Eighteen gametophytes includingL. japonica, L. ochotensis andL. longissima, were verified with random amplified polymorphic DNA (RAPD) technique. Eighteen ten-base primers were chosen from 100 primers selected for final amplification test. Among the total of 205 bands amplified, 181 (88.3%) were polymorphic. The genetic distance among different strains ranged from 0.072 to 0.391. The dendrogram constructed by unweighted pair-group method with arithmetic (UPGMA) method showed that the female and male gametophytes of the same cell lines could be grouped in pairs respectively. It indicated that RAPD analysis could be used not only to distinguish different strains ofLaminaria, but also to distinguish male and female gametophyte within the same cell lines. There is ambiguous systematic relationship if judged merely by the present data. It seems that the use of RAPD marker is limited to elucidation of the phylogenetic relationship among the species ofLaminaria. Artificial seedling production of Sargassum thunbergii is an effective way to relieve pressure on natural resources. In order to improve the utilization of zygotes and reduce the loss of seedlings, studies on the characteristic of the zygotes release, the development of rhizoids, the attachment of germlings, and the influence of jet washing were conducted. Results show that the percent of zygotes released was increased with time in the first 60 h. The capacity of germlings attached to the substratum was significantly increased, especially coincident with the time of the new rhizoids emerged and elongated. The detachment rate of germlings significantly decreased with the delay of starting time of jet washing or the reduction of jet washing velocity. However, the jet washing at any level applied in the experiment could cause considerable loss of germlings within the 20 days after the attachment. Our study provided some parameters to optimize the operation in the early stage of seedling production.  相似文献

We conducted a phenological evaluation of Sargassum thunbergii, a common algal species, along the coast of Lidao Bay, Rongcheng, China. The local S. thunbergii population entered a maturation period from early June to mid-August, when seawater temperature was 14.4 to 25.1°C. Our results suggest an increase in temperature and day length during this period initiated thalli maturation. Inhabitants of the low tidal zone had higher thalli length and biomass compared with those from the mid-and high tidal zones. We observed a switch in the length frequency distribution and contribution of length classes to biomass between the small (<20 cm) and long (20–79.9 cm) length classes at the time of thallus maturity. This suggests there is a trade-offbetween sexual reproduction and vegetative regeneration.  相似文献

Saccharina japonica is a common macroalga in sublittoral communities of cold seawater environments,and consequently may have highly effi cient ribulose-1,5-bisphosphate carboxylase/oxygenase(Rubisco)activity for carbon assimilation.In our study,we cloned the full-length Rubisco gene from S.japonica(SJ-rbc).It contained an open reading frame for a large subunit gene(SJ-rbcL)of 1 467 bp,a small subunit gene(SJ-rbcS)of 420 bp,and a SJ-rbcL /S intergenic spacer of 269 bp.The deduced peptides of SJ-rbcL and SJ-rbcS were 488 and 139 amino acids with theoretical molecular weights and isoelectric points of 53.97 kDa,5.81 and 15.84 kDa,4.71,respectively.After induction with 1 mmol/L isopropyl-β-Dthiogalactopyranoside for 5 h and purifi cation by Ni 2+ affi nity chromatography,electrophoresis and western blot detection demonstrated successful expression of the 55 kDa SJ-rbcL protein.Real-time quantitative PCR showed that the mRNA levels of SJ-rbcL in gametophytes increased when transferred into normal growth conditions and exhibited diurnal variations: increased expression during the day but suppressed expression at night.This observation implied that Rubisco played a role in normal gametophytic growth and development.In juvenile sporophytes,mRNA levels of SJ-rbcL,carbonic anhydrase,Calvin-BensonBassham cycle-related enzyme,and chloroplast light-harvesting protein were remarkably increased under continuous light irradiance.Similarly,expression of these genes was up-regulated under blue light irradiance at 350 μmol/(m 2·s).Our results indicate that long-term white light and short-term blue light irradiance enhances juvenile sporophytic growth by synergistic effects of various photosynthetic elements.  相似文献

