Pulse amplitude modulation (PAM) fluorometry is ideally suited to measure the sub-lethal impacts of photosystem II (PSII)-inhibiting herbicides on microalgae, but key relationships between effective quantum yield [Y(II)] and the traditional endpoints growth rate (mu) and biomass increase are unknown. The effects of three PSII-inhibiting herbicides; diuron, hexazinone and atrazine, were examined on two tropical benthic microalgae; Navicula sp. (Heterokontophyta) and Nephroselmis pyriformis (Chlorophyta). The relationships between Y(II), mu and biomass increase were consistent (r(2)0.90) and linear (1:1), validating the utility of PAM fluorometry as a rapid and reliable technique to measure sub-lethal toxicity thresholds of PSII-inhibiting herbicides in these microalgae. The order of toxicity (EC(50) range) was: diuron (16-33nM)>hexazinone (25-110nM)>atrazine (130-620nm) for both algal species. Growth rate and photosynthesis were affected at diuron concentrations that have been detected in coastal areas of the Great Barrier Reef.  相似文献

Phytoplankton species composition was analyzed inside and outside of the iron-enriched patch during the SEEDS experiment. Before the iron-enrichment, the phytoplankton community consisted of similar proportions of pico-, nano- and micro-sized phytoplankton, and the micro-phytoplankton was dominated by the pennate diatom Pseudo-nitzschia turgidula. Although all the diatoms, except the nano-sized Fragilariopsis sp., increased during the two weeks of the observation period, the flora in the patch dramatically changed with the increase of phytoplankton biomass to a centric diatom-dominated community. Neritic diatoms, especially Chaetoceros debilis, showed higher growth rates than other diatoms, without any delay in the initiation of growth after the enrichment, and accounted for 90% of the micro-phytoplankton after day 9. In contrast, the oceanic diatoms showed distinct delays in the initiation of growth. We conclude that the responses of the diatoms to the manipulation of iron concentration were different by species, and the fast and intensive response of the phytoplankton to iron-enrichment resulted from the presence of a small amount of neritic diatoms at the study site. The important factors that determine the dominant species in the bloom are the potential growth rates under an iron-replete condition and the growth lag. Abundant species in the patch are widely distributed in the North Pacific and their relative contributions in the Oyashio area and at Stn KNOT are high from spring to summer. However, a characteristic difference of species composition between the SEEDS bloom and natural blooms was the lack of Thalassiosira and Coscinodiscus species in the patch, which usually account for a major part of the phytoplankton community under blooming conditions in the western North Pacific.  相似文献

The analysis of oxygen isotope ratios (δ18O) in biogenic silica (e.g. diatoms, sponge spicules, phytoliths) is an increasingly utilised palaeoclimatic proxy for lakes and oceans, providing an alternative to carbonate δ18O. Currently, the problems associated with the cleaning and extraction of pure diatom silica, especially from lake sediments, may compromise the precision associated with δ18Odiatom records. This is because, despite improvements in cleaning techniques, it is still difficult to remove silica-containing contaminants (e.g. tephra, silts and clays) from biogenic silica. Where it has proved difficult to remove all the silica bearing contaminants from a sample, mass balance calculations involving point counting have been used, but these are inherently inaccurate as they only consider the surface area of the contaminant, rather than volume. The new method described here assesses the volume of contamination in each sample of biogenic silica by using major and trace element geochemistry, allowing the δ18O of the contamination to be removed from the biogenic silica δ18O value by linear mass balance. In this case, diatom silica was contaminated with tephra, however the method should also be applicable to other contaminants such as silts and clays.  相似文献

