The bacteriophage P13 that infects Klebsiella serotype K13 contains a heat-stable depolymerase capable of effective degradation of exopolysaccharide(EPS) produced by this microorganism. In this study, the titer of phage P13, initially 2.0 × 107 pfu mL-1, was found increasing 20 min after infection and reached 5.0 × 109 pfu mL-1 in 60 min. Accordingly, the enzyme activity of depolymerase approached the maximum 60 min after infection. Treatment at 70℃ for 30 min inactivated all the phage, but retained over 90% of the depolymerase activity. Addition of acetone into the crude phage lysate led to precipitation of the protein, with a marked increase in bacterial EPS degradation activity and a rapid drop in the titer of phage. After partial purification by acetone precipitation and ultrafiltration centrifugation, the enzyme was separated from the phage particles, showing two components with enzyme activity on Q-Sepharose Fast Flow. The soluble enzyme had an optimum degradation activity at 60℃ and pH 6.5. Transmission electron microscopy demonstrated that the phage P13 particles were spherical with a diameter of 50 nm and a short stumpy tail. It was a double-strand DNA virus consisting of a nucleic acid molecule of 45976 bp. This work provides an efficient purification operation including thermal treatment and ultrafiltration centrifugation, to dissociate depolymerase from phage particles. The characterization of phage P13 and associated EPS depolymerase is beneficial for further application of this enzyme.  相似文献

The Kendekeke polymetallic deposit, located in the middle part of the magmatic arc belt of Qimantag on the southwestern margin of the Qaidam Basin, is a polygenetic compound deposit in the Qimantag metallogenic belt of Qinghai Province. Multi-periodic ore-forming processes occurred in this deposit, including early-stage iron mineralization and lead-zinc-gold-polymetallic mineralization which was controlled by later hydrothermal process. The characteristics of the ore-forming fluids and mineralization were discussed by using the fluid inclusion petrography, Laser Raman Spectrum and micro-thermometry methods. Three stages, namely, S1-stage(copper-iron-sulfide stage), S2-stage(lead-zinc-sulfide stage) and C-stage(carbonate stage) were included in the hydrothermal process as indicated by the results of this study. The fluid inclusions are in three types: aqueous inclusion(type I), CO2-aqueous inclusion(type II) and pure CO2 inclusion(type III). Type I inclusions were observed in the S1-stage, having homogenization temperature at 240–320oC, and salinities ranging from 19.8% to 25.0%(wt % NaCl equiv.). All three types of inclusions, existing as immiscible inclusion assemblages, were presented in the S2-stage, with the lowest homogenization temperature ranging from 175 oC to 295oC, which represents the metallogenic temperature of the S2-stage. The salinities of these inclusions are in the range of 1.5% to 16%. The fluid inclusions in the C-stage belong to types I, II and III, having homogenization temperatures at 120–210oC, and salinities ranging from 0.9% to 14.5%. These observations indicate that the ore-forming fluids evolved from high-temperature to lowtemperature, from high-salinity to low-salinity, from homogenization to immiscible separation. Results of Laser Raman Spectroscopy show that high density of CO2 and CH4 were found as gas compositions in the inclusions. CO2, worked as the pH buffer of ore-forming fluids, together with reduction of organic gases(i.e. CH4, etc), affected the transport and sediment of the minerals. The fluid system alternated between open and close systems, namely, between lithostatic pressure and hydrostatic pressure systems. The calculated metallogenic pressures are in the range of 30 to 87 Mpa corresponding to 3 km mineralization depth. Under the influence of tectonic movements, immiscible separation occurred in the original ore-forming fluids, which were derived from the previous highsalinity, high-temperature magmatic fluids. The separation of CO2 changed the physicochemical properties and composition of the original fluids, and then diluted by mixing with extraneous fluids such as meteoric water and groundwater, and metallogenic materials in the fluids such as lead, zinc and gold were precipitated.  相似文献

