Three years of regular weekly/biweekly monitoring of seasonal changes in temperature, transparency, chlorophyll a (CHL) and bacteria [erythrosine-stained microscopic counts and cultivable colony forming units (CFUs)] at the vertical profile in the South basin of Lake Baikal (51°54′195″N, 105°04′235″E, depth 800 m) were evaluated. In more detail, the structure and function of phytoplankton and the microbial loop in the euphotic layer at the same site were investigated during the late-winter–early-spring period under the ice. The depth of euphotic zone (up to 1% of surface irradiation) was 35 to 40 m. Primary production was measured three times a week with the 14C method in 2, 10, 20, 30 and 40 m. Maximum production was found in 10 m, with lower values towards the surface (light inhibition) and towards the lower layers. The total production in cells larger than 1 μm in the column (0–40 m) was 204–240 mg C d−1 m−2, 30–40% of it being in cells 1–3 μm (mostly picocyanobacteria), which represented roughly 9% of the total chlorophyll a (estimated from pigment analyses). A major part of phytoplankton biomass was formed by diatoms (Synedra acus Hust., Asterionella formosa Hass. and Stephanodiscus meyerii Genkal & Popovskaya). Total production (including extracellular, dissolved organic matter) was 235–387 mg C day−1 m−2, and the exudates were readily used by bacteria (particles 0.2–1 μm). This part amounted to 1–5% of cellular production in 2 to 20 m and 11–77% of cellular production in 20–40 m, i.e., in light-limited layers. From 0 to 30 m, chlorophyll a concentration was 0.8 to 1.3 μg l−1, wherefrom it decreased rapidly to 0.1 μg l−1 towards the depth of 40 m. Bacteria (DAPI-stained microscopic counts) reached 0.5–1.4×106 ml−1; their cell volumes measured via image analysis were small (average 0.05 μm−3), often not well countable when erythrosine stain was used. Bacterial biomasses were in the range of 6–21 μg C l−1. Numbers of colony forming units (CFUs) on nutrient fish-agar were c. 3–4 orders lower than DAPI counts. The amounts of heterotrophic protists were low, whereby flagellates reached 6 to 87 ml−1 and ciliates, 0.2–1.2 ml−1 (mostly Oligotrichida). Bacterial production was measured in the same depths as primary production using 3H-thymidine (Thy) and 14C-leucine (Leu) uptake. Consistently, bacterial abundances, biomasses, thymidine and leucine production were higher by 30–50% in layers 2, 10 and 20 m compared with that in the deeper 30 and 40 m, where cellular primary production was negligible. Leucine uptake in the deeper layers was even three times lower than in the upper ones. From the comparison of primary and bacterial production, bacteria roughly use 20–40% of primary production during 24 h in the layers 2 to 20 m.  相似文献

积雪是地表特征的重要参数,对辐射收支、气候和长期天气变化均有重要影响。雪本身又是一个重要的天气现象和水文气象参数,过量的降雪也会带来严重的雪灾,如牧区雪灾、雪崩和融雪洪水灾害等。因此对积雪的监测,尤其是对山区的积雪监测,具有多方面的意义。利用卫星遥感技术监测积雪已有50余年的历史,并已形成了系列业务产品。青藏高原平均海拔超过4 000 m,该地区的积雪具有重要的水文、气候和生态环境意义。由于地形复杂,人迹罕至,地面观测站点稀少,受较强太阳辐射的影响,积雪消融迅速、区域差异消融以及风吹雪等因素导致积雪分布破碎化严重,对使用遥感资料监测该地区的积雪造成的极大的困难和不确定性。随着国内外传感器技术的不断发展,光学和被动微波遥感数据的同步获取技术已经非常成熟,综合利用光学遥感数据高空间分辨率和被动微波数据不受云干扰的特点,结合机器学习、无人机等技术,将环境参数加入反演模型中,有助于提高青藏高原积雪参数反演精度。  相似文献