Conventional fluorescence microscopy and visible light region fluorescence microspectrometry have been combined with laser scanning fluorescence microscopy (LSFM) to examine and characterize soluble sedimentary organic matter (SOM) in Holocene diatomaceous laminites from ODP core 1034, Saanich Inlet, British Columbia, Canada. Microscopic SOM in the laminites is dominated by two components: red-fluorescing chlorophyllinite, and yellow-fluorescing, diatom-derived oils. Both are commonly preserved within the interior of siliceous diatom auxospores and appear as an abundant and intense stain on mineral matter throughout the laminites. The mineral stain is classified as matrix chlorophyllinite and matrix oil, respectively. Accessory insoluble SOM consists of dinoflagellate, prasinophyte and coccoidal alginites, and rare woody huminite, funginite, and sporinite. Visible light region fluorescence microspectrometry of chlorophyllinite reveals a predominance at λ max 670–675 nm, which is consistent with a chlorophyll a source based on comparison with reference samples of chlorophyll a and b. The consistency in λmax and spectral character of chlorophyllinite with increasing depth in Saanich Inlet correlates with a high degree of chlorophyll preservation. Its distribution in the laminites is readily imaged by LSFM using either UV (363 nm) or blue (488 nm) laser excitation and >665 nm emission. Yellow-fluorescing diatom oils have λmax ranging from 485 to 520 nm, suggesting that they may comprise up to 70% saturates. The distribution of entrapped yellow-fluorescing diatom oils as well as matrix oils is also readily imaged with LSFM using a combination of UV laser excitation (363 nm) and 510 ±40 nm emission. Laminae in the Saanich Inlet samples which are highly enriched in both diatom oils and ‘reproductive’ auxospore cells are interpreted as recording intense episodic spring diatom blooms. The observations reported here document microscopically, in situ, possibly pristine preservation of diatom oils (including possible fatty acids) at appreciable depths within Holocene organic-rich laminites. Their recognition in recent and older strata is important, for they serve as potential biomarkers for assessing long term geo-environmental changes such as water temperature. The periodic presence of anomalously high amounts of diatom-derived biological oils in laminae within the recent sediments may mark times of unusually high nutrient levels and paleoproductivity.  相似文献

Diatoms play a significant role in the global carbon cycle through their role in biogenic silica production and the transport of organic matter to the seafloor. Recent work has shown that silicified diatom frustules contain a significant amount of organic matter, and that the proportion of diatom-bound organic matter increases with depth in the water column and sediments. Here, we investigate the association between organic matter and the mineral phase. We used a combination of scanning transmission X-ray microscopy (STXM) and carbon X-ray absorption near-edge structure (XANES) spectroscopy to characterize the distribution and composition of organic matter in frustules of the diatom Cylindrotheca closterium and a biomimetic silica gel. To our knowledge, this study represents the first successful attempt to simultaneously image and obtain chemical information about the organic matter within a diatom frustule using X-ray spectromicroscopy near the carbon edge. Organic carbon, most likely protein, was distributed throughout the frustules and was not removed by harsh chemical treatment. The physical structure of the frustules appeared to be related to the chemical composition of this organic matter, with aromatic or unsaturated carbon being concentrated in the most intricately patterned regions of the frustule. A similar physical and chemical structure was observed in a biomimetic silica gel precipitated spontaneously with polylysine. These results are consistent with the theory that organic constituents of diatom frustules direct silica precipitation and become incorporated within the silica matrix as it forms. The relationship between organic matter composition and silica morphology, the failure of harsh chemical treatments to remove this organic matter, and the spontaneous nature of the co-precipitation of silica and organic matter indicate some chemical interaction between the siliceous and organic components of diatom frustules. Frustule-bound organic matter should therefore be protected from decomposition in the water column or diagenetic alteration in sediments unless the frustule dissolves.  相似文献

Geogenic dust is commonly believed to be one of the most important environmental problems in the Middle East.The present study investigated the geochemical characteristics of atmospheric dust particles in Shiraz City(south of Iran).Atmospheric dust samples were collected through a dry collector method by using glass trays at 10 location sites in May 2018.Elemental composition was analysed through inductively coupled plasma optical emission spectrometry.Meteorological data showed that the dustiest days were usually in spring and summer,particularly in April.X-ray diffraction analysis of atmospheric dust samples indicated that the mineralogical composition of atmospheric dust was calcite+dolomite(24%)palygorskite(18%)quartz(14%)muscovite(13%)albite(11%)kaolinite(7%)gypsum(7%)zircon=anatase(3%).The high occurrence of palygorskite(16%-23%) could serve as a tracer of the source areas of dust storms from the desert of Iraq and Saudi Arabia to the South of Iran.Scanning electron microscopy indicated that the sizes of the collected dust varied from 50 μm to0.8 μm,but 10 μm was the predominant size.The atmospheric dust collected had prismatic trigonal-rhombohedral crystals and semi-rounded irregular shapes.Moreover,diatoms were detected in several samples,suggesting that emissions from dry-bed lakes,such as Hoor Al-Azim Wetland(located in the southwest of Iran),also contributed to the dust load.Backward trajectory simulations were performed at the date of sampling by using the NOAA HYSPLIT model.Results showed that the sources of atmospheric dust in the study area were the eastern area of Iraq,eastern desert of Saudi Arabia,Kuwait and Khuzestan Province.The Ca/Al ratio of the collected samples(1.14) was different from the upper continental crust(UCC) value(UCC=0.37),whereas Mg/A1(0.29),K/Al(0.22) and Ti/Al(0.07) ratios were close to the UCC value(0.04).This condition favours desert calcisols as the main mineral dust sources.Analysis of the crustal enrichment factor(EF_(crustal)) revealed geogenic sources for V,Mo,Pb,Sr,Cu and Zn(2),whereas anthropogenic sources affected As,Cd,Cr and Ni.  相似文献

