We have applied the technique of random amplification of polymorphic DNA (RAPD) to the analysis of the relationships among four species of brine shrimp: Artemia franciscana, A. urmiana, A. sinica, and A. parthenogenetica. Seventy ten-base synthetic oligonucleotides were used to amplify a total of 458 distinct fragments. DNA polymorphisms were found in all the species examined; the highest percentage of polymorphic bands was found in A. parthenogenetica, with 28.8 per cent. Each species was scored for the presence or absence of every amplification product and the data entered into a binary data matrix. Cluster analysis was then performed to create a dendrogram using UPGMA by the NTSYS program. There are significant differences between bisexual species and parthenogenetic populations. A. parthenogenetica provided 94 specific molecular markers, while bisexual species gave 27 specific molecular markers. A. sinica is a species distinct from the other Old World bisexual species.  相似文献

应用聚丙烯酰胺凝胶电泳技术分析了孤雌生殖卤虫产生的罕见雄体、两性卤虫及其杂交后代 (F1)的酯酶 (EST)、超氧物歧化酶 (SOD)、过氧化物酶 (PRX)、碱性磷酸酶 (AL P)、四唑氧化酶(TO)和乳酸脱氢酶 (L DH)的同工酶酶谱表型。对编码 5种酶的 19个位点、35个等位基因的表达进行了遗传分析。结果表明 ,在 EST、PRX和 TO酶谱中既有共显性等位基因 ,又有沉默等位基因的表达 ;卤虫 PRX、AL P和 SOD为单体酶 ,在 EST和 TO酶谱中还存在着二聚体同工酶 ;除 LDH为单态外 ,其余 5种同工酶为多态酶。  相似文献

用不同水平(分别为T1组:0 mg/L;T2组:50 mg/L;T3组:100 mg/L;T4组:150 mg/L)乳铁蛋白(lactoferrin,简称LF)强化的卤虫(Artemia franciscana)幼体投喂9~23日龄大黄鱼(Pseudosciaena crocea)仔鱼。结果表明,摄食LF强化卤虫幼体的大黄鱼仔鱼其生长无明显改善(P0.05),但大黄鱼仔鱼的成活率和抗应激能力显著提高,T3(60%)和T4(80%)组的成活率显著高于T1(40%)和T2(30%)组(P0.05)。T4组大黄鱼仔鱼的耐干露能力最强,T4组和T3组的抗高盐度(65)应激能力明显的强于T1和T2组(P0.05),T4组和T3组之间无明显的差异(P0.05)。经4 h高温(32°C)曝露后,T1和T2组的仔鱼全部死亡,T4组和T3组的成活率分别为30%和23.3%。即仔鱼的成活率和抗应激能力受卤虫强化LF水平的影响显著,LF强化的有效剂量为不低于100 mg/L强化水体。  相似文献

研究了有毒赤潮藻塔玛亚历山大藻Alexandrium tamarense对甲壳动物卤虫的干重及能值的影响.实验表明,经过18h的染毒实验,7 500尾卤虫Artemia parthenogenetica在水体中有塔玛亚历山大藻细胞或过滤液(细胞密度均为200cells·ml-1)存在的情况下,其总干重及总能值的增加明显受到抑制.卤虫总干重,对照组增加0.010 4g,而滤液组和藻细胞组均增加了0.002 2g,仅为对照组的21.1%;卤虫总能值,对照组增加0.143 5kJ,而滤液组和藻细胞组则分别增加0.018 0kJ和0.056 8kJ,分别为对照组的39.5%和12.5%.经SAS软件分析,塔玛亚历山大藻细胞对卤虫总干重的影响与对照组比较有显著性差异(p=0.037 0),而对其总能值增加则无显著差异(p=0.1251);而塔玛亚历山大藻过滤液对卤虫的总干重、总能值的增加与对照组都有显著性差异(p分别为0.021 5和0.013 5).另外,无论是藻细胞或是过滤液对卤虫的比能值均无影响.有毒塔玛亚历山大藻使卤虫总干重减少可影响其他相关海洋动物的食物供给,而对卤虫能值的影响则表明毒藻可改变食物链中能量的传递效率,从而影响处于较高营养层的生物.研究还显示该藻的不利影响可出现在赤潮发生期及消退期,这为全面了解赤潮危害提供新的视角.  相似文献

本文用数值分类的方法对来自我国山西、内蒙古以及青藏高原的共15个两性生殖卤虫品系的卵、无节幼体进行了研究.通过卵和无节幼体的聚类分析,青藏高原卤虫品系和其它品系得到很好的分离.发现青藏高原卤虫品系的卵有三个特征,即卵径大,卵壳厚,卵呈紫红色,这些特征均是对那里高寒环境的适应.研究表明,随着分布区海拔的升高,中国各卤虫品系的卵和无节幼体有增大的趋势.其中西藏拉果错卤虫的卵和无节幼体是世界上所有已知品系中最大的.  相似文献

In this study, we report on the existence of a new bisexual Artemia species, found in the high plateaus of Tibet (P. R. China). Different disciplines have been used to characterize this new population: biometrics of cysts and nauplii, morphometry of adults, cytogenetics, allozyme and DNA analyses and cross-breeding/fertility tests with known Artemia species. The results obtained justify that there is enough evidence to support the view that Artemia from Tibet is a new species with the proposed name Artemia tibetiana.  相似文献

The structure of the naturally occurringArtemia parthenogenetica population inhabiting the Mahalewaya Saltern in Sri Lanka was studied over an annual cycle, together with accompanying variations in the physico-chemical characteristics of its pond waters.Artemia was found only in the serially connected earthen ponds that serve as condenser pans in the saltern. Over the 12-month period of study, salinities in these ponds ranged from 81.5 to 199.6 ppt, temperatures from 28.2 to 33.6°C, pH from 7.9 to 9.1 and dissolved oxygen from, 1.1 to 9.0 mg L–1. The physico-chemical characters varied along a gradient across the three ponds, with salinity and temperature increasing with proximity of the condenser pans to the crystallizers of the saltern and dissolved, oxygen values showing the opposite trend. The ponds containingArtemia were shallow and had a maximum depth of 37 cm. Gross primary productivity ranged from negative values to 686.4 mg C m–2 day–1. Rainfall was distributed non-uniformly almost throughout the year, most of it being experienced from August to October, with a 254 mm maximum in October and lesser rainfall from December to May. Population numbers ranged from zero to 125 individuals L–1 and declined with increasing salinities and temperatures which related, spatilly, to increasing proximity of ponds to crystallizers and temporally, to the dry weather periods. Recruitment was best at low salinities and temperatures (104 to 114 ppt and 28.2 to 30.5°C) but also took place, although, at reduced intensities, up to 174 ppt salinity and 32.2°C temperature. Populations were mostly dominated by instar I stage nauplii with adult stages predominating only during periods of high salinity and temperature. Water management practices for salt production in the salterns included maintaining shallow depths that lead to high water temperatures and pond irrigation procedures that resulted in abrupt salinity changes. Such practices limited the size of theArtemia population as well as prevented its spread into the numerous ponds of the saltern.  相似文献
