The bacteriophage P13 that infects Klebsiella serotype K13 contains a heat-stable depolymerase capable of effective degradation of exopolysaccharide(EPS) produced by this microorganism. In this study, the titer of phage P13, initially 2.0 × 107 pfu mL-1, was found increasing 20 min after infection and reached 5.0 × 109 pfu mL-1 in 60 min. Accordingly, the enzyme activity of depolymerase approached the maximum 60 min after infection. Treatment at 70℃ for 30 min inactivated all the phage, but retained over 90% of the depolymerase activity. Addition of acetone into the crude phage lysate led to precipitation of the protein, with a marked increase in bacterial EPS degradation activity and a rapid drop in the titer of phage. After partial purification by acetone precipitation and ultrafiltration centrifugation, the enzyme was separated from the phage particles, showing two components with enzyme activity on Q-Sepharose Fast Flow. The soluble enzyme had an optimum degradation activity at 60℃ and pH 6.5. Transmission electron microscopy demonstrated that the phage P13 particles were spherical with a diameter of 50 nm and a short stumpy tail. It was a double-strand DNA virus consisting of a nucleic acid molecule of 45976 bp. This work provides an efficient purification operation including thermal treatment and ultrafiltration centrifugation, to dissociate depolymerase from phage particles. The characterization of phage P13 and associated EPS depolymerase is beneficial for further application of this enzyme.  相似文献

The altimeter normalized radar cross section(NRCS) has been used to retrieve the sea surface wind speed for decades, and more than a dozen of wind speed retrieval algorithms have been proposed. Despite the continuing efforts to improve the wind speed measurements, a bias dependence on wave state persists in all wind algorithms. On the basis of recent evidence that short waves are essentially modulated by local winds and much less affected by wave state, we proposed a physics-based approach to retrieve the wind speed from the dual-frequency difference in terms of the mean square slope of short waves. A collocated dataset of coincident altimeter/buoy measurements were used to develop and validate the approach. Validation against buoy measurements indicates that the approach is almost unbiased and has an overall root mean square error of 1.24 m s-1, which is 5.3% lower than the single-parameter algorithm in operational use(Witter and Chelton, 1991) and 2.4% lower than another dual-frequency approach(Chen et al., 2002). Furthermore, the results indicate that the new approach significantly improves the wave-dependent bias compared to the single-parameter algorithm. The capacity of altimeter to retrieve sea surface wind speed appears to be limited for the case of winds below 3 m s-1. The validity of the approach at high winds needs to be further examined in the future study.  相似文献

岩性识别一直是火山岩油气勘探中的重要课题,基于测井数据的岩性识别也逐渐成为火山岩研究的需要,大数据背景下的机器学习算法为测井岩性识别提供了一个新方向。为提高某研究区火山岩岩性识别符合率,本文采用K近邻、支持向量机和自适应增强3种经典机器学习算法,对研究区内的粗面岩、非致密粗面岩、辉绿岩、辉长岩、玄武岩和非致密玄武岩等6类中基性火山岩进行岩性识别。从常规测井系列中优选对研究区岩性敏感的自然伽马、声波时差、补偿中子、深侧向电阻率和补偿密度等5种测井参数作为岩性识别模型的输入向量,从研究区内5口有岩心样品或薄片鉴定资料的目标层中选取测井数据点1 440个,其中960个作为训练样本,其余480个作为测试样本。以识别符合率和时间作为评价指标,对3种算法的识别结果进行对比分析,实验表明:自适应增强算法的分类准确率最高,6类岩性平均识别符合率达到82.10%;支持向量机算法表现良好,平均识别符合率为81.04%;K近邻算法平均识别符合率为76.04%。  相似文献

The North Qilian orogenic belt is the key to figure out the evolution and assembly of Asia. The Upper Silurian Hanxia Formation which is deposited in the north area of North Qilian Orogenic Belt is of utmost important to reveal the architecture and orogenic process of the North Qilian belt. So provenance analysis of the Hanxia Formation is of significance to reveal not only that the tectonic evolution of the central orogenic belt China, but also that Paleozoic Asia plate reconstruction. The ratios of elements and some discrimination diagrams based on geochemistry indicate that felsic rocks constitute their main source rocks. The chemical index of alteration is less than 80, indicating that the source rocks are relatively fresh and of low maturity. On the Th/Sc versus Zr/Sc scatter plot, samples from Hanxia Formation occur along the magmatic compositional variation trend of rocks, indicating that the rocks did not undergo obvious sedimentary sorting and recycling. The major and trace elements discrimination diagrams imply that Hanxia Formation rocks were derived from continental island arc. Recent studies show that the North China plate subducted southwards and produced subduction-related arc magmatism along the southern margin of the North Qilian Terrane during the Silurian. Therefore, we infer that in the late Silurian period the subduction-related arc became accreted to the Central Qilian terrane to the south, forming a composite continental arc, and the North Qilian belt accumulated in a fore-arc basin.  相似文献

