial for understanding the formation mechanism of carbonate minerals. 相似文献

A marine bacterium,Pseudoalteromonas sp.BC228 was supplemented to feed in a feeding experiment aiming to determine its ability of enhancing the digestive enzyme activity and immune response of juvenile Apostichopusjaponicus.Sea cucumber individuals were fed with the diets containing 0 (control),105,107 and 109 CFU g-1 diet of BC228 for 45 days.Results showed that intestinal trypsin and lipase activities were significantly enhanced by 107 and 109 CFU g-1 diet of BC228 in comparison with control (P〈0.01).The phagocytic activity in the coelomocytes of sea cucumber fed the diet supplemented with 107 CFUg-1 diet of BC228 was significantly higher than that of those fed control diet (P〈0.05).In addition,105 and 107 CFUg-1 diet of BC228 significantly enhanced lysozyme and phenoloxidase activities in the coelomic fluid of sea cucumber,respectively,in comparison with other diets (P 〈 0.01).Sea cucumbers,10 each diet,were challenged with Vibrio splendidus NB 13 after 45 days of feeding.It was found that the cumulative incidence and mortality of sea cucumber fed with BC228 containing diets were lower than those of animals fed control diet.Our findings evidenced that BC228 supplemented in diets improved the digestive enzyme activity of juvenile sea cucumber,stimulated its immune response and enhanced its resistance to the infection of V.splendidus. 相似文献

本研究预测naa cluster中的naaB基因负责催化烟酸到6-羟基烟酸的反应。通过对NaaB的蛋白序列进行分析,发现该酶含有三个亚基,分别由naaBL1、naaBS、naaBL2基因编码,其中naaBL1和naaBL2分别编码2个含有钼辅因子结合结构的大亚基,naaBS基因编码一个含有[2Fe-2S]簇的小亚基。本研究对naaB基因进行了克隆,将构建的重组质粒pME6032-naaBL1SL2分别转化到无色杆菌(Achromobacter sp.)和大肠杆菌(Escherichia coli)中,通过HPLC和LC-MS检测验证了NaaB负责催化烟酸到6-羟基烟酸,并且在无色杆菌和大肠杆菌中NaaB均能够转化烟酸,这与之前报道烟酸羟化酶不能在大肠杆菌表达的结果不同,说明NaaB的基因编码和催化功能具有独特性。通过序列分析推测NaaB的这一特性可能跟该酶同时含有两个钼结合结构域的大亚基有关。 相似文献

Thorium(IV) biosorption is investigated by citric acid treated mangrove endophytic fungus Fussarium sp. #ZZF51 (CA-ZZF51) from South China Sea. The biosorption process was optimized at pH 4.5, equilibrium time 90 min, initial thorium(IV) concentration 50 mg L−1 and adsorbent dose 0.6 g L−1 with 90.87% of removal efficiency and 75.47 mg g−1 of adsorption capacity, which is obviously greater than that (11.35 mg g−1) of the untreated fungus Fussarium sp. #ZZF51 for thorium(IV) biosorption under the condition of optimization. The experimental data are analyzed by using isotherm and kinetic models. Kinetic data follow the pseudo-second-order model and equilibrium data agree very well with the Langmuir model. In addition, FTIR analysis indicates that hydroxyl, amino, and carbonyl groups act as the important roles in the adsorption process. 相似文献

PAH-degrading bacteria, including Novosphingobium sp. PCY, Microbacterium sp. BPW, Ralstonia sp. BPH, Alcaligenes sp. SSK1B, and Achromobacter sp. SSK4, were isolated from mangrove sediments. These isolates degraded 50–76% of 100 mg/l phenanthrene within 2 weeks. Strains PCY and BPW also degraded pyrene at 98% and 71%, respectively. Furthermore, all of them probably produced biosurfactants in the presence of hydrocarbons. Interestingly, PCY has a versatility to degrade various PAHs. Molecular techniques and plasmid curing remarkably revealed the presence of the alpha subunit of pyrene dioxygenase gene (nidA), involving in its pyrene/phenanthrene degrading ability, located on megaplasmid of PCY which has never before been reported in sphingomonads. Moreover, genes encoding ferredoxin, reductase, extradiol dioxygenase (bphA3A4C) and exopolysaccharide biosynthetase, which may be involved in PAH degradation and biosurfactant production, were also found in PCY. In this study,the ability of macroalgae Gracilaria sp.of removing eutrophication factors and toxic heavy metals Al,Cr,and Zn in a closed cultivation system is reported.The results show that the concentration of the three heavy metals decreased significantly during the experimental period in an algal biomass dependent manner.The biofiltration capacity of the alga for Al,Cr,and Zn is 10.1%-72.6%,52.5%-83.4% and 36.5%-91.7%,respectively.Using more materials resulted in stronger heavy metal removal.Additionally... 相似文献
