2018年4月在浙江省舟山近海海域的渔业资源调查中采集到19尾菖鲉属鱼类标本,经鉴定发现为三色菖鲉(Sebastiscus tertius),为中国大陆近海海域的新记录种。本研究对采集到的19尾三色菖鲉标本拍摄照片并对其进行形态特征以及DNA条形码研究。三色菖鲉具有以下主要形态特征:背鳍数ⅩⅡ-12,胸鳍数18—19,腹鳍数Ⅰ-5,臀鳍数Ⅲ-5;第一鳃弓总鳃耙数6—7+13—17;前鳃盖骨有5枚硬棘,鳃盖骨后沿有2枚硬棘;眶前骨下后角有1枚硬棘;胸鳍处有一明显黑斑。测定了19尾标本的12S rRNA和COI序列,结合GenBank中所有学名为Sebastiscus tertius的同源序列进行分析,发现12S rRNA序列完全一致,可作为该物种的微型DNA条形码,而COI序列中所有个体明显分为两个分支,两者遗传距离达到0.044,表明两分支可能为不同的有效种。COI同源序列分析结果显示, GenBank中学名为Sebastiscus tertius的鱼种与褐菖鲉(Sebastiscus marmoratus)聚为一支,表明GenBank中的Sebastiscus tertius存在错误...  相似文献

宏条形码分析(metabarcoding analysis)是通过对野外样本的分子标记(即条形码)进行PCR扩增、测序,并利用测序结果分析野外样本物种组成、物种相对丰度,及其时空动态变化。这项在近20年里逐步发展起来的分子技术,得益于DNA测序技术的快速发展,包括测序通量的提升和测序成本的降低以及生物信息学分析方法的快速开发。基于可操作分类单元(operational taxonomic units,OTUs)分析方法,特别是最近的扩增子序列变异(amplicon sequence variants,ASVs)方法的开发,极大促进了宏条形码分析的应用。在世界及我国的海洋生态领域,宏条形码分析的应用还处于起步阶段。随着分析方法的日趋成熟,可适用于不同应用领域的分子标记不断增加,以及参考数据库的不断完善,宏条形码分析将释放出广阔的应用潜力。可以预期,宏条形码分析将在系统分析我国近岸海域有害藻华物种(及其他浮游生物物种)的组成和相对丰度中起关键作用,通过发现新物种,辨别隐存种,跟踪物种的生物地理学,进而帮助解析有害藻华暴发机理。  相似文献

为了解海南尖鳍鲤(Cyprinus acutidorsalis)线粒体基因组结构特征及其与鲤科(Cyprinidae)鱼类的进化关系,本文通过二代测序获得海南万泉河口尖鳍鲤线粒体基因组全序列,并对结构特征进行分析,结果显示具有典型的环状闭环双链分子,全长16581bp,碱基组成为A(31.96%)、G(15.69%)、C(27.53%)和T(24.82%),包含13个蛋白质编码基因(protein-codinggenes,PCGs)、2个r RNA基因、22个tRNA基因和1个可变控制区D环。对碱基含量分析发现尖鳍鲤的碱基组成中具有较高AT含量的偏向性,13个PCGs中有不少的偏好性密码子,如CGA(RSCU>3.506)、CCA(RSCU>2.208)等。除tRNAGln、tRNAAla、tRNAAsn、tRNACys、tRNALeu(CUN)、tRNATyr、tRNASer(UCN)、tRNAPro  相似文献

藻胆蛋白(Phycobiliprotein,PBP)是原核藻类和真核藻类进行光合作用的捕光色素蛋白,占藻体干重的2%左右,主要分为3类:藻红蛋白(phycoer-ythrin,PE)、藻蓝蛋白(phycocyanin,PC)和别藻蓝蛋白(allophycocyanin,APC)[1].藻胆蛋白一般由α亚基和β亚基组成,部分藻胆蛋白中还存在γ亚基.一般由α和β亚基形成稳定的单体(αβ),再由单体聚合为多聚体(αβ)n.从蓝藻和红藻中分离的藻胆蛋白为三聚体(αβ)3或六聚体(αβ)6[2].  相似文献

Brackish water ecosystems are often exposed to wide variations in environmental variables, including temperature and salinity, which may cause strong selective pressures on organisms modifying the genetic patterns of species. The aim of this work was to test whether there is a ‘divergence-with-gene flow’ in coastal lagoon populations of white seabream (Diplodus sargus) (Ria Formosa, S Portugal and Mar Menor, SE Spain) respect to four marine populations, by using partial sequences of cyt b mitochondrial gene and information from nine microsatellite loci. Genetic diversity was highest in both coastal lagoons (Mar Menor and Ria Formosa) considering mitochondrial and nuclear markers. Although some of FST population pairwise comparisons were not significant, analyses of molecular variance (AMOVAs) detected differences between groups (coastal lagoon and marine) close to significance. Also, only two haplotypes (Cytb-17 and Cytb-18) were detected in both coastal lagoon sampling sites and these localities (Mar Menor and Ria Formosa) showed the highest number of singletons, some of them with a high number of mutations, as has been already described for other Mar Menor populations (Pomatochistus marmoratus and Holothuria polii). Also, several tests detected significant positive and balancing selection considering mtDNA and microsatellite data. These data support the hypothesis of selection as one of the drivers of the genetic differences found between coastal lagoon and marine populations. The life strategy adopted by Diplodus sargus in coastal lagoons allows it to decrease its mortality rate and improve the heritability of its genes. Also, the increase time spent in coastal lagoons with different temperatures and salinities favours the fitness selection and the maintenance of exclusive haplotypes and genotypes in coastal lagoon inhabitants favouring the ‘divergence-with-gene-flow’.  相似文献

