1,4-蒽醌衍生物的合成

通过高酞酸酐与2-氯-5-甲基-1,4-苯醌反应,经过最长线性步骤6步和3步反应,分别以16%和47%的总产率合成了2个1,4-蒽醌衍生物12和14。其中,化合物14的合成为首次报道。所有合成的化合物均采用核磁共振波谱和质谱方法对其进行了表征和确证,并对高酞酸酐与苯醌反应生成1,4-蒽醌的反应机理进行了探讨。     

微型近红外光谱仪测量北极积雪覆盖海冰内部太阳辐照度可行性研究

The extremely low temperature, high humidity and limited power supply pose considerable challenges when using spectrometers within the Arctic sea ice. The feasibility of using a miniature low-power near-infrared spectrometer module to measure solar radiation in Arctic sea ice environments was investigated in this study.Temperature and integration time dependences of the spectrometer module were examined over the entire target operating range of –50℃ to 30℃, well below the specified operating range of this spectrometer. Using these observations, a dark output prediction model was developed to represent dark output as a function of temperature and integration time. Temperature-induced biases in the saturation output and linear operating range of the spectrometer were also determined. Temperature and integration time dependences of the signal output were evaluated. Two signal output correction models were developed and compared, to convert the signal output at any temperature within the operating temperature range and integration time to that measured at the reference temperature and integration time. The overall performance of the spectrometer was evaluated by integrating it into a refined fiber optic spectrometry system and measuring solar irradiance distribution in the ice cover with thickness of 1.85 m in the Arctic during the 9th Chinese National Arctic Research Expedition. The general shape of the measured solar irradiance above the snow surface agreed well with that measured by other commercial oceanographic spectroradiometers. The measured optical properties of the sea ice were generally comparable to those of similar ice measured using other instruments. This approach provides a general framework for assessing the feasibility of using spectrometers for applications in cold environments.

低温对不同规格刺参幼参生长与耗氧率的影响

碳水化合物的组合合成

碳水化合物的组合合成是一项新兴技术，该技术可以在短时间内合成大量用于进行生物活性筛选的寡糖及拟糖物。液相与固相合成技术可以极大地加快药物研究与开发进程。作者针对该技术在碳水化合物合成方面的研究进展情况进行讨论。     

产复合酶菌株Pseudomonas sp.NJ197产酶发酵条件的研究

从南极普利兹湾深海900m的沉积物中筛选到一株同时产多种低温复合酶的菌 株NJ197,作者对其进行碳源、氮源、无机盐、起始pH值、培养时间、接种量等发酵条 件的优化实验．实验结果证明,在以0．5％可溶性淀粉为碳源,以0．5％豆饼粉为氮 源,无机盐:0．424％NH4H2PO4、0．075％K2HPO4、0．02％MgSO4、0．5％CaCO3,起始 pH值8．5,接种36h种龄的种子10％,20℃、250r／min的条件下在旋转摇床中培养 72h,产复合酶菌株产脂肪酶和淀粉酶活性最高．最佳的生理条件下复合酶中的碱性 脂肪酶和淀粉酶的产量分别提高到22．4U／cm3和30．9U／cm3,该产复合酶菌株可以 作为诱变育种、基因工程菌改造的出发菌株,在洗涤和制革工业中有良好的应用前 景．     

乳糖诱导重组别藻蓝蛋白基因在大肠杆菌中的表达

以重组别藻蓝蛋白表达工程菌JM109（DE3）／pET28-APC作为研究对象，对于JM109（DE3）菌株在乳糖诱导下表达重组蛋白的规律进行了研究。比较分析了不同生长阶段进行诱导，最佳乳糖浓度、诱导持续时间和诱导温度等参数对重组蛋白表达的影响。实验结果表明，采用JM109（DE3）为宿主菌，经过条件优化，乳糖诱导重组蛋白的表达量可以达到IPTG诱导的水平；较低的诱导培养温度能有效提高可溶性重组蛋白的表达。最后在摇瓶发酵结果的基础上，实现了乳糖诱导工程菌的5L全自动发酵罐培养。研究结果为在大肠杆菌大规模发酵中，运用廉价、无毒的乳糖作为诱导剂提供了一定的借鉴。
