The extracellular polysaccharide hydrolase-producing strain EP-1 was isolated from seawater and identified as Paenibacillus pabuli. Furthermore, a homogeneous extracellular polysaccharide hydrolase from Paenibacillus pabuli EP-1 was purified by combining ion-exchange chromatography and size exclusion chromatography with a purification fold of 90.69 and recovery of 16.23%. Characterization of the purified polysaccharide hydrolase revealed a molecular mass of 38 k Da and optimum activity at 45℃ and pH 6.0. The polysaccharide hydrolase maintained its stability within a wide range of pH(3.0–12.0) and thermal stability when the temperature was below 50℃. The presence of Hg2+, Fe2+, Mn2+, Co2+ and SDS notably decreased hydrolase activity, and organic solvents such as formaldehyde, acetone, DMF and acetonitrile completely inhibited hydrolase activity. The purified hydrolase had no activity on agar, carrageenan, gellan gum, sodium alginate, or starch, but effectively hydrolyzed the polysaccharide from Ulva prolifera. The Km and Vmax values of this hydrolase were 43.84 mg m L-1 and 4.33 mg m L-1 min-1, respectively. The patterns of C:N:P stoichiometry across ecosystems are important in understanding biogeochemical processes. The stoichiometry of nutrients at the leaf and root level have been reported previously, but relationships of other plant organs, such as stems and the reproductive organs, remain unclear. We collected 228 samples of leaves, roots, stems and reproductive organs from 11 common plant species at 25 sites on the Tibetan Plateau to explore the relationships of C:N:P stoichiometry both within and across plant organs. The average C concentrations in the roots, leaves, stems and reproductive organs were 427.32, 410.51, 421.11 and 416.72 mg g~(-1), respectively. The shoot tissues(leaves, stems and reproductive organs) had significantly higher N and P concentrations than the roots. The N and P concentrations had a significant positive correlation within the same organ. The nutrient concentrations(N and P) and nutrient ratios(C:N, C:P and N:P) were significantly correlated across all pairwise organ combinations. Our data suggest that alpine perennial herbs share similar evolutionary histories and have constrained patterns of covariation for C concentrations, with differential patterns for N and P stoichiometry across organs. Our data also indicate that covarying sets of nutrient traits are consistent across environments and biogeographical regions and demonstrate convergent evolution in plant nutritional characteristics in extreme alpine environments. 相似文献
Urban agglomeration is caused by the continuous acceleration of the urbanization process in China. Studying the expansion of construction land can not only know the changes and development of urban agglomeration in time, but also obtain the great significance of the future management. In this study, taking Changsha-Zhuzhou-Xiangtan (Chang-Zhu-Tan) urban agglomeration in Hunan province as a study area, Landsat images from 1995 to 2014 and Autologistic-CLUE-S model simulation data were used. Moreover, several factors including gravity center, direction, distance and landscape index were considered in the analysis of the expansion. The results revealed that the construction area increased by 132.18%, from 372.28 km2 in 1995 to 864.37 km2 in 2014. And it might even reach 1327.23 km2 in 2023. Before 2014, three cities had their own respective and discrete development directions. However, because of the integration policy implementation in 2008, the Chang-Zhu-Tan began to gather, the gravity center moved southward after 2014, and the distance between cities decreased, which was in line with the development plan of urban expansion. The research methods and results were relatively reliable, and these results could provide some reference for the future land use planning and spatial allocation in the urbanization process of Chang-Zhu-Tan urban agglomeration.
