Molecular Characterization and Expression and DNA Methylation Analyses of a Galectin-Related Protein Gene from Cynoglossus semilaevis

Galectins,a family of ?-galactoside-binding proteins,participate in both innate immunity and adaptive immunity.This study identified one novel galectin-related protein from half-smooth tongue sole Cynoglossus semilaevis,which was designated as Cs GRP.The full-length c DNA of its encoding gene was 785 bp in length with a 528 bp open reading frame encoding a putative protein of 176 amino acids.The deduced Cs GRP contained a putative 131-aa galactoside-binding lectin domain and 3 critical residues responsible for carbohydrate binding(R~(93),W~(109) and R~(114)).Genomic structural analysis revealed that Cs GRP consisted of five exons and four introns.Cs GRP showed 68% similarity with Poecilia latipinna GRP and 67% similarity with Stegastes partitus GRP.Cs GRP showed the highest expression level in liver,although its expression was detected in all tested tissues.When challenged with Vibrio harveyi,the expression of Cs GRP was significantly down-regulated in liver(P 0.05).In addition,we found that in spleen and kidney of C.semilaevis,the Cp G island of Cs GRP showed significantly higher(P 0.05) methylation level in disease-resistant family of C.semilaevis(DR-Cs) than in disease-susceptible ones(DS-Cs).Our results suggested that Cs GRP may play important roles in the immune response of C.semilaevis.Moreover,DNA methylation patterns provided valuable data for understanding the relationship between epigenetic regulation and immunity,which would assist the animal genetic research and improve the animal breeding in the future.     

深海海底光缆敷设施工余量控制的原理和控制软件的应用

为解决深海有中继海底光缆项目技术难题,中国海底电缆建设有限公司开发了海底光缆敷设施工控制软件,填补我国相关领域的空白。文章介绍海底光缆敷设施工余量控制的原理和控制软件的操作流程:海底光缆余量包括区域余量、底部余量和释放余量,余量控制是海底光缆敷设施工中最关键的核心技术环节,应用控制软件可极大地降低计算量和提高计算精确度,并可通过在施工中不断调整计划,从而极大地提高施工质量,具有传统人工计算不可比拟的优势。     

鲁中南岩溶水资源综合类型及合理调蓄研究

鲁中南岩溶区是中国北方半干旱温带岩溶区中具有代表性的地区之一。该区岩水资源分布具有明显的不均匀性和大面积分散补给而局部富集等特征。岩溶形态以溶蚀裂隙洞穴为主，地下水类型多为裂隙溶洞型。在对该区岩溶水补给，富集，径流及排泄特征的研究基础上，初步将本区地表水和地下水资源划分为两大类型，即地下岩溶水径带变化带水资源类型和排泄带水资源及地下岩溶水资源类型。对该地区地表水和地下水的合理调蓄问题进行了系统的分析和总结，并提出了相关的对策。     

低聚羧甲基壳聚糖的抗氧化性能

将低聚壳聚糖(chitosan oligosaccharide,COS)醚化得到O-羧甲基壳聚糖(O-carboxymethyl chito-sanolig osaccharide,O-CMCOS)和N-羧甲基壳聚糖(N-carboxymethyl chitosanolig osaccharide,N-CMCOS),其取代度(substituting degree,DS)均为0.54,用红外光谱对其结构进行表征。同时,考察了其结构对超氧阴离子的清除活性以及还原能力。结果表明,COS及其衍生物清除超氧阴离子的活性及还原能力的强弱顺序均为:O-CMCOSCOSN-CMCOS。     

