Utilization of cheap renewable carbon feedstock for polyhydroxyalkanoate (PHA) production not only brings down its production cost but also ensures sustainability. The scope of this study was to evaluate the potential of sap extracted from felled oil palm trunk (OPT) as a novel inexpensive renewable carbon source for PHA production. OPT sap was found to be nutritionally rich and contained various fermentable sugars (5.5% w/v) as its major constituent. Termite gut isolate, Bacillus megaterium MC1 grew profoundly in mineral medium with OPT sap as carbon source and a cell density of 10.9 g/L was attained after 16 h of cultivation in shake flask cultures. A maximum poly‐3‐hydroxybutyrate [P(3HB)] content (% cell dry weight; CDW) of 30 wt% and a P(3HB) concentration of 3.28 g/L was recorded. Additionally, OPT sap extracted from younger tree trunks with prolonged storage had higher sugar content (10.8% w/v) and, when used as a growth medium without the addition of any nutrients, supported bacterial growth comparable to commercially available media.  相似文献

A mud volcano LUSI initiated its eruption on 29 May 2006, adjacent to a hydrocarbon exploration well in East Java. Ground subsidence in the vicinity of the LUSI eruptive vent was well recorded by a Synthetic Aperture Radar (SAR) PALSAR onboard the Japanese ALOS satellite. We apply an Interferometric SAR (InSAR) technique on ten PALSAR data scenes, acquired between 19 May 2006 and 21 May 2007, in order to obtain continuous maps of ground displacements around LUSI. Although the displacements in the area closest to the eruptive vent (spatial extension of about 1.5 km) are not detectable because of the erupted mud, all the processed interferograms indicate subsidence in an ellipsoidal area of approximately 4 km (north–south) × 3 km (east–west), centered at the main eruptive vent. In particular, interferograms spanning the first four months until 4 Oct. 2006 and the subsequent 46 days between 4 Oct. 2006 and 19 Nov. 2006 show at least about 70 cm and 80 cm of displacements away from the satellite, respectively. Possible causes of the subsidence, i.e., 1) loading effect of the erupted mud, 2) creation of a cylindrical mud conduit, and 3) pressure decrease and depletion of materials at depth, are investigated. The effects of the first two causes are found to be insufficient to explain the total amount of subsidence observed in the first six months. The third possibility is quantitatively examined using a boundary element approach by modeling the source of deformation as a deflating oblate spheroid. The spheroid is estimated to lie at depths of a few hundred to a thousand meters. The estimated depths are significantly shallower than determined from analyses of erupted mud samples; the difference is explained by presence of significant amount of inelastic deformation including compaction and downward transfer of material.  相似文献

In this project we investigated the ecotoxicological effects of endocrine disrupters in a four-year survey of the Mediterranean population of swordfish (Xiphias gladius). In the Mediterranean environment, top predators, such as swordfish, accumulate high concentrations of polyhalogenated aromatic hydrocarbons (PHAHs) and toxic metals, potentially incurring high toxicological risk. The effects of organochlorines and trace elements (Hg, Cd and Pb) in 192 swordfish specimens, caught in the Strait of Messina, Sicily, Italy, were investigated using vitellogenin (Vtg), zona radiata proteins (Zrp) and CYP1A (BPMO, EROD) activities. Vtg and Zrp were found to be dramatically induced in some adult male specimens, suggesting that this species is highly exposed to estrogens in the Mediterranean Sea. A role of organochlorines in this induction phenomenon is suggested by the statistically significant correlations between Zrp in plasma and PCB concentrations in muscle (p<0.032) and Vtg in plasma and PCB concentrations in liver (p<0.034) of male specimens. Levels of trace elements in liver were in the following ranges: Hg 1-22, Cd 1-28 and Pb 0-1.6 ppm d.w. These data indicate potential reproductive alterations in large pelagic fish and suggest the need for continuous monitoring to avoid reductions in the population of this fish species of high commercial and ecological interest.  相似文献

The chemical zoning profile in metamorphic minerals is often used to deduce the pressure–temperature (PT) history of rock. However, it remains difficult to restore detailed paths from zoned minerals because thermobarometric evaluation of metamorphic conditions involves several uncertainties, including measurement errors and geological noise. We propose a new stochastic framework for estimating precise PT paths from a chemical zoning structure using the Markov random field (MRF) model, which is a type of Bayesian stochastic method that is often applied to image analysis. The continuity of pressure and temperature during mineral growth is incorporated by Gaussian Markov chains as prior probabilities in order to apply the MRF model to the PT path inversion. The most probable PT path can be obtained by maximizing the posterior probability of the sequential set of P and T given the observed compositions of zoned minerals. Synthetic PT inversion tests were conducted in order to investigate the effectiveness and validity of the proposed model from zoned Mg–Fe–Ca garnet in the divariant KNCFMASH system. In the present study, the steepest descent method was implemented in order to maximize the posterior probability using the Markov chain Monte Carlo algorithm. The proposed method successfully reproduced the detailed shape of the synthetic PT path by eliminating appropriately the statistical compositional noises without operator’s subjectivity and prior knowledge. It was also used to simultaneously evaluate the uncertainty of pressure, temperature, and mineral compositions for all measurement points. The MRF method may have potential to deal with several geological uncertainties, which cause cumbersome systematic errors, by its Bayesian approach and flexible formalism, so that it comprises potentially powerful tools for various inverse problems in petrology.  相似文献