Microalgae are significant contributors to biodiversity and primary production in shallow coastal habitats. The invasive Brazilian pepper (Schinusterebinthifolius) can occupy former mangrove habitats in disturbed areas, and its allelopathic capability can affect growth rates of microalgae. The four microalgae tested against dilution extracts of Schinus fruits varied in their growth response, suggesting variable but unpredictable impacts on microalgal biodiversity, primary production, and community structure.  相似文献

The siliceous sinter deposits of El Tatio geothermal field in northern Chile have been examined petrographically and mineralogically. These sinters consist of amorphous silica (opal-A) deposited around hot springs and geysers from nearly neutral, silica-saturated, sodium chloride waters. Water cooling and evaporation to dryness are the main processes that control the opal-A deposition in both subaqueous and subaerial settings, in close spatial relation to microbial communities. All fingerprints of organisms observed in the studied sinter samples represent microbes and suggest that the microbial community is moderately diverse (cyanobacteria, green bacteria, and diatoms). The most important ecological parameter is the temperature gradient, which is closely related to the observed depositional settings: 1) Geyser setting: water temperature = 70–86 °C (boiling point at El Tatio: 4200 m a.s.l.); coarse laminated sinter macrostructure with rapid local variations; biota comprises non-photosynthetic hyperthermophilic bacteria. 2) Splash areas around geysers: water temperature = 60–75 °C; laminated spicule and column macrostructure, locally forming cupolas (< 30 cm); predominant Synechococcus-like cyanobacteria. 3) Hot spring setting: water temperature = 40–60 °C; laminated spicules and columns and subspherical oncoids characterize the sinter macrostructure; filamentous cyanobacteria Phormidium and diatoms (e.g., Synedra sp.) are the most characteristic microbes. 4) Discharge environments: water temperature = 20–40 °C; sinter composed of laminated spicules and oncoids of varied shape; cyanobacterial mats of Phormidium and Calothrix and diatoms (e.g., Synedra sp.) are abundant. El Tatio is a natural laboratory of great interest because the sedimentary macrostructures and microtextures reflect the geological and biological processes involved in the primary deposition and early diagenesis of siliceous sinters.  相似文献

Laminated diatom-rich marine sediments from California and the Arctic Ocean provide a window into the seasonal climate and oceanography of the mid- and high-latitude Late Cretaceous. These remarkable shallow-buried sediments constitute palaeo-sediment traps that record exceptionally well-preserved sequential biogenic and lithogenic flux events. Many of the diatom laminae are composed of a few dominant taxa, and we use a species-based approach for palaeoecological interpretation. Contrary to many earlier interpretations, results indicate that both sites preserve a major flux of taxa adapted to exploit a strongly stratified ocean. The uppermost Maastrichtian Marca Shale of California records a seasonal cycle initiated with a spring bloom flux of diatom resting spores followed by a summer lithogenic sediment input likely driven by monsoonal storms providing river runoff and aeolian input. This is followed by a dominant diatom flux of species that thrived in the summer stratification in deep chlorophyll maxima and were sedimented in the fall when this stratification broke down. Dominant taxa comprising this group include Hemiaulus, Stephanopyxis, Stellarima and Rhizosolenia. The uppermost Campanian CESAR 6 core from the Arctic Alpha Ridge, records a spring bloom flux of resting spores followed by a more dominant summer and fall flux of diatom vegetative cells which were likely concentrated in subsurface summer blooms that generated a “summer export pulse” and by the breakdown of stratification in the fall that gave massive flux of deep chlorophyll maxima species in the fall dump. The dominance and diversity of Hemiaulus in the CESAR 6 core together with widespread evidence of N2-fixation may indicate that some diatom blooms were powered by intracellular N2-fixing cyanobacteria as in the modern oligotrophic ocean. Thin lenses of fine lithogenic sediment that occur mostly in the spring layer represent rafting by winter sea ice and support other evidence that suggests intermittent winter freezing in the Late Cretaceous Arctic.  相似文献
