Detection of white spot syndrome virus (WSSV) ofPenaeus chinensis by in situ hybridization

White Spot Syndrome Virus (WSSV) was purified from hemolymph of infected shrimp. After nucleic acid extraction from the purified virus particles, EcoR I-digested fragments of the WSSV genome were cloned; three of these fragments were used as non-radioactive probes labeled with DIG-11-dUTP. The probes hybridized in situ, with sections located in the nuclei of all WSSV-infected tissues. The virus was detected in the gill, stomach, epidermis, and connective tissue and so on, but not detected in healthy shrimp tissues and epithelial cells of hepatopancreatic tubules of diseased shrimp. 大伙房库区近30年来植被覆盖变化分析

根据像元二分模型原理,利用1983年、2000年、2011年大伙房库区遥感影像数据提取植被覆盖遥感信息,结合植被指数计算植被覆盖度,分析比较大伙房库区不同时期植被覆盖的变化,研究大伙房库区这三个时期植被覆盖动态变化规律,为大伙房库区的生态建设和可持续发展提供决策支持。结果显示:1近30年以来,大伙房库区植被覆盖等级分布不均衡,高植被覆盖区过分集中在北部地带,西部则多为低植被覆盖区;2大伙房库区高植被覆盖面积在减少、低植被覆盖面积在增加;3大伙房库区植被覆盖总面积在下降,库区的生态环境已经遭到了破坏;4政策建议:加强监测与监管力度,合理开发利用矿产资源,积极响应国家"退耕还林、退耕还草"号召。     

Hematological changes in white spot syndrome virus-infected shrimp, Fenneropenaeus chinensis (Osbeck)

The pathological changes of hemocytes in the haemolymph and hepatopancreas were examined in experimentally and naturally WSSV (white spot syndrome virus) infected Fenneropenaeus chinensis. The results showed that the pathological manifestations of hemocytes were similar among moribund shrimps infected via injection, feeding and by nature. Firstly, the total hemocyte counts (THCs) in WSSV-infected shrimp were significantly lower than those in healthy shrimp. Secondly, necrotic, broken and disintegrated cells were often observed, and a typical hematolysis was present in the haemolymph smear of WSSV-infected shrimp. Thirdly, necrosis and typical apoptosis of hemocytes were detected with TEM in the peripheral haemolymph of WSSV-infected shrimp. Hyalinocytes and semi-granulocytes with masses of WSSVs in their nuclei often appeared, whereas no granular hemocytes with WSSV were found in the hepatopancreas of moribund infected shrimps. All our results supported that hemocytes were the main target cells of WSSV, and hyalinocytes and semigranular hemocytes seemed to be more favorable for WSSV infection in F. chinensis.     

Localization and dynamic expression of a 27.8 kDa receptor protein for lymphocystis disease virus infection in sea bass (<Emphasis Type="Italic">Lateolabrax japonicus</Emphasis>) tissues

Lymphocystis disease virus (LCDV) infects target cells by attaching to a 27.8 kDa receptor (27.8R) protein in flounder Paralichthys olivaceus, and anti-27.8R monoclonal antibodies (MAbs) have been developed. However, the 27.8R existence in tissues of sea bass (Lateolabrax japonicus) and its role in LCDV infection have remained unclear. In this study, the results of western blotting demonstrated that the same 27.8R was shared by flounder and sea bass. LCDV-free sea bass individuals were intramuscularly injected with LCDV, and viral copies were detected in tissues from 3 h post infection and showed a time-dependent increase during 9 days infection. Distribution and synthesis of 27.8R in sea bass tissues were investigated by using anti-27.8R MAbs as probes. It was found that 27.8R was distributed in all the tested tissues. The levels of 27.8R protein were highest in gill and skin, then a bit lowly in stomach, head kidney and heart, followed by spleen, intestine, blood cells, gonad and liver, and least in kidney and brain in healthy sea bass. Upon LCDV infection, 27.8R synthesis was up-regulated in each tissue, and higher in the tissues with higher LCDV copies. The 27.8R and LCDV were detected in some peripheral blood leukocytes but not in red blood cells. These results suggested that 27.8R was widely distributed in sea bass tissues, and it served as a receptor and correlated with tissue tropism of LCDV infection. Furthermore, leukocytes had the potential of being a LCDV carrier and were responsible for a systemic infection of LCDV in sea bass.     

The Gastric Shield and the Undrelying Epithelium of Chlamys Farreri:Morphological,Histological and Histochemical Characterizations

The gastric shield and underlying stomach epithelium of Chlymas ffarreri were examined at the light and ultrastructural levels.The results showed that the gastric shield consisted of two different size lobes joined together by a narrow middle piece,the thicker lobe was shaped like a funnel,but unclosed at the lateral side;the other lobe was irregularly triangular-shaped.The transverse section of the thicker lobe was obviously laminated and gradually decreased in thickness from the peak to the margins of the shield.The underlying epithelium bore numerous about 3μm diameter spherical processes formed by the apical plasmalemma of the epithelial cells becoming blunt pseudopodia.Microvilli and some interspersed cilia were present in the areas among the spherical processes regions where only microvilli existed.Rough endoplasmic reticulum,Golgi apparatus,different-sized electron-dense secretory granules and electron-lucent vacuoles as well las abundant mitochondria were present in the underlying epithelial cells.Fused droplets of the secretion from the underlying epithelial cells formed the gastric shield.     

中国地震电磁现象的观测与研究

对近年来中国地震电磁现象(包括地面电磁场、大气电场、地磁脉冲和电离层扰动等)的观测研究进行了重点介绍。     

Grain-size related nitrogen distribution in southern Yellow Sea surface sediments

Forty-eight surface sediments of the southern Yellow Sea are separated into three grain-size fractions. Four forms of extractable nitrogen (nitrogen in ion-exchangeable form (Nie), nitrogen in weak-acid extractable form (Nwa), nitrogen in strong-alkali extractable form (Nsa) and nitrogen in strong-oxidant form (N50))are obtained by the sequential extraction. The results show that the contents and the distributions of the extractable nitrogen in the southern Yellow Sea surface sediments are closely related to sediment grain size. The distributions ofNie, Nso and total nitrogen (TN) present positive correlations with fine particles content, while Nwa and Nsa does not have such correlation. The net contents of all the forms of nitrogen increase with sediment grain size finer.