相比传统垂直摄影模式,倾斜摄影更符合人眼直观感受,且能提供更加丰富的三维信息,因而在测绘领域越来越受到重视。对比分析了国内外主要倾斜数字相机的性能参数,归纳了倾斜摄影测量作业的一般数据结构、成像规律。为衡量单张航摄倾斜影像的绝对倾斜程度或是两张倾斜影像之间的倾斜差异,引入了倾斜度概念。通过不同倾斜度条件下的ASIFT匹配实验,研究分析倾斜度变化对于匹配有效性的影响,验证了倾斜度计算方法的合理性。  相似文献

基于2004~2013年战略性新兴产业上市公司跨区布址数据,运用社会网络分析法对长江三角洲城市群空间结构演变规律进行了分析。研究发现:长江三角洲城市网络的主要联系流由 “Z”字形空间结构演变为“金字塔”形结构。 高行政级别城市在网络要素资源配置中占据主导地位,省会城市的中介能力呈现“倒U”型发展趋势。沪杭宁甬苏合6城市构成了长三角城市网络的核心层,核心-边缘层联系成为占据主导地位的联系方式;城市网络凝聚子群具有明显的地理粘着性,沪宁和杭州凝聚子群分别是群内联系和群际联系的标杆。  相似文献

The effect of solar wind (SW) on the North Atlantic sea surface temperature (SST) in boreal winter is examined through an analysis of observational data during 1964-2013. The North Atlantic SSTs show a pronounced meridional tripolar pattern in response to solar wind speed (SWS) variations. This pattern is broadly similar to the leading empirical orthogonal function (EOF) mode of interannual variations in the wintertime SSTs over North Atlantic. The time series of this leading EOF mode of SST shows a significant interannual period, which is the same as that of wintertime SWS. This response also appears as a compact north-south seesaw of sea level pressure and a vertical tripolar structure of zonal wind, which simultaneously resembles the North Atlantic Oscillation (NAO) in the overlying atmosphere. As compared with the typical low SWS winters, during the typical high SWS winters, the stratospheric polar night jet (PNJ) is evidently enhanced and extends from the stratosphere to the troposphere, even down to the North Atlantic Ocean surface. Notably, the North Atlantic Ocean is an exclusive region in which the SW signal spreads downward from the stratosphere to the troposphere. Thus, it seems that the SW is a possible factor for this North Atlantic SST tripolar mode. The dynamical process of stratosphere-troposphere coupling, together with the global atmospheric electric circuit-cloud microphysical process, probably accounts for the particular downward propagation of the SW signal.  相似文献

Polysaccharides were extracted from Grateloupia livida(Harv.) Yamada using hot water(extracted product denoted WGW) and then degraded in dilute sulfuric acid(degraded product denoted WGWD). The degraded mixture was then separated into four fractions through anion exchange chromatography on 2-diethylaminoethanol(DEAE)-Bio-Gel Agarose FF gel. Electrospray ionization collision-induced dissociation tandem mass spectrometry(ESI-CID-MS/MS) was performed to elucidate the structural features of all fractions. In combination with nuclear magnetic resonance spectroscopy(NMR)and infrared spectroscopy(IR) data, the major polysaccharide structures were concluded to be μ-carrageenan and κ-carrageenan. μ-Carrageenan usually has a backbone of alternating 1,3-linked β-D-galactopyranose residues sulfated at C-4 and 1,4-linked a-D-galactopyranose residues sulfated at C-6, while κ-carrageenan consists of alternating 1,3-linked β-D-galactopyranose residues sulfated at C-4 and 1,4-linked a-D-3,6-anhydrogalactopyranose residues. Trace v-carrageenan, composed of 1,3-linked β-D-galactopyranose residues sulfated at C-4 and 1,4-linked a-D-galactopyranose residues sulfated at C-2 and C-6, was also detected. Furthermore, the polysaccharide had a backbone comprising 1,3-linked β-D-galactopyranose and1,4-linked α-L-galactopyranose sulfated at C-6, which is the agarose precursor. The hydroxy groups in the galactopyranose were partially substituted by methyl and pyruvic acid acetal(PA) groups. The anticomplementary activities of WGW and its derivatives against classical pathways were measured. The native polysaccharides in WGW had higher activities, while the derivatives had much weaker activities. This indicated that the molecular weight and sulfate content were important factors affecting the anti-complement activity.  相似文献
