This study measures the presence of bacteria‐sediment associations (BSAs) in an alpine, glacier‐fed watershed in the Southern Coast Mountains of British Columbia, Canada. The impact of BSAs on the creation of flocculated particles and their settling velocity are quantified using a laser transmissometer. Results from the study indicate that BSAs are present in the watershed and vary over both space and time. The percentage of bacteria associated with sediment particles was found to range from < 1% to 40%. Major sources of planktonic bacteria such as agricultural land and wastewater treatment outflow co‐occur with large decreases in the BSA ratio. Laboratory analysis demonstrates that an increase in the concentration of bacteria was associated with a decrease in the volume concentration of small particles, and a decrease in both estimated density and measured settling velocity for particles in larger size classes; consistent with flocculated particles of increasing complexity arising from combinations of primary particles and/or BSAs. Paleoenvironmental reconstructions using laminated lake sediments in alpine, glacier‐fed systems benefit from a fuller understanding of the geomorphologic processes by which they formed. While bacteria are noted to enhance formation of flocculated particles in laboratory systems, their impact upon geomorphic processes in natural systems have yet to be fully explored. © 2018 The Authors. 三种碳源对乌梁素海好氧不产氧光合细菌群落结构的影响   总被引:3,自引:1,他引:2  
好氧不产氧光合细菌(AAPB)营光合异养,在水体碳循环中起着重要作用.但是,碳源对AAPB群落结构的影响尚不明确.本文基于16S rDNA和光合反应中心合成中的关键基因——pufM基因,运用变性梯度凝胶电泳(DGGE)技术,比较分析了在微生物分离培养中常用的3种碳源(葡萄糖、丙酮酸钠、酵母提取物)对乌梁素海水体总细菌及AAPB群落结构的影响.结果表明,不同碳源诱导7~21 d后,水体总细菌群落丰富度及多样性指数均有不同程度的提升,AAPB群落则呈现先升后降的趋势.3种碳源中,丙酮酸钠添加后对总细菌及AAPB菌群的丰富度和多样性的提升效果最好.测序及系统发育结果表明,乌梁素海水体经诱导后新检测到了一些原环境中未发现的菌属,一些具有特殊碳代谢能力的菌属丰度得到了提升.本研究为充分认识和理解AAPB的生态学作用和意义,及其富集、分离和培养提供了基础依据.  相似文献

考察了膜-生物反应器(MBR)与活性污泥工艺(CAS)处理无机氨氮废水的对比研究.结果表明,同CAS相比,MBR处于高容积负荷、低污泥负荷的运行状态下,硝化效果较好,除了短期的pH控制故障外,氨氮转化为硝酸盐氮的转化率可达99%.随着运行时间的增加,膜组件的截留作用导致MBR内胞外多聚物的积累,电镜扫描(SEM)不能观察到运行后期微生物的形态变化,然而,CAS中微生物形态变化不大.2个反应器中硝化菌、亚硝化菌和异养菌的变化趋势是一致的,异养菌仅为硝化细菌的万分之一;但与MBR相比, CAS中硝化菌数量少32%,亚硝化菌的数量少35%,异养菌数量少24%.  相似文献

海水中致病菌种类繁多、致病性强,对水产养殖和人体健康存在着潜在危害。传统的检测方法培养过程复杂繁琐、耗时长、效率低,难以满足实际工作的需求。因此,建立海水中致病菌的快速、灵敏、准确的检测技术势在必行。本文综述了流式细胞技术、分子生物学方法、免疫学方法和生物传感器技术等在海水致病菌检测中的应用,总结了上述方法在海水致病菌检测中的研究现状。  相似文献

为了解重金属Hg对锦州湾海域表层沉积物中异养细菌的主要代谢酶的影响,采用Hakanson潜在生态危害指数法对锦州湾表层沉积物中重金属Hg开展潜在生态风险评价,利用平板扩散法测定了9个站位沉积物中异养细菌四种水解酶活性。结果显示:锦州湾海域表层沉积物中重金属污染严重,Hg的含量和潜在生态危害河口最为严重,向外逐步递减,表明本海域表层沉积物中的Hg主要由陆源输入;表层沉积物中异养细菌水解酶活性与抗性异养菌比例均表现为西南部河口区高于东北部外海海域,与重金属Hg的含量分布及Hakanson潜在生态危害指数基本相似,这表明长期高浓度的重金属污染,改变了锦州湾海域表层沉积物中的异养菌的生理代谢过程,在生理或遗传物质上对重金属产生了一定的抗性;磷酸酶、蛋白酶、脂肪酶与沉积物中Hg的含量具有显著正相关性,说明其对重金属Hg较为敏感,在一定程度上可作为反映锦州湾海域沉积物中重金属Hg污染状况的生物学指标。  相似文献

Prevalence of bacterial species involved in biomineralization of manganese on titanium (Ti) surfaces in marine environment was revealed in this research work. This study involves one year sea water exposure of Ti and their periodical biofilm characterization was carried out to quantify the manganese oxidizing bacterial (MOB) presence in the biofilm formed on titanium surfaces. The total viable count study of Ti coupons exposed to sea water for one year resulted in 60% of the MOB in overall biofilm population. The biochemical characterization of MOB isolates were performed for the genus level identification of the seven bacterial isolates. Further, the seven strains were subjected to 16S rRNA gene sequencing. Evolutionary analysis was performed using MEGA 7 to obtain closely related strains within the groups. The manganese oxidizing ability of the bacterial isolates were determined with Leucoberbelin Blue Assay (LBB) and Atomic Absorption Spectroscopy studies (AAS). The results show that among the isolated marine MOB species, Bacillus sp. and Leptothrix sp. have the maximum Mn oxidizing property. The microtitre plate assay was performed to determine the biofilm forming ability of the isolated marine MOB species. All the results have confirmed the prevalence of Bacillus sp. among the biofilm colonizers on Ti surfaces when exposed in sea water.  相似文献

The effect of kelp Ecklonia maxima inclusion in formulated feeds on abalone growth and gut bacterial communities has not been previously investigated in South Africa. An eight-month on-farm growth trial was conducted with sub-adult Haliotis midae (~43 mm shell length) fed graded levels of kelp in formulated feeds. Kelp inclusion (0.44–3.54% of pellet dry mass) promoted faster growth (65.7–74.5% total mass gain), with better feed and protein conversions (apparent feed conversion ratio [FCR] 1.4–1.8, apparent protein efficiency ratio [PER] 2.3–2.7), as compared with the non-supplemented feed (52.3% total mass gain, FCR 2.1, PER 1.9; p < 0.001). Abalone-gut bacterial DNA was sequenced using 16S rRNA gene pyrosequencing and the sequences were clustered into operational taxonomic units (OTUs) at a 97% similarity level. A supplementary 16S rRNA gene denaturing gradient gel electrophoresis (DGGE) analysis was employed. The dominant OTUs differed in terms of their relative abundances, with an autochthonous Mycoplasma strain being significantly more abundant (p = 0.03) in the gut of abalone fed a kelp-supplemented feed. The DGGE band patterns displayed higher within-group variability for abalone fed the control diet, suggesting that dietary kelp inclusion promotes gut-bacteria homeostasis. This may contribute to better feed utilisation and growth in abalone fed kelp-supplemented feeds.  相似文献
