利用紫外诱变法对三角褐指藻(Phaeodactylum tricornutum)进行诱变育种。实验得到三角褐指藻的最佳紫外辐射剂量为18 W的紫外灯距离藻液35 cm照射15 min。通过单细胞分离技术获得1株突变株UP1,与出发藻株相比,突变株UP1的EPA产量提高10.2%。研究了诱变株的最适生长及产EPA的条件,结果表明UP1在NaNO_3 75 mg/L,p H 7.5,昼夜温度17~15℃,接种量为10%时培养7天,具有最大的生长速率和EPA产量。探讨了诱变株的遗传稳定性,结果表明诱变株可稳定遗传。  相似文献

We evaluated the performances of a submersible ultraviolet fluorometer (EnviroFlu-HC, TriOS Optical Sensors) dedicated to the real time measurement of polycyclic aromatic hydrocarbons (PAHs) in the aquatic media. We conducted calibration experiments and in situ measurements in the coastal Mediterranean Sea. We found that the EnviroFlu-HC was not strictly specific to PAHs, even though it exhibited the highest sensitivity for phenanthrene, but could response to tryptophan-like material as well, and in a much less extent, to humic substances. The sensor signal showed great spatial and temporal variations in clean and polluted sites, with likely a high contribution of PAHs in the harbors, and a high contribution of tryptophan-like and humic-like materials in the sewage effluent. We conclude that the EnviroFlu-HC is a good tool for monitoring anthropogenic inputs in the coastal waters, although its utilization should be combined to other fluorescence measurements to improve the information about the nature of the aromatic compounds detected.  相似文献

The relative ability of Coriolus versicolor to grow on coir fiber as a ligninocellulosic material was examined. Addition of yeast extract to the culture increased laccase activity, which was further enhanced to the level of 1976 U/L by addition of 1 mM copper sulfate. Laccase thus produced was used without further purification for the decolorization of various dye solutions. Decolorization efficiency was compared with the conventional environment friendly oxidation technique using hydrogen peroxide in the presence of UV radiations. Laccase showed good decolorization in most of the cases. Excellent results were achieved when the dye solution was treated successively with laccase and UV/H2O2 wherein more than 80% decolorization was achieved. This value is remarkably higher than that attained either by the enzyme or UV/H2O2 photolysis alone.  相似文献

Faint absorption bands in the visible range of the smoothed vestoid spectra have been found. The bands centered near 505, 530, and 550 nm are attributed to ferrous iron in low-calcium pyroxene and are typical for pyroxene-bearing vestoid surfaces. In accordance with characteristics of the faint absorption bands around 600 and 650 nm the studied vestoid spectra can be sorted into five types. Since the same absorptions are also seen in the laboratory spectra of the minerals and meteorites, which appear to be similar to vestoid material, spectral types of the vestoids can be related to their surface compositions. Regolith of the Type-I vestoids consists of pure low-calcium pyroxenes. Minor amount of olivine along with pyroxene appear to be on the Type-II vestoids whereas the mixtures of low-calcium pyroxene with minor chromite define the Type-III and -IV. The causes for the fifth spectral type in terms of minor mineral phases are unclear now. Simulation of the spectra of vestoids was employed to estimate content of olivine (∼6-12 vol%) and chromite (∼12-30 vol%) on their surfaces.  相似文献

In this work, the treatment of actual agro‐industrial wastewaters (IWW) by a UV/H2O2 process has been investigated. The aqueous wastes were received from industrial olive oil mills and then treated by laboratory scale physicochemical methods, i. e., coagulation using ferrous and aluminum sulfate, decantation, filtration and adsorption on activated carbon. These wastes are brown colored effluents and have a residual chemical oxygen demand (COD) in the range of 1800 to 3500 mgO2 L–1, which cannot be further eliminated with physicochemical processes. The UV/H2O2 treatments were carried out under monochromatic irradiation at 254 nm using a thermostated reactor equipped with a mercury vapor lamp located in an axial position. The effects of initial H2O2 concentration, initial COD, pH and temperature have been studied in order to determine the optimum conditions for maximum color and COD removals. The experimental results reveal the suitability of the UV/H2O2 process for both removal of high levels of COD and effectively decolorizing the solution. In particular, 95% of color removal and 90% of COD removal were obtained under conditions of pH = 5 and 32°C using 2.75 g H2O2 g–1 COD L–1 during 6 h of UV‐irradiation. The treatment is unaffected by pH over the range 2 to 9. In addition, the COD removal is improved by increasing the temperature, whereas the color removal has not been affected by this parameter. The results show that the hydroxyl radicals generated from the catalytic decomposition of H2O2 by UV‐irradiation of the solution could be successfully used to mineralize the organics contained in IWW. The mineralization of the organics seems to occur in three main sequential steps: the first is the rapid decomposition of tannins leading to aromatic compounds, which are confirmed by the decolorization of the IWW; the second step corresponds to the oxidation of aromatics leading to aliphatic intermediates, which occurs by the cleavage of an aromatic ring, and is established by the removal of aromatics, and the final step is the slow oxidation of the aliphatic intermediates, which is measured by the COD removal.  相似文献