Timber harvest temporarily increases water yield; however, relationships between hydrologic and nutrient chemistry changes have not been consistent. This study quantified the effects of forest harvesting and site preparation without fertilization and with modern best management practices on nutrient concentrations and yields in small headwater streams of the Southeastern Coastal Plain. We monitored two watershed pairs for 2 years prior to and 1 year following timber harvest and for 2 more years following site preparation and planting. Treatment watersheds were clearcut, and downstream portions of streamside management zones were thinned in Fall 2003. Site preparation (herbicide application and burning) and planting followed a year later. All operations followed 1999 Georgia forestry best management practices. Previously published research revealed a large increase in water yield following harvest. Nutrient concentrations varied significantly within and between monitoring periods, even in reference watersheds. Silvicultural activities had no discernible effect on phosphorus and ammonium concentrations; however, statistically significant increases in nitrate/nitrite (67–340 µg L−1) and total nitrogen concentrations (100–400 µg L−1) in treatment watersheds followed stand re‐establishment. Nutrient yields increased after timber harvest largely as a result of increased water yields, although increased nutrient yields were small relative to inter‐annual and inter‐watershed variability and variability. Annual water yield largely explained the variability in annual nitrogen and phosphorus export from reference and treatment streams (r2 values from 0.65 to 0.98). High NOx concentrations coming from an upstream agricultural area decreased 1600–1800 µg L−1 over several hundred metres in the treatment streams by dilution, uptake or denitrification. Simultaneous measurement of coupled water, heat, and solute transport in unsaturated porous media is made possible with the multi-functional heat pulse probe (MFHPP). The probe combines a heat pulse technique for estimating soil heat properties, water flux, and water content with a Wenner array measurement of bulk soil electrical conductivity (ECbulk). To evaluate the MFHPP, we conducted controlled steady-state flow experiments in a sand column for a wide range of water saturations, flow velocities, and solute concentrations. Flow and transport processes were monitored continuously using the MFHPP. Experimental data were analyzed by inverse modeling of simultaneous water, heat, and solute transport using an adapted HYDRUS-2D model. Various optimization scenarios yielded simultaneous estimation of thermal, solute, and hydraulic parameters and variables, including thermal conductivity, volumetric water content, water flux, and thermal and solute dispersivities. We conclude that the MFHPP holds great promise as an excellent instrument for the continuous monitoring and characterization of the vadose zone.  相似文献

Most behavioral studies on hermaphroditic fishes have focused on small‐sized species, which are tractable for research. Although many species of large hermaphroditic fishes are important fishery resources, their proximate mechanisms (visual, chemical and/or behavioral cues) in the social regulation of sex change have not been determined. Determination of these would inform resource management and aquaculture. In order to get closer to understanding the proximate mechanisms underlying the social regulation of female‐to‐male sex change in large hermaphroditic fishes, this study reports situations that induced female‐to‐male sex change in black‐spot tuskfish, Choerodon schoenleinii, a species of large protogynous fish, in massive laboratory tanks. The situations differed in the possibility of male‐to‐female tactile contact and in the group sex ratio, enabling us to infer plausible proximate mechanisms underlying sex change induction. Tactile contact between individuals is suspected to be closely related to the incidence of female‐to‐male sex change in C. schoenleinii. Visual and chemical cues alone may be insufficient to inhibit such sex changes. Male‐to‐female tactile contact may have an important influence on female‐to‐male sex change, i.e., inhibition of this sex change, in this species. The effect of sex ratio of a social group on the incidence of sex change may be due to the relative frequency and intensity of male tactile contact with each female, which may vary with the number of females. In the absence of a dominant male, tactile contact among females may affect the incidence of sex change, as well as determine which individuals change sex.  相似文献

The Raman spectra of carbonaceous material (CM) from 19 metasediment samples collected from six widely separated areas of Southwest Japan and metamorphosed at temperatures from 165 to 655°C show systematic changes with metamorphic temperature that can be classified into four types: low‐grade CM (c. 150–280°C), medium‐grade CM (c. 280–400°C), high‐grade CM (c. 400–650°C), and well‐crystallized graphite (> c. 650°C). The Raman spectra of low‐grade CM exhibit features typical of amorphous carbon, in which several disordered bands (D‐band) appear in the first‐order region. In the Raman spectra of medium‐grade CM, the graphite band (G‐band) can be recognized and several abrupt changes occur in the trends for several band parameters. The observed changes indicate that CM starts to transform from amorphous carbon to crystallized graphite at around 280°C, and this transformation continues until 400°C. The G‐band becomes the most prominent peak at high‐grade CM suggesting that the CM structure is close to that of well‐crystallized graphite. In the highest temperature sample of 655°C, the Raman spectra of CM show a strong G‐band with almost no recognizable D‐band, implying the CM grain is well‐crystallized graphite. In the Raman spectra of low‐ to medium‐grade CM, comparisons of several band parameters with the known metamorphic temperature show inverse correlations between metamorphic temperature and the full width at half maximum (FWHM) of the D1‐ and D2‐bands. These correlations are calibrated as new Raman CM geothermometers, applicable in the range of c. 150–400°C. Details of the methodology for peak decomposition of Raman spectra from the low to medium temperature range are also discussed with the aim of establishing a robust and user‐friendly geothermometer.  相似文献