Wave simulation was conducted for the period 1976 to 2005 in the South China Sea (SCS) using the wave model, WAVEWATCH-III. Wave characteristics and engineering environment were studied in the region. The wind input data are from the objective reanalysis wind datasets, which assimilate meteorological data from several sources. Comparisons of significant wave heights between simulation and TOPEX/Poseidon altimeter and buoy data show a good agreement in general. By statistical analysis, the wave characteristics, such as significant wave heights, dominant wave directions, and their seasonal variations, were discussed. The largest significant wave heights are found in winter and the smallest in spring. The annual mean dominant wave direction is northeast (NE) along the southwest (SW)-NE axis, east northeast in the northwest (NW) part of SCS, and north northeast in the southeast (SE) part of SCS. The joint distributions of wave heights and wave periods (directions) were studied. The results show a single peak pattern for joint significant wave heights and periods, and a double peak pattern for joint significant wave heights and mean directions. Furthermore, the main wave extreme parameters and directional extreme values, particularly for the 100-year return period, were also investigated. The main extreme values of significant wave heights are larger in the northern part of SCS than in the southern part, with the maximum value occurring to the southeast of Hainan Island. The direction of large directional extreme H s values is focus in E in the northern and middle sea areas of SCS, while the direction of those is focus in N in the southeast sea areas of SCS.  相似文献

我国已在多个地区进行了20余口煤层气U型水平井施工,但总体尚处于勘探实验阶段。为了研究潘庄地区煤层气U型水平井施工技术工艺,以寺河井田西区煤层气U型水平井为例,分析了实验区地质条件的可行性,重点研究了钻井技术工艺并初步考察了产气效果。结果表明:潘庄地区3#煤层含气量大、构造及水文地质条件简单,煤层产状稳定且厚度较大,原生结构煤体发育,比较适合U型水平井的施工。煤层气U型水平井具有技术工艺复杂、增产效果明显的特点,先进的工具仪器和技术工艺为煤层气U型水平井施工成功提供了技术保障,其最高产气量达到22 000m^3/d,相当于5-6口普通高产垂直井的产气效果,可为煤矿区煤层气抽采利用提供关键技术支撑。  相似文献

以水成铀矿理论为指导,通过对托斯特凹陷的构造演化、岩性岩相、氧化带及地球化学、水文地质、铀源条件分析,认为托斯特凹陷铀主要成矿阶段为渐新世末-上新世.找矿目的层古近系乌伦古河组具较好岩性岩相条件,尤其是发育稳定疏松砂体的Ⅲ旋回为砂岩型铀矿化赋存的主砂体.托斯特凹陷西部发育层间氧化带,东部发育潜水-层间氧化带,砂岩型铀矿化主要受层间氧化带控制,该凹陷具较好找矿前景,有利地段位于托斯特凹陷西部.  相似文献

Daily temperature and pressure series from 55 European meteorological stations covering the 20th century are analyzed. The overall temperature mean displays a sharp minimum near 1940 and a step-like jump near 1987. We evaluate the evolution of disturbances of these series using mean squared inter-annual variations and “lifetimes”. The decadal to secular evolutions of solar activity and temperature disturbances display similar signatures over the 20th century. Because of heterogeneity of the climate system response to solar forcing, regional and seasonal approaches are key to successful identification of these signatures. Most of the solar response is governed by the winter months, as best seen near the Atlantic Ocean. Intensities of disturbances vary by factors in excess of 2, underlining a role for the Sun as a significant forcing factor of European atmospheric variations. We speculate about the possible origin of these solar signatures. The last figure of the paper exemplifies its main results.  相似文献

晚奥陶世凯迪期早期与凯迪期晚期之交,是沉积演化和盆山转换极为关键的时期,在这个短暂时期内,川南—黔北地区几乎全部从碳酸盐沉积转换为碎屑岩沉积,详细研究这一转换时期的沉积特征及其岩相古地理变迁对探讨该区的沉积演化和盆地演化具有重要的意义。为了良好地揭示凯迪期早期与凯迪期晚期之交这一沉积转换关键时期的岩相古地理变化,采用层序岩相古地理编图思路,以凯迪阶涧草沟组与五峰组之间的界面作为编图的等时界面,编制了凯迪期早期涧草沟组顶部与凯迪期晚期五峰组底部2幅岩相古地理图,凯迪期早期涧草沟组顶部古地理面貌表现为古隆起、古暴露区、古剥蚀区、潮坪、局限台地、开阔台地和浅滩7个古地理单元;凯迪期晚期五峰组底部古地理面貌表现为古隆起、潮坪、浅水陆棚、深水陆棚和浅滩5个古地理单元。首次恢复了暴露区及其分布范围,更加客观地反映了凯迪期早期与凯迪期晚期之交岩相古地理格局及其演化过程。  相似文献