Ulva species can grow rapidly in nutrient-rich habitats causing green tides and marine fouling. A more complete understanding of the reasons behind these outbreaks is urgently required. Accordingly, this study attempts to use microsatellite markers based expressed sequence tag(EST) to analyze the genetic variation of several Ulva prolifera populations in the South Yellow Sea of China. Two hundred and thirty-eight SSRs were identified from 8 179 unique ESTs(6 203 newly sequenced and 1 976 downloaded from NCBI database) and 37 primer pairs were successfully designed according to the ESTs; 11 pairs were selected to detect the genetic diversity and relationship of 69 attached U. prolifera samples and 13 free-floating samples collected from coastal and off-coast areas of the South Yellow Sea. The results of cross-species transferability showed that six of the 11 EST-SSR primers could give good amplification in other five Ulva species and the average allele number was 4.67. Genetic variation analysis indicated that all 82 U. prolifera samples were clearly divided and most samples collected from the same site clustered together as a group in the dendrogram tree produced by unweighted pair-group mean analysis(UPGMA) method and the cluster results showed some consistency with the geographical origins. In addition, 13 free-floating samples(except HT-001-2) were grouped as a single clade separated from the attached samples.  相似文献

采用钻/测井、地震及米兰科维奇旋回相结合的方法,对柴达木盆地三湖坳陷第四系七个泉组进行了高分辨率层序地层学研究,在七个泉组可识别出6个层序界面,进而划分出5个三级层序。根据蒿藜花粉含量、碳酸盐含量、δ18O、GR、氯离子含量等对比分析,本区气候总体上由温暖变为干冷,存在5次温湿与干冷气候的次级旋回,分别对应sq1、sq2、sq3、sq4、sq5五个层序,每个旋回内部波动与基准面上升半旋回和基准面下降半旋回相一致。层序Ⅱ(sq2)基准面上升半旋回时期发育的三角洲前缘砂体尖灭及砂岩透镜体圈闭是进一步勘探的有利目标。  相似文献

通过对辽东湾高分辨率浅地层剖面声学地层与典型钻孔沉积地层的对比分析,揭示了研究区晚第四纪MIS5以来的地层层序。辽东湾高分辨率浅地层剖面自下而上划定的6个声学地层单元(SU5、SU4、SU3、SU2、SU12、SU11)与钻孔岩芯划分的6个沉积地层单元(DU5、DU4、DU3、DU2、DU12、DU11)具有良好的对应关系。分别与MIS4期、MIS2期低海面时期的沉积间断密切相关的两个层序界面R5、R3,将辽东湾识别出的地层单元自下而上划分为3个层序(SQ3、SQ2、SQ1)。其中SQ3仅识别出上部的海侵体系域与高水位体系域,对应MIS5期海平面相对较高时期的滨浅海相沉积(DU5);SQ2自下而上由低水位体系域(MIS...  相似文献

Connectivity between estuarine fish nurseries and coastal adult habitats can be affected by variations in juvenile growth and survival. Condition indices are renowned proxies of juvenile nutritional status and growth rates and are valuable tools to assess habitat quality. Biochemical (RNA:DNA ratio) and morphometric (Fulton's condition factor K) condition indices were determined in juveniles of Solea solea, Solea senegalensis, Platichthys flesus, Diplodus vulgaris and Dicentrarchus labrax collected in putative nursery areas of nine estuaries along the Portuguese coast (Minho, Douro, Ria de Aveiro, Mondego, Tejo, Sado, Mira, Ria Formosa and Guadiana) in the Spring and Summer of two consecutive years (2005 and 2006) with distinct climatic characteristics. Individual condition showed significant variation amongst species. The combined use of both condition indices highlighted the low correlation between them and that RNA:DNA had a higher sensitivity. RNA:DNA varied between years but overall the site relative patterns in condition were maintained from one year to the other. Higher RNA:DNA values were found in Spring than in Summer in most species. Intra-estuarine variation also occurred in several cases. Species specific trends in the variability of condition amongst estuaries were highlighted. Some estuaries had higher juvenile condition for more than one species but results did not reveal an identical trend for all species and sites, hindering the hypotheses of one estuarine nursery promoting superior growth for all present species. Significant correlations were found between condition indices, juvenile densities and environmental variables (water temperature, salinity and depth) in the estuarine nurseries. These influenced juvenile nutritional condition and growth, contributing to the variability in estuarine nursery habitat quality. Management and conservation wise, interest in multi-species approaches is reinforced as assessments based on a single species may not reflect the overall nursery habitat quality.  相似文献