The observed meridional overturning circulation (MOC) and meridional heat transport (MHT) estimated from the Rapid Climate Change/Meridional Circulation and Heat Flux Array (RAPID/MOCHA) at 26.5°N are used to evaluate the volume and heat transport in the eddy-resolving model LASG/IAP Climate system Ocean Model (LICOM). The authors find that the Florida Current transport and upper mid-ocean transport of the model are underestimated against the observations. The simulated variability of MOC and MHT show a high correlation with the observations, exceeding 0.6. Both the simu-lated and observed MOC and MHT show a significant seasonal variability. According to the power spectrum analysis, LICOM can represent the mesoscale eddy characteristic of the MOC similar to the observation. The model shows a high correlation of 0.58 for the internal upper mid-ocean transport (MO) and a density difference between the western and eastern boundaries, as noted in previous studies.  相似文献

This paper emphasizes that the interactive constraints of geology and isotopic dating is the best approach to construct the geological event sequence, and has compiled 106 data of reasonable isotopic ages for the igneous rocks of the Yanshan belt. We propose a sequence of mgmatic-tectonic events in the Jurassic-Cretaceous Yanshan orogen of North China. Five orogenic episodes are divided, (1) pre-and initial orogenic episode (Early Jurassic); (2) early orogenic episode (Middle Jurassic); (3) peak orogenic episode (Late Jurassic); (4) late orogenic episode (early Early Cretaceous), and (5) post-orogenic episode. Each episode is a short cycle, all of the orogenic processes construct a longer cycle, and they, in general, followed a counter-clockwise (ccw) PTt path. Finally, it is suggested that the Yanshanian movement was so intensive that the magmatism and tectonic deformation had involved all the lithosphere thickness and the late-Achaean-formed cratonic lithosphere had been significantly reworked.  相似文献

应用1995-2000年NOAA卫星资料,结合广西贵港市的数字化土地利用信息,通过时序分析方法找出典型作物代表区,并计算各典型代表区的平均、最大、最小VSWI(Vegetation Supply Water Index;植被供水指数)特征值,归纳分析水田和旱地的干旱指标,继而根据典型代表区的平均VSWI值划分旱情等级,生成农田旱情遥感图像,评估干旱情况。试验表明,该方法可用于湿润、半湿润地区的农田干旱遥感监测。  相似文献

A new large-bodied theropod dinosaur,Datanglong guangxiensis gen.et sp.nov.,was recovered from the Lower Cretaceous Xinlong Formation of the Datang Basin,Guangxi.It is unique in several features including:posteriormost dorsal vertebra with teardrop-shaped pneumatic foramen confined by enlarged pcdl,acpl and the centrum; posteriormost dorsal with well-developed,horizontal prpl; posteriormost dorsal with a parapophysis projecting more laterally than the diapophysis; brevis fossa shallow with short,ridge-like medial blade; and iliac pubic peduncle with posteroventrally expanded margin.Cladistic analysis supports the idea that this new taxon Datanglong guangxiensis is a primitive member of the Carcharodontosauria in possessing two unambiguous synapomorphies:large external pneumatic foramina and internal spaces present in the lateral surface of ilium,and a peg-and-socket ischiac articulation with the ilium.The presence of the new taxon from Guangxi further confirms that Carcharodontosauria were cosmopolitan large-bodied predators during the Early–mid Cretaceous.  相似文献

Sediment cores from the harbour and the coastal zone of Mytilene, island of Lesvos, Greece, were used to study the metal contamination caused by the discharge of untreated urban effluents into the sea. In the harbour. the upper layers were highly enriched in Cd, Cu, Pb and Zn, while no metal enrichment was recorded in the cores from the wider coastal zone. The metal data were normalized to Li (conservative element) to compensate for the natural textural and mineralogical variability. It was found that only the upper 18 cm of the core collected from the harbour of Mytilene could be reported as metal contaminated. Also, through the normalization procedure, it was found that the surface layers of coastal sediments assumed 'clean' were enriched in Pb, probably as a result of atmospheric transportation of the metal from the nearby town.  相似文献
