僧帽牡蛎碱性磷酸酶性质的研究   总被引:12,自引:2,他引:12  
从僧帽牡蛎分离纯化出一种碱性磷酸酶,采用Sephadex G-200凝胶过滤柱层析法测得该酶分子量为1.57×10~5.以对硝基苯磷酸为底物,该酶的最适温度为43℃,最适pH值为10.0.在37℃和pH值为10.0的条件下,酶催化反应的米氏常数(K_m)为9.68×10~(-4)mol/dm~3,活化能(E_a)为45.85kJ/mol,温度系数(Q_(10))为1.80(30~40℃).对该酶的热稳定性研究表明:其在45℃以下较为稳定,50℃以上明显失活,并测定了该酶的热失活速度常数.金属离子对酶活力的效应研究结果表明:Mg~(2+)、Mn~(2+)、Ca~(2+)、Co~(2+)对酶有激活作用,Ag~+、Hg~(2+)、Zn~(2+)表现为抑制作用,Cd~(2+)几乎没有影响.Mg~(2+)是较有效的激活剂,表现为部分非竞争性效应,激活常数(K_a)为2.48×10~(-4)mol/dm~3,有Mg~(2+)存在时该酶活性中心的转换数量是无Mg~(2+)时的4.0倍.几种效应物对酶的抑制机理研究表明:HPO_4~(2-)、HAsO_4~(2-)、Cys为竞争性,其抑制常数分别为1.61、1.18和0.15mmol/dm~3;L-Phe为反竞争性,抑制常数为3.26mmol/dm~3.  相似文献

A long time series (170 years) of daily flows of the river Warta (Poland) are subject to fractal analysis. A binary variable (renewal stream) illustrating excursions of the process of flow is examined. The raw series is subject to de-seasonalization and normalization. Fractal dimensions of crossings of Warta flows are determined using a novel variant of the box-counting method. Temporal variability of the flow process is studied by determination of fractal dimensions for shifted horizons of 10 or 30 years length. Spectral properties are compared between the time series of flows, and the fractional Brownian motion which describes both the fractal structure of the process and the Hurst phenomenon. The approach may be useful in further studies of non-stationary of the process of flow, analysis of extreme hydrological events and synthetic flow generation.  相似文献

Geotechnical properties from a series of deep-sea sites in the North Atlantic and North Pacific oceans are examined to evaluate overall trends and to compare with similar fine-grained soils found on land. The study areas encompass a range of sedimentary environments dominated by combinations of turbidite and pelagic deposits. Carbonate content in excess of 20% is seen to result in a decrease in liquid limit and compressibility. Vertical profiles of geotechnical properties in the North Pacific show broader changes in down-core geotechnical properties compared to the North Atlantic and reflect the effects of long-term climatic changes and seafloor spreading. Sediments in the North Atlantic indicate significant differences depending on location, which is attributed to variability in turbidite deposition, water depth, distance from sediment sources, and the effects of bottom currents. Compared to equivalent fine-grained soils on land, deep-sea sediments are generally softer, more compressible and have higher friction angles at comparable Atterberg limits. Deeper and older sediments in the North Pacific are characterized by unusually large plastic limits, which are attributed to the presence of volcanic fractions. Empirical relationships for compression index and friction angle are discussed for sediments from both oceans.  相似文献

The complex burial and diagenetic histories of the Jurassic Fulmar and Triassic Skagerrak sandstones in the UK Central North Sea present significant challenges with regard to reservoir quality and rock property prediction. Commercial reservoir quality is retained despite deep burial and associated high temperatures and pressures. Shallow marine Fulmar sands are normally compacted (mean IGV = 26 ± 3%) yet have porosities of 21–33%. Porosity was preserved through inhibition of quartz cementation by clay and microquartz coatings, and enhanced by dissolution of framework grains (∼5%). Skagerrak fluvial sands are more compacted (mean IGV = 23 ± 2%), exhibit minor feldspar dissolution (<1%), and have porosities of 16–27%. Quartz cement averages only 2 ± 1.5% due to robust chlorite coats that cover 80% (±13%) of quartz surfaces.We modeled reservoir quality evolution using the forward diagenetic model Touchstone, which simulates porosity loss due to compaction and quartz cementation. Quantitative petrographic analyses and burial history data were used to calibrate Touchstone model parameters. The results were applied to deeper prospects for pre-drill prediction of porosity and permeability. In parallel, petrophysical data were used to characterize the elastic properties of the sandstones to provide a basis for quantitative seismic forward modeling. Experimental data and core-calibrated petrophysical results, reflecting variable in situ fluids and saturations, were used to build an elastic properties model. The model is robust and was used to generate fluid-filled sandstone properties, incorporating Touchstone results, for prospect-specific seismic attribute modeling. Well results from exploration wells are in good agreement with pre-drill Touchstone and elastic properties model predictions.  相似文献

