In order to select an optimum extraction method for the target glycoprotein (TGP) from jellyfish (Rhopilema esculentum) oral-arms, a high performance liquid chromatography (HPLC)-assay for the determination of the TGP was developed. Purified target glycoprotein was taken as a standard glycoprotein. The results showed that the calibration curves for peak area plotted against concentration for TGP were linear (r = 0.9984, y = 4.5895x+47.601) over concentrations ranging from 50 to 400 mgL-1. The mean extraction recovery was 97.84% (CV2.60%). The fractions containing TGP were isolated from jellyfish (R. esculentum) oral-arms by four extraction methods: 1) water extraction (WE), 2) phosphate buffer solution (PBS) extraction (PE), 3) ultrasound-assisted water extraction (UA-WE), 4) ultrasound-assisted PBS extraction (UA-PE). The lyophilized extract was dissolved in Milli-Q water and analyzed directly on a short TSK-GEL G4000PWXL (7.8 mm×300 mm) column. Our results indicated that the UA-PE method was the optimum extraction method selected by HPLC.  相似文献

枣庄市薛城区煤炭资源丰富,辖区内含煤面积达180km2,2000年以前4个乡镇曾有矿井70多对,目前正在生产的矿井还有16对,由于废弃矿井多,部分闭坑矿井措施不彻底、不到位,特别是陶枣矿区露头煤距地表埋藏较浅,给违法分子非法盗采留下可乘之机。非法盗采煤炭资源行为既扰乱了正常的矿业秩序,造成资源极大浪费,严重危及周边合法煤矿的安全生产,同时又对社会安全稳定带来了隐患,严厉打击非法盗采煤炭资源行为刻不容缓。  相似文献

通过对平阴县周边及其西部农田区采样调查,平阴县周边元素异常具有异常衬度较高,富集系数大等特征,西部农田区异常具有各元素套合关系好,多数异常衬度不高,富集系数小的特点。结果表明,西部农田区异常是以As,Cd为主要成分的异常,元素含量与SiO2,Al2O3,Fe2O3等常量组分间具有显著的相关性,随SiO2含量的增加而降低,随Al2O3,Fe2O3含量的增加而增加,说明土壤常量组分及其所决定的土壤质地是导致异常形成的主要因素;而平阴城区异常是以S,Hg,As,Zn,Mn等元素为主的异常,是典型的人为综合因素(化工烟尘、工厂污水、生活垃圾)产生的异常。  相似文献

This paper considers higher-harmonic forces due to wave focusing on a vertical circular cylinder. A series of experiments has been conducted in a wave flume. The first six-harmonic components of the measured wave forces are analyzed using the scale-averaged wavelet spectrum. It is noted that due to the transient nature of focused (freak) waves, Fourier analysis would not provide equivalent information to that gleaned from the analyses used herein. The results for the experiments with very steep wave crests show significant amplitudes at the fourth and fifth harmonics. These harmonics exhibit amplitudes that are the same order as the second harmonic, but much larger than those of the third harmonic. The wavelet-based bicoherence is used to detect the quadratic nonlinearity of the measured forces. And the bicoherence spectra reveal the primary mathematical reason for the existence of the striking amplitudes of the fourth and fifth harmonics: the interaction between the lower-harmonic components couple more strongly with the fourth and fifth harmonics, thus the fourth and fifth harmonics glean more energy than those of the third-harmonic components. 基于2005年至2007年上海市陈行水库和青草沙水源地的监测资料,应用基于组合权重法的水质综合评价模型对水质状况进行了综合评价。并将综合评价结果与单因子评价结果进行比较分析。研究结果表明:基于组合权重法的水质综合评价模型简便、适用,利用样本信息较为全面,能够实现对水质客观、准确、量化的分类和排序,是一种较为有效的水质评价方法。  相似文献

Denitrification in Qi'ao Island coastal zone, the Zhujiang Estuary in China   总被引:1,自引:0,他引:1  
Samples of sediments and the overlying water were collected in the Qi'ao Island coastal zone, the Zhujiang (Pearl River) Estuary (ZE). Denitrification rates, sediment oxygen demand (SOD) , and fluxes of inorganic nitrogen compounds were investigated with N2 flux method, using a self-designed continuous flow through and auto-sampling system. The results indicate that the denitrification rates varied between 222 and 908 μmol/(m2·h) with an average of 499 μmol/(m2·h). During incubation, the sediments absorbed dissolved oxygen in the overlying water with SOD ranging from 300 to 2 363 μmol/(m2·h). The denitrification rates were highly correlated with the SOD (r2 =0.77) regardless of the NO3- + NO2- concentrations in the overlying water, organ- ic carbon contents in sediments and water temperature, suggesting that the SOD was probably the main environ-mental factor controlling the denitrification in the Qi'ao Island coastal zone. There was a net flux of NO3- + NO2-into the sediments from the overlying water. The NH4+ flux from sediments into water as the result of mineraliza-tion was between 12. 3 and 210. 3 μmol/(m2·h) ,which seems limited by both organic carbon content in sedi-ment and dissolved oxygen concentration in the overlying water.  相似文献

The concentration of zinc, chromium, cadmium, nickel, lead and copper was measured on both surface sediments and core sediments in the Shenzhen Bay. The average contents of all metals have exceeded the background values in the coastal zone of Guangdong Province. Cadmium is a heavy pollution matter which indicates a strong pollution level according to the ecological risk coefficient method. The ecological risk index of the whole area is 299 which belongs to middle and strong level. The area with strong pollution level is the near shore area closing to the inside of Shenzhen Bay, Shakou area is in the middle level while the weak pollution level occurs in Shenzhenhe Estuary according to ecological risk index. The four sediment cores can be divided into two parts based on lead-210 dating, sediments size analysis and economic development period in surrounding areas since 1950 with strong human intervention. The sediment size, deposition rate and human activities have positive relationship with the contents of heavy metals which directly impact the pollution level of heavy metals in sediments in Shenzhen Bay.  相似文献

The current study was carried out over a period of one year to characterise the coastal migrant fishery of Kenya. The study looked at gears and vessels used, and ownership, demographic factors including ages of the fishers and family sizes, migrant activity and resource conservation at two main fishing villages in Kenya; Vanga and Mayungu in the south and north coasts, straddling at 4.663°S and 39.215°E and 3.214°S and 40.135°E respectively. Further, the fishers were categorised with regard to fishing, gear and vessel operation and trade, and evolution upon entry into the fishery was also assessed in order to define fisher-stake in the fishery for resource management and conservation planning. Structured questionnaires were used to interview the fishers, and data and information recorded from 1018 fishers during the survey. Migrants accounted for over 63% of the fishers in the two study sites, with majority of the fishers lying in the 15–45 year age bracket. Dependence level averages at 4–6 person families per fisher. Entry to the fishery was mainly at seamen level, progressing to fishermen and finally to fish dealers (tajiris), with the latter holding >62% capital in the fishery. Resource management in the fishery was low and only 10% of the fishers were active participants in marine conservation and community beach management issues. Fisher migrations were mainly monsoon season-linked (>58%) although social factors such as family location determined to a great extent the expanse of the migrations. The revival of fisheries cooperatives and active participation in community resource management and conservation groups is envisaged as the key to the sustainability of both the marine resources and the economies associated with this high mobility, cross-border fishery.  相似文献