 Bacterial interaction with oil and sea water, in a porous medium, was studied using a transparent micro model mounted on a cybernetic-controlled positioning board of an inverted microscope. This was interfaced with computer-operated image analysis and surveillance equipment. Consecutive flooding of the model allowed oil to become trapped by capillary forces. Hydrocarbonoclastic bacteria were inoculated. The interaction of biofilm and oil was monitored during a period of 25 days, while mineral salts were supplied intermittently. A unique feature of biofilm behaviour was discovered. The phenomenon is discussed, and calculations are presented. A possible explanation of the phenomenon appears to be local reductions of the surface tension. The phenomenon helps explain why bacteria are able to mobilise oil trapped by capillary forces without reducing the interfacial tension to a level that leads to an increase in the capillary number sufficient to explain incremental oil recovery. Subterranean microorganisms and radioactive waste disposal in Sweden   总被引:2,自引:0,他引:2  
In 1987, microbiology became a part of the Swedish scientific program for the safe disposal of high level nuclear waste (HLW). The goal of the microbiology program is to understand how subterranean microorganisms will interact with the performance of a future HLW repository. The Swedish research program on subterranean microbiology has mainly been performed at two sites in granitic rock aquifers at depths ranging from 70 m down to 1240 m, the Stripa research mine in the middle of Sweden and the Äspö Hard Rock Laboratory (HRL) situated on the south eastern coast of Sweden. Some work has also been performed in cooperation with other national or international research groups in Sweden, Canada and at the natural analogue sites Oklo in Gabon and Maqarin in Jordan. The following conclusions are drawn. There is a very high probability of the existence of a deep subterranean biosphere in granitic rock. The documented presence of a deep biosphere implies that relevant microbial reactions should be included in the performance assessment for a HLW repository. A HLW repository will be situated in a subterranean biosphere that is independent of solar energy and photosynthetically produced organic carbon. The ultimate limitation for an active microbial life will be the availability of hydrogen as energy source over time, and hydrogen has indeed been found in most deep groundwaters. Sulphide producing microorganisms are active in environments typical for a Swedish HLW repository, and the potential for microbial corrosion of the copper canisters must be considered. The bentonite buffer around the copper canisters will be a hostile environment for most microbes due to the combination of radiation, heat and low water availability. Discrete microbial species can cope with each of these constraints, and it is theoretically possible that sulphide producing microbes may be active inside a buffer, although the experiments conducted thus far have shown the opposite. Microorganisms have the capability to enzymatically recombine radiolysis oxidants formed by radiation of water. It has earlier been concluded that the migration of radionuclides due to sorption on microorganisms can be neglected. The influence of microbially produced complexing agents remains to be studied at realistic conditions in deep groundwater. Microorganisms have been found in natural alkaline groundwaters, but it could not be conclusively demonstrated that they were in situ viable and growing, rather than just transported there from neutral groundwater. A possible hypothesis based on the obtained results from investigations of natural alkaline groundwaters is that fresh concrete may be a bit too extreme for active life even for the most adaptable microbe – but this remains to be demonstrated.  相似文献

Water quality in Singapore's coastal area was evaluated with microbial indicators, pathogenic vibrios, chemical tracers and physico-chemical parameters. Sampling sites were grouped into two clusters (coastal sites at (i) northern and (ii) southern part of Singapore). The coastal sites located at northern part of Singapore along the Johor Straits exhibited greater pollution. Principal component analysis revealed that sampling sites at Johor Straits have greater loading on carbamazepine, while turbidity poses greater influence on sampling sites at Singapore Straits. Detection of pathogenic vibrios was also more prominent at Johor Straits than the Singapore Straits. This study examined the spatial variations in Singapore's coastal water quality and provided the baseline information for health risk assessment and future pollution management.  相似文献