The unique material properties of volcanic soils may cause stability problems within the soil. However, few studies have examined the composition and engineering characteristics of volcanic soils below sea level. The objective of this study is to investigate the engineering properties of volcanic soils sampled from Ulleung Island. For the volcanic soils, the index properties, particle geometry, and mineralogy are analyzed in the laboratory. An oedometer cell incorporated with bender elements is used to measure the small-strain stiffness and compressibility of the volcanic soils. To obtain the large strain strength parameter and hydraulic conductivity of the volcanic soils, direct shear tests, and constant head permeability tests are performed. The experimental results show that the basic index properties of volcanic soils sampled from Ulleung Island are very similar to the values of previously published reference data: poorly graded with a median grain size, very low fine fraction, and slightly high specific gravity. In addition, the particle surface texture features and elementary analysis indicates a dark grain color, small pits or holes in the grain, and relatively low SiO2 and high Fe2O3 contents. The friction angle of the volcanic soils depends on the relative density, and the hydraulic conductivity varies according to e3/(1 + e) and D102. The characterization of the electrical properties in Ulleung Island needs to be conducted with caution due to the high Fe2O3 content in the volcanic soils.  相似文献

Details of all known data on New Zealand snapper tagging operations to 1963 are presented and briefly analysed. Nearly 8,000 fish have been tagged, with 43 (0.5%) returned. Fishing methods have included beach‐seining, trawling, and hand‐lining; tags used have been “pig‐rings”, operculum strap tags, or Petersen discs attached by two different methods. The greatest number of returns were from line‐caught fish with Petersen discs attached through the body by nylon thread. Of the 43 fish recaptured, 33 were taken less than six miles from their tagging site, and seven had moved between 10 and 260 miles (no recapture site was given for three fish. Days at liberty ranged from one to 1,127, with a mean of 164. There were insufficient growth data for analysis.  相似文献

Abstract

The advanced piston cover (APC) has been used by the Ocean Drilling Program since 1985 for recovering soft sediments from the ocean floor. The pullout force measured on extracting the core barrel from the sediment is shown to correlate with the average shear strength of the sediment core measured in the ship's laboratory. A simple rule of thumb is derived relating the shear strength of the sediment to the pullout force. Multiple APC holes at individual sites allow the consistency of the pullout measurements to be assessed. The effects of different operational procedures during APC coring are also explored. Although generally applicable, the correlation between pullout force and laboratory measurements of shear strength breaks down for some APC holes, possibly because of the disturbance of some sediment types during the APC coring process. A better understanding of the physical process of APC coring, and its effect on the properties of the sediment both inside and immediately outside the core barrel, would indicate what confidence can be put on the measurement of pullout force as a way of evaluating the in situ shear strength of deep sea sediments.  相似文献

ABSTRACT

Using physical (acoustical engineering mechanics and scanning electron microscope) and other technical methods, is study makes a comprehensive analysis of the structural characteristics and engineering mechanics properties of sediments, the sediment structure, microstructure and their classification characteristics and in the light of the physical-mechanical characteristic parameters, the relations between the grain contact, grain accumulation, and porosity with the physical-mechanical characteristics of sediment, and reports the relationship between the acoustic physical parameters and the stress-strain properties of sediment. Results show that there are six structural types in the shelf seabed sediment of the northern South China Sea, among which, the mixed contact structural type sediment has a higher compressive strength and a higher sound velocity than others, and that in the shallow seabed there exists a mesoscale structure with high and low velocity layers.  相似文献

The effects of the 3 main wastewater discharges on the benthic fauna of the Manawatu River were studied between March 1979 and January 1980. At least 4 replicate Surber samples were taken from each of 6 sites, on 5 occasions during this period. Deleatidium sp. (Ephemeroptera), Hydora sp. (Coleoptera), and the Chironomidae were the most useful indicator organisms, according to a ranking system based on the occurrence of significant differences (P<0.05) between populations at different sites. These taxa exhibited significant differences between sites in about 70% of all inter‐site comparisons, using Mann‐Whitney U‐tests. The Coefficient of Similarity was successful at summarising the comparisons between the clean‐water and down‐stream sites, ranging between 95% similarity to the clean‐water site for a recovery site in winter, and less than 10% similarity to the clean water site for the lower reaches in summer. It is suggested that relative indices may be more appropriate than absolute indices for measuring water pollution. Water quality, indicated by species diversity, was generally good (allowing for the effects of agricultural runoff), although midsummer organic loading induced marked changes in benthic species composition, including the disappearance of Deleatidium sp. from several sites.  相似文献
