Expressed sequence tag analysis and cloning of trehalose-6-phosphate synthase gene from marine alga Laminaria japonica (Phaeophyta)

A high quality cDNA library was constructed from the brown alga Laminaria japonica,with the titer of 1.2×10 5 pfu/ml.The average insert size of the cDNA library is about 1.6 kb.From the cDNA library,591 cDNA clones were randomly selected and sequenced.As a result,574 EST(expressed sequence tag) sequences were generated.All of the 574 ESTs were submitted to the dbEST database section of GenBank with the accession numbers from CX942625 to CX943198.The cDNA library was screened with a α-32 p labeled 453 bp T P S gene probe,which is a partial sequence yielded from Porphyra yezoensis.Four positive cDNA clones were screened and the sequencing data showed that these four cDNA clones covered majority of L.japonica TPS cDNA sequence.After PCR amplification,sequencing and assembling,the entire ORF(open reading frame) sequence of the T P S gene was obtained,which was named LjTPS.LjTPS encodes a protein containing 908 amino acids with a calculated molecular mass of 101 674 Daltons.The LjTPS gene was successfully expressed in E.coli and rice.The LjTPS gene has potential application both in plant breeding to stress tolerance and in deciphering the T P S gene function and mechanism to stress tolerance.     

Construction of cDNA library from intestine, mesentery and coelomocyte of Apostichopus japonicus Selenka infected with Vibrio sp. and a preliminary analysis of immunity-related genes

The aquaculture of sea cucumber Apostichopus japonicus (Echinodermata, Holothuroidea) has grown rapidly during recent years and has become an important sector of the marine industry in Northern China. However, with the rapid growth of the industry and the use of non-standard culture techniques, epidemic diseases of A. japonicus now pose increasing problems to the industry. To screen the genes with stress response to bacterial infection in sea cucumber at a genome wide level, we constructed a cDNA library from A. japonicus Selenka (Aspidochirotida: Stichopodidae) after infecting them with Vibrio sp. for 48 h. Total RNA was extracted from the intestine, mesentery and coelomocyte of infected sea cucumber using Trizol and mRNA was isolated by Oligotex mRNA Kits. The ligated cDNAs were transformed into DH5α, and a library of 3.24×105 clones (3.24×105 cfu mL-1) was obtained with the sizes of inserted fragments ranging from 0.8 to 2.5 kb. Sequencing the cDNA clones resulted in a total of 1106 ESTs that passed the quality control. BlastX and BlastN searches have identified 168 (31.5%) ESTs sharing significant homology with known sequences in NCBI protein or nucleotide databases. Among a panel of 25 putative immunity-related genes, serum lectin isoform, complement component 3, complement component 3-like genes were further studied by real-time PCR and they all increased more than 5 fold in response to Vibrio sp. challenge. Our library provides a valuable molecular tool for future study of invertebrate immunity against bacterial infection and our gene expression data indicates the importance of the immune system in the evolution and development of sea cucumber.     

细角螺微卫星DNA 富集文库构建及特征分析

采用生物素磁珠富集法,用生物素标记的(GT)15和(CT)15两种探针与细角螺基因组 MseI 酶切的300~1200bp 片段杂交,杂交复合物与链霉亲和素磁珠结合,捕获含有重复序列的微卫星片段。最后将磁珠捕获到的重复序列与PMD19-T载体连接后克隆到DH5α中构建微卫星基因组文库。通过PCR检测出354个阳性克隆,从中随机选取248个片段大于500bp的阳性克隆进行测序,结果显示,在220个成功测序的阳性克隆中共获得278个微卫星序列,其中完美型171个,占61.5%;非完美型81个,占29.1%;复合型26个,占9.4%。除探针使用的GT和CT的重复序列外,还筛选到三碱基GTT、TGG、GAA、CAA;四碱基TCTA、ACAG、TAGA、GTGA、GTCT、GATA及五碱基TTTTG的重复序列。在278条序列中共有82条可以设计引物。     

基于GIS模式的数据仓专题图研究

针对基于GIS的BTEX油气化探数据仓地质专题图制作系统进行研究,分析了基于自动化制图理念的数据流,提出了系统的总体构架及基于国家标准的地质符号库,并根据油田地质成果图规范开发了BTEX油气化探数据仓数据挖掘成果图的制图模板。将该系统应用于渤中凹陷的BTEX数据综合评价图的制作,结果表明该软件具有较好的人机交互性和扩展性,在满足BTEX油气化探数据仓的处理结果专题地图的制作与成果展示的基础上,能够提高BTEX油气化探地质专题图的制图效率。     

SVG WebGIS中地图符号交互配置方法的探讨

采用SVG技术实现WebGIS具有许多独特的优势。从推广SVG WebGIS应用、促进其商用化的角度出发,本文研究了SVG地图符号交互配置方法的设计和实现:它具有较好的实用性、通用性、可移植性和可扩展性,以及较高的地图符号化效率等优点。重点解决了SVG地图点、线、面符号的设计和交互配置等问题。     

分布式遥感模型库的构建及其运行机制

波谱特性是地物目标的基本特性之一,是定性、定量遥感的基础。我国新一代地物波谱库建设以目标地物结构、波谱、知识库、模型库的相互配套为特色,旨在解决以前地物波谱库建设和使用中存在的一些问题,使地物波谱库在空间信息产业化过程中发挥更大的作用。论述了遥感模型库在新一代波谱数据库中的地位和作用,并就遥感模型库的组成、网络环境下的体系结构与开发技术,以及波谱知识库和元数据支持下的遥感模型库运行机制进行了探讨。提出基于网络的三层分布式遥感模型库体系结构和基于组件的模型库构建方法。同时还根据遥感模型库建设的具体情况,讨论了遥感模型方法元数据,并提出基于元数据的遥感模型库管理和运行机制。上述设计思想保证了遥感模型库具有动态的扩充机制和较强的代码可重用性,使模型库能够得到及时更新和扩充,从而保持库的先进性与实用性。     

地理信息系统中一种全面的符号管理方案

符号和符号的管理是地形图制图的数据可视化的核心内容，在地理信息系统中占有重要的地位，文中提出一个新的符号管理方案，通过对符号表和符号库的双重管理，解决了符号迁移问题，同时也使得GIS中一些常用功能的实现变的直接，高效，最后还就该方案同现有一些GIS系统的符号管理方案作了比较分析，进一步说明了它的全面性和实用性。     

ASP组件WEBGIS系统中ASP函数库的设计与实现

在WEGBIS研究日趋白热化的今天，笔者所在的研究小组提出了一种基于ASP组件技术的WEBGIS解决方案，并以MAPGIS为平台，开发研制出了一组功能强大的组件。然而让二次开发用户学习、掌握这些纷杂的组件，比较困难。为减轻二次开发用户对该组件库的学习难度，实现快速、简洁地建站，对有规律的ASP代码进行了封装，形成一定规模的ASP函数库，从而降低了学习组件的难度，加快了建站的速度，促进了WEBGIS的推广。     

基于关系数据库的三维模型库技术

提出了一个利用关系数据库来实现三维模型库的方法,对数据库设计、模型数据的建模要求、客户端实现以及模型库的维护做了说明,解决了文件方式管理三维模型数据的弊端。