单环刺螠硫醌氧化还原酶相互作用蛋白质的筛选

硫化物是沿海水域中常见的有害物质,对于多数生物来讲,其在微摩尔级水平便可导致生物体中毒。硫醌氧化还原酶(SQR)是一种谷胱甘肽还原酶家族的膜结合黄素蛋白,为硫化物氧化解毒的一种关键酶。本研究以体外原核表达并复性的单环刺螠SQR为材料,采用His-pulldown技术和SDS-PAGE分析,从单环刺螠体壁细胞裂解液中筛选出两个可能与SQR结合的蛋白质,分子质量分别为40 ku和60 ku。通过毛细管液相色谱-离子肼质谱分析,初步判断其为细胞色素P450(cytochrome P450)和腺苷三磷酸结合盒转运蛋白(ATP-binding cassette transporter),并初步探讨了其可能的功能。     

时空分析在MAGIS平台中的集成与实现

针对MAGIS时空分析种类多、差异大、难以集成的难题,提出了一套新的集成方法。这种方法依据时空分析复杂度函数划分分析种类,利用Batch处理与Work Flow技术多路执行分析,同时根据分析功能和操作数结构减少分析服务,并使用优先级管理服务。该方法降低了服务维护费用,减少了批量分析的整体执行时间,并提高了分析的成功率。     

SWY11-2高精度小型测温仪

介绍一种新型的SWY11-2高精度小型水温测量仪器。该仪器传感器的感温元件采用负温度系数(NTC)热敏电阻,它具有温度系数大、灵敏度高、热容量小、响应速度快、分辨率高和成本低的特点;电路系统使用24位A/D转换器、低功耗单片机,并通过充分开发芯片资源,提高系统电路集成度,从而达到体积小、功耗低、操作简单的特点;为了达到海上恶劣条件下快速操作,其结构还采用了无工具拆装的设计。仪器在海上现场与加拿大RBR公司的TR1060水温仪比测试验,结果证明仪器性能接近国外高精度产品的水平,SWY11-2型测温仪与进口同类仪器相比,具有成本低和服务方便的优势,今后定有广阔的应用前景。     

基于C/S模式的海洋地质调查数据保密框架设计与实现

海洋地质调查数据数据量庞大,结构复杂,对数据安全和保密性要求甚高。特别是涉及国家机密的成果数据、成果图件及地震图件,既要满足科研人员的查询浏览要求,又要确保图件不被恶意盗取。针对传统密码保护措施的不足与其潜在的危险性,通过对海洋地质调查数据库结构、图件索引机制、数据加密算法以及计算机底层API函数的研究,设计了一套适用于海洋地质调查数据安全的管理框架,并在广州海洋地质调查数据库管理系统中实现,系统运行稳定,确保了数据的保密性及安全性。     

Cryopreservation of Veliger Larvae of Trumpet Shell,Charonia sauliae: an Essential Preparation to Artificial Propagation

Trumpet shell, Charonia sauliae, is an endangered and valuable species, but its artificial propagation protocol has not been successfully established. To estimate the possibility of cryopreservation for larvae of C. sauliae, which is a potential preparation for its artificial reproduction and further research, in this study a protocol for the cryopreservation of veliger larvae of trumpet shell was optimized. Through a two-step cryopreservation procedure, four kinds of cryoprotectants (ethylene glycol, 1, 2-propanediol,dimethyl sulfoxide and glycerol) were employed at three concentrations (1.0, 1.5 and 2.0 molL-1) respectively and survival rates of larvae were determined after a storage of 1h. The larvae frozen with these four cryoprotectants after 1h storage were cultured, and then survival rates were determined at 24, 72 and 120h after thawing. Dimethyl sulfoxide at a concentration of 1.5 molL-1 showed the best protective effect in all experiments (p<0.05). And survival rates of larvae frozen with dimethyl sulfoxide were determined after 1,7 and 15 d of storage. The survival rates of larvae frozen with 1.5 molL-1 dimethyl sulfoxide after 1h, 1d, 7 d and 15 d of storage were 80.77% ±7.51%, 80.34% ±11.28%, 83.10% ±9.14% and 77.23%±6.22% respectively. No significant differences in survival rates of larvae frozen with dimethyl sulfoxide were observed after various storage periods (p>0.05).
