鲨肝刺激物质类似物基因在大肠杆菌中的表达与产物活性分析

构建了鲨肝刺激物质类似物基因片段的原核表达载体质粒 (pET-sHSS) ,转化大肠杆菌后通过IPTG诱导获得原核表达产物 ,并利用凝胶层析方法对表达产物进行了分离纯化 ;进一步利用MTT比色法研究了该重组产物对肝癌细胞株SMMC -7721的增殖影响。结果表明 ,在1mmol/L的IPTG的诱导下 ,鲨肝刺激物质类似物基因片段在大肠杆菌BL21菌株中获得表达 ,表达产物的相对分子质量大小约为17500u,占菌体可溶性蛋白总量的38%左右 ;纯化后的产物在低浓度下 (<50ng/L)具有明显刺激SMMC7721细胞株增殖的活性 ,高浓度下 (>100ng/L)则明显抑制该肿瘤细胞株的生长 ,与天然鲨肝刺激物质的生物活性类似     

A method to estimate the longshore sediment transport at ebb-tidal deltas based on their volumetric growth: Application to the Guadiana (Spain–Portugal border)

Current techniques assessing longshore sediment transport rates have large uncertainties, pleading for the development of alternative and complementary approaches. The present study proposes a method to estimate the decadal average rate of longshore transport at modern ebb-tidal deltas based on a sediment budget analysis of the outer shoal growth. This transport is obtained as the balance of the other contributions to the shoal with the total sediment input rate obtained from an inverse application of the inlet reservoir model. The method is applied to the Guadiana ebb-tidal delta, yielding an average longshore sediment transport rate (~85 000 m3 year⁻¹) in good agreement with expectations for the region. It is exemplified that this decadal averaged rate can be used to improve longshore sediment transport expressions in order to study its variability over shorter time scales. At the Guadiana, the yearly longshore sediment transport from the improved formula ranges from ~25 000 m³ (westward) to ~245 000 m³ (eastward) and is related to the North Atlantic Oscillation index. Overall, the proposed method constitutes an alternative tool to constrain the average longshore sediment transport rate over decades in the vicinity of tidal inlets. It is applicable to ebb-tidal deltas where the outer shoal growth (from an early to a mature stage) is well documented by bathymetric maps, and where the main transport pathways towards the outer shoal can be specified. © 2019 John Wiley & Sons, Ltd. © 2019 John Wiley & Sons, Ltd.     

三角帆蚌对水体悬浮物和叶绿素a消除量的研究

本研究在不同温度条件下分别测定了一龄(△1)和三龄(△3)三角帆蚌对水体中悬浮物和叶绿素a的消除量;研究了三角帆蚌对叶绿素消除效果与水体中藻类生物量之间的关系,并在池塘中挂养1200只三角帆蚌四个月。结果表明:三角帆蚌对水体中悬浮物和叶绿素a的消除率均随水温升高而提高,其中消除效果在总重量相同时△3>△1,较高水温条件下三角帆蚌在悬浮物中优先利用藻类;同一温度下,水体中藻类量增大时,三角帆蚌对叶绿素a的消除量增大而消除率却降低;在池塘挂养测试中与对照池塘比较,四个月后三角帆蚌因大量消除了池塘水中的悬浮物和叶绿素a,使池塘水体透明度提高从26cm提高到了80cm。

Effect of particles on the photodegradation of PAHs in natural waters-A case study for the Yellow River

Photodegradation of chrysene, benzo （a） pyrene and benzo （g, h, i） perylene in natural water of the Yellow River was studied using simulation sunlight. The effects of particulates on the photodegradation were explored. Several results arose from this study. （1） The photodegradation of PAHs can be fitted with first-order kinetics when no particulate exists in water system, and the first-order constant increases with decreasing of initial concentration of PAHs. The photodegradation rates of the three PAHs are related to their molecule absorption spectrum. （2） The existence of loess exerts two kinds of effects on the photodegradation of PAHs, including the inner filter effects and the photosensitizing effects of humic substance in loess. These two contrary effects lead to the difference of net effects among different contents of loess. When the loess contents are 0.1 and 5.0 g/L, the existence of loess stimulates the photodegradation of chrysene, benzo （a） pyrene. When the loess content is 5.0 g/L, the existence of loess stimulates the photodegradation of benzo （g, h, i） perylene. In addition, the photodegradation of PAHs can be fitted with the second-order kinetics when there is loess in the water system. （3） The soluble humic substances in loess can accelerate the photodegradation of PAHs while the in soluble part cannot. （4） Since the soluble humic substances in the suspended solids of the river have been dissolved in water,     

应用6σ质量管理方法尝试评价多目标样品氯等项目检测质量水平

应用6σ质量管理方法评价X射线荧光光谱法测定多目标地球化学样品分析中4个日常监控土壤标准物质中Cl、S、总碳、N、Na₂O、MgO、Al₂O₃、SiO₂、P、K₂O、CaO、Ba、Ti、V、Cr、Mn、TFe₂O₃、Zn、Ga、Br、Pb、Th、Rb、Sr、Y、Zr、Nb等27个项目检测质量水平。研究结果显示,随着高精度、自动化仪器的应用,多目标样品分析多个项目的精密度、准确度都得到大幅度的提高,部分检测项目的性能已经达到6σ质量水平;但有少数项目如N、总碳、Cr 等低于3σ质量水平,其精密度准确度均需改进,分析性能有待进一步提高,建议更换检测方法。用6σ质量管理方法评价地质实验分析测试质量水平与地质实验室传统质量控制方法得出的结果具有一致性,其评价方法简便、量化、直观、具有一定的应用价值。     

柴河铅锌矿成矿物质来源初步探讨

柴河铅矿成矿物质来源是多源的。矿质来源主要来自下伏的老基底岩石,部分来自矿床围岩——高于庄组条带状白云岩。基底岩石中的含矿热卤水,沿NF1这条切割震旦系地层到达基底岩石的断裂上升,运移到震旦系高于庄组成矿有利围岩（条带状白云岩）中并在有利部位富集成矿。     

射线透射图像中轻物质自动识别算法的研究

目的:针对射线透射图像中轻物质的自动识别存在的难点问题进行研究。方法:提出了基于轮廓特征和灰度特征的识别算法,结果:详细地阐述了利用识别算法进行识别的过程;结论:给出了识别算法的实验结果。     

不同过滤条件下黄色物质的吸收光谱演化特征分析

在实验室对青岛近岸现场水样进行处理、分析,获得了原始状态下水样和被过滤（0.22μm,0.45μm）黄色物质水样的吸收光谱数据,除进一步验证了黄色物质光谱分布模式外,还测得黄色物质光吸收随滤膜孔径的变化。30个现场样品在波长380nm处吸收系数统计平均值^—Ay（380）分析表明,当使用0.45μm过滤器对水样进行处理,所获得的^—Ay（380）值要比未经过处理的现场水样所测得的^—Ay（380）值高约2.1％;而用0.22μm过滤器对现场水样进行处理,所获得的^—Ay（380）值要比未经过处理的现场样品所测得的^—Ay（380）值高约12．6％,要比用0．45μm过滤器处理的现场样品所测得的^—Ay（380）值高约10.3％,这些数据的获得为现有已获黄色物质数据的“归一化”和统一评判以及未来黄色物质测量数据的准确性判断与比较提供了参考。