Meteorological extreme events (heavy rainfall, heat waves) may lead to fast changes in nutrient load and water temperature in temperate lakes. We conducted laboratory experiments with an artificial phytoplankton community to mimic a rapid temperature increase (from 21 °C to 29 °C) at low nutrient levels (‘heat wave scenario’), respectively temperature decrease (from 21 °C to 16 °C) and increased nutrient load (‘heavy rainfall scenario’). We hypothesised that there is a taxon specific nutrient x temperature interaction, leading to significant shifts in the phytoplankton community composition when both variables change. To separate the temperature effect from the nutrient effect, we performed another experimental series at a reduced temperature but without addition of nutrients. As expected, the nutrient effect was overall more important than temperature and significantly affected all five taxa tested that represented different algal classes. However, temperature also played an important role for community composition, because the cryptophyte Cryptomonas sp. and the dinoflagellate Peridinium sp. reached significantly higher biovolumes at lower temperatures. The nutrient x temperature interaction was significant in the green alga Scenedesmus obliquus. These findings suggest that our experimental results cannot be interpreted primarily by species competition for nutrients. Heterotrophic bacteria were present in all experiments. Bacterial biomass was significantly positively related to temperature and nutrients. However, relative to phytoplankton biovolume, bacterial biovolume decreased under nutrient replete conditions. In conclusion, our results demonstrate that short-term environmental change may significantly affect both the phytoplankton community (in terms of species dominance and total biomass) and the ratio between autotrophs and heterotrophs in temperate lakes.  相似文献

The Mawsmai cave and Krem Phyllut caves, East Khasi hills, Meghalaya, India has so far not yet attracted the attention of geomicrobiologists. Observations and hypotheses on the possible influence of identified microorganisms for speleothem formations in Meghalaya are reported for the first time. XRD studies identified calcite in speleothems and gypsum in cave wall deposits as the dominant minerals. SEM-EDAX showed interesting microfabric features showing strong resemblance with fossilised bacteria, calcified filaments, needle calcite and numerous nano scale calcite crystals, highly weathered and disintegrated crystals of calcite, that point towards a significant microbial influence in its genesis. Thin section petrography showed laminated stromatolitic features. The microorganisms identified by conventional isolation and further evaluation of isolates by molecular techniques include Bacillus cereus, Bacillus mycoides, Bacillus licheniformis, Micrococcus luteus, and Actinomycetes. Microscopic observations also showed unidentifiable cocci and four unidentifiable strains of CaSO4 (gypsum) precipitating bacteria. Experimental studies confirmed that these bacteria are able to precipitate calcium minerals (calcite, gypsum, minor amounts of dolomite) in the laboratory. These results allow us to postulate that species like these may contribute to active biogenic influence in the cave formations at Meghalaya.  相似文献

通过已构建的泥蚶血液cDNA文库,克隆得到泥蚶Tg-HbIIA基因全长cDNA序列,对其进行生物信息学、组织表达及免疫相关分析。结果表明,泥蚶Tg-HbIIA cDNA全长731bp,包含63bp5′非编码区(5′-UTR),246bp3′非编码区(3′-UTR),开放阅读框450bp,编码150个氨基酸;其氨基酸序列与毛蚶(Scapharca kagoshimensis)和不等壳毛蚶(Scapharca inaequivalvis)的序列有较高的同源性,分别达到89%和88%;对其结构预测显示该蛋白具有血红蛋白功能域,含有10个潜在血红素结合位点。qRT-PCR检测结果显示:泥蚶不同组织部位中,Tg-HbIIA mRNA在血液表达量最大,其次为外套膜和鳃,表达量最低的是肝胰脏;在副溶血弧菌(Vibrio parahaemolyticu)、脂多糖、肽聚糖刺激后,泥蚶血液Tg-HbIIA mRNA表达量显著上调,表明Tg-HbIIA在贝类抗菌免疫防御中可能起重要作用,而Tg-HbIIA对不同致病因子免疫反应的灵敏度和表达时序存在一定差异。  相似文献