The inactivation of enzymes is of great interest for many industrial applications. The effectiveness of photoinactivation of alpha‐amylase, catalase, and urease with 222 nm radiation was investigated in comparison to that at 254 nm. The enzymes were irradiated with different fluence rates of 222 nm radiation emitted by a KrCl‐excimer lamp and with 254 nm radiation produced by a low‐pressure mercury lamp. The relative activities were calculated before and after irradiation. Degradation caused by UV‐radiation was assessed by SDS‐PAGE analysis. The results clearly demonstrated that inactivation of the proteins is much more effective with the 222 nm excimer lamp compared to the 254 nm mercury lamp. Irradiation with the excimer lamp and a UV‐fluence rate of 1000 J/m2 was sufficient to reduce the relative activities of amylase and urease to 15% and that of catalase to 60%. After irradiation with 4000 J/m2, the enzyme activity was almost completely inhibited. In contrast, after irradiation with the mercury lamp with an UV‐fluence rate of 4000 J/m2, the relative activity was still above 85%. The gel patterns showed no visible degradation after irradiation at 254 nm, but a strong and unspecific degradation was obvious after treatment at 222 nm, presumably caused by cleavage of the peptide bonds.  相似文献

在不同紫外辐射波段下,南极棕囊藻(Phaeocystis antarctica)细胞的生长率、叶绿素a、细胞内DMSP含量和DMS释放量变化的测定结果表明;UV-B对南极棕囊藻细胞生长率和叶绿素a含量有抑制效应,UV-B还可加快DMSP分解成DMS和丙烯酸的分解速率,而UV-A对该藻细胞的DMSP合成有强烈的抑制效应。鉴于在每年春季极地海洋浮游植物繁殖期间,南极棕囊藻在南极海冰带海洋浮游植物种群结构中占有的优势地位,以及该藻是极地海洋浮游植物中DMS的主要释放者,推测南极“臭氧空洞”所增加的紫外辐射可能会对南极海域的DMS释放率产生一定的影响。  相似文献

Previous studies using observations made at low spatial and spectral resolution showed that the resonance lines of He  i and He  ii are anomalously strong in the quiet Sun when compared with other transition region lines formed at similar temperatures. Here, the higher spatial and spectral resolution provided by the Coronal Diagnostic Spectrometer ( cds ) instrument on board the Solar and Heliospheric Observatory ( SOHO ) is used to re-examine the behaviour of the He  i and He  ii lines and other transition region lines, in quiet regions near Sun centre. Supergranulation cell boundaries and cell interiors are examined separately. Near-simultaneous observations with the sumer instrument provide information on the lower transition region and the electron pressure. While the lines of He  i and He  ii have a common behaviour, as do the other transition region lines, the behaviour of the helium lines relative to the other transition region lines is significantly different. The emission measure distributions that account for all transition region lines, except those of helium, fail to produce sufficient emission in the He  i and He  ii resonance lines by around an order of magnitude, in both supergranulation cell boundary and cell interior regions. The electron pressure appears to be higher in the cell interiors than in the average cell boundaries, although the uncertainties are large. While the VAL-D model gives a closer match to the He  i 584.3-Å line, it does not successfully reproduce other transition region lines.  相似文献

根据中山站 Brewer臭氧光谱仪的观测结果 ,对中山站地区近六年春季臭氧低值的特征进行研究 ,并与 TOMS卫星观测结果进行了对比分析。分析表明中山站春季臭氧值的逐日变化与极地涡旋动力过程有密切联系。Brewer和 TOMS观测结果总体上接近 ,但 TOMS要略高于地基观测结果 6~ 1 0 DU。在春季臭氧低值期间 ,UV- B测值要高于夏至时的 UV- B测值 ,30 0~30 5nm波段的 UV- B辐射强度比 32 0~ 32 5nm波段更易受春季臭氧低值的影响  相似文献
