Expression of putative zinc-finger protein lcn61 gene in lymphocystis disease virus China (LCDV-cn) genome 总被引:1,自引:0,他引:1
An open reading frame (lcn61) of lymphocystis disease virus China (LCDV-cn), probably responsible for encoding putative zinc-finger proteins was amplified
and inserted into pET24a (+) vector. Then it expressed in E. coli BL21 (DE3), and His-tag fusion protein of high yield was obtained. It was found that the fusion protein existed in E. coli mainly as inclusion bodies. The bioinformatics analysis indicates that LCN61 is C2H2 type zinc-finger protein containing
four C2H2 zinc-finger motifs. This work provides a theory for functional research of lcn61 gene.

Amphioxus, a cephalochordate, is an important model fish for studies in evolution and comparative biology. A successful cell
culture from amphioxus tissues in vitro would help understanding some basic issues. To determine the optimal culture conditions for proliferation of amphioxus cells,
primary cultures were initiated from buccal cirri, tail, gill, gut and metapleural fold of amphioxus Branchiostoma belcheri tsingtauense. The media tested were L-15, F-12, M 199, MEM, DMEM, PRMI 1640 and LDF, each was supplemented with 20% fetal bovine serum.
The optimal conditions include tail tissue cultured in L-15 or F-12 with supplement of 20% FBS and 1.5% NaCl at about 25°C.

A Lagrangian tracer model is set up for Hangzhou Bay based on Coupled Hydrodynamical Ecological model for Regional Shelf Sea
(COHERENS). The study area is divided into eight subdomains to identify the dominant physical processes, and the studied periods
are March (the dry season) and July (the wet season). The model performance has been first verified by sea-surface elevation
and tidal current observations at several stations. Eight tracer experiments are designed and Lagrangian particle tracking
is simulated to examine the impact of physical processes (tide, wind and river runoff) on the transport of passive tracer
released within the surface layer. Numerical simulations and analysis indicate that: (1) wind does not change the tracer distribution
after 30 days except for those released from the south area of the bay during the wet season; (2) the tide and the Qiantang
River runoff are important for particle transport in the head area of the bay; (3) the Changjiang River runoff affects the
tracer transport at the mouth of the bay, and its impact is smaller in the dry season than in the wet season.

All gravity field functionals obtained from an Earth gravitational model (EGM) depend on the underlying terrestrial reference
frame (TRF), with respect to which the EGM’s spherical harmonic coefficients refer to. In order to maintain a coherent framework
for the comparison of current and future EGMs, it is thus important to investigate the consistency of their inherent TRFs,
especially when their use is intended for high precision studies. Following the methodology described in an earlier paper
by Kleusberg (1980), the similarity transformation parameters between the associated reference frames for several EGMs (including
the most recent CHAMP/GRACE models at the time of writing this paper) are estimated in the present study. Specifically, the
differences between the spherical harmonic coefficients for various pairs of EGMs are parameterized through a 3D-similarity
spatial transformation model that relates their underlying TRFs. From the least-squares adjustment of such a parametric model,
the origin, orientation and scale stability between the EGMs’ reference frames can be identified by estimating their corresponding
translation, rotation and scale factor parameters. Various aspects of the estimation procedure and its results are highlighted
in the paper, including data weighting schemes, the sensitivity of the results with respect to the selected harmonic spectral
band, the correlation structure and precision level of the estimated transformation parameters, the effect of the estimated
differences of the EGMs’ reference frames on their height anomaly signal, and the overall feasibility of Kleusberg’s formulae

本文从非均匀介质中波动方程出发,提出了基于一般标架的相空间(局部角度域)波传播的步进算法. 该方法在构造单程波的步进算法时,在选择标架或正交基等方面有更大的自由度. 我们以不随频率及深度变化且具有变尺度特性的Gabor_Daubechies紧标架为例,给出了单程波传输算子的具体形式及相应的波场步进算法;详细讨论了基于Gabor_Daubechies标架的传输算子的高频渐近展开问题,得出了在高频、小传输步长条件下传输算子的近似解析表达式,并给出使用条件. 通过模型算例,比较了精确传输算子与高频近似传输算子用于非均匀介质中波传播的结果,说明在一定条件下由两者得出的波场几乎是相同的. 相似文献

本文提出了一种基于双平方根算子的共炮检距道集波动方程保幅叠前深度偏移方法,将振幅误差补偿作为偏移的一部分与“运动学偏移”一起在偏移过程中实现.其基本内容包括:(1)从保幅的单平方根算子方程出发,推导出由双平方根算子定义的保幅单程波方程;(2)根据地震波摄动理论把速度场分裂为层内常速背景和变速扰动,分别在频率-波数域和频率-空间域求得波场深度延拓的偏移时移量及振幅校正系数,从而得到最终的DSR保幅波场延拓算子;(3)在高频假设条件下,把DSR保幅波场延拓公式中的积分运算进行稳相近似,得到保幅波场延拓的相移公式.理论分析和模型数值试验表明,该方法不但可以使散射能量聚焦、归位,提高成像精度;而且可以输出正确反映地下反射系数的振幅信息,为后续的地震属性分析(如AVO/AVA)提供更真实的地震信息. 相似文献
