采用体外测定细菌浓度、胞外产物(ECP)蛋白含量和蛋白酶活力的方法,进行了鳗弧菌M3菌株在2216E培养基中的培养条件研究。结果表明,该菌株用固体培养基培养至24h左右,可得到较高的菌体浓度、ECP蛋白含量和蛋白酶活力。采用响应面分析方法设计实验,用SAS统计软件分析数据,得到NaCl浓度、pH值和温度对菌体生长及蛋白酶产量影响的回归模型。在2216E培养基的基础上,添加不同氮源、碳源物质以及不同浓度蛋白胨进行生长研究。结果表明,胰大豆蛋白胨能促进菌体生长及ECP蛋白分泌;NH4Cl与酪蛋白水解物可抑制蛋白酶的产生;牙鲆肌肉匀浆对菌体、ECP蛋白产量和蛋白酶产生有不同程度的促进作用;培养基中蛋白胨浓度为4%时菌体量与ECP蛋白含量达最高值,在蛋白胨浓度为2%时蛋白酶分泌量已稳定;1%的葡萄糖、蔗糖、甘油均能显著地提高菌体及ECP蛋白产量,却抑制了蛋白酶的产生。  相似文献

采用不同浓度的锶胁迫方法研究了叉鞭金藻生长、叶绿素、MDA含量及三种抗氧化酶SOD、CAT、GPX活性的变化。结果表明,在实验设计的各种Sr^2 浓度内叉鞭金藻均能生长,但Sr^2 浓度较高时生长受到不同程度的抑制,细胞数量比对照组分别减少15.9%和51.0%;叉鞭金藻中叶绿素含量随着Sr^2 浓度的升高而降低,呈负相关关系;叉鞭金藻中MDA含量在较低Sr^2 浓度下稍呈下降趋势,但与对照组相比并无显著差异,而在23.04mmol/L Sr^2 浓度时是对照的1.25倍;低Sr^2 浓度时,叉鞭金藻中蛋白质含量没有大的变化,当浓度超过5.76mmol/L时则引起蛋白质含量下降;叉鞭金藻中抗氧化酶SOD在低Sr^2 时分别降低12.5%、7.5%和9.5%,高Sr^2 时显著升高,为对照组的85.7%;CAT、GPX活性在低Sr^2 浓度时差别不大,但当浓度为23.04mmol/L时升高显著,比对照组分别增加了19.8%和74.4%。  相似文献

Using the National Center for Enviromental Prediction/National Center for Atmospheric Research (NCEP/NCAR) reanalysis data and NOAA satellite-obser ved outgoing long-wave radiation (OLR) data, the development of the South China Sea (SCS) summer monsoon and intraseasonal (30-60 d) oscillation (ISO) have been examined. The results show that there exists obvious interannual variability of intraseasonal oscillaiton. Using the 16 a time series of filtered OLR averaged over the SCS, an index is defined to define ““onset events““ over the SCS on the ISO time scales. Of the 16 a examined here, 10 shows a strong ISO signal in the onset of monsoon convection over the SCS. In these cases, the ISO initially suppresses the seasonal development of southwesterly and cyclonic circulation over the SCS before the ISO onset. As the ISO propagates northeastward, the low frequency cyclonic circulation anomaly occurs in the SCS and the low frequency southwesterly wind and convection over here dramatically intensify. The northeast progression of the ISO anomaleis plays a role in the initial suppression and then acceleration of the seasonal cycle of the SCS summer monsoon.  相似文献

Study on form characteristics of nitrogen in marine sediments is the primary method to research its biogeochemical cycling and nitrogen form characteristics in core sediments can reflect the process and results of early diagenesis in a certain degree. In this paper, Sequential extraction process in natural grain size was used for studying the existent forms of nitrogen in five core sediments of the southern Bohai Sea for the first time. Nitrogen was divided into two parts -transferable and fixed based on whether it could be extracted by the reagent. Distributions and early diagenesis of transferable nitrogen forms in the southern Bohai Sea were researched integratedly. Results indicate that IEF - N and OSF-N are predominant forms in transferable part in the studied core sediments. Contents of different nitrogen forms vary differently with depth, and have different diagenesis process. 主要根据近几年来中韩黄海水循环动力学合作调查结果,结合有关观测资料,进一步分析了冬至初春黄海暖流的路径和起源.与以往类似研究不同的主要有两点:(1)初步探讨了黄海暖流路径的季节和年际变异,并指出这种变异与北向风的强弱密切相关;(2)通过分析济州岛西侧海域混合水的去向,进一步确认了部分混合水绕济州岛运行,并进入济州海峡这一事实.同时,初步揭示进入黄海的混合水,即黄海暖流水,含有更多的东海陆架水成分.  相似文献

A total of 67 samples from the upper and lower sediment traps in the central South China Sea were analyzed, which were collected during 1993~1996. It is indicated that the distribution of stable isotope values, surface primary productivity, fluxes of total particulate matter, carbonate, biogenic opal, organic carbon, planktonic foraminiferal species and their total amount exhibit obviously seasonal and annual fluctuations. High values of the fluxes occurred in the prevailing periods of the northeastern and southwestern monsoons, and the low values occurred during the periods between the two monsoons. The fluxes of some planktonic foraminiferal species (Globigerinoides sacculifer, G. ruber, Globigerinita glutinata, Neogloboquadrina dutertrei) and their percentages also exhibit two prominent peaks during the prevailing periods of the northeastern and southwestern monsoons respectively, while those of Globigerina bulloides, Globorotalia menardii and Pulleniatina obliquiloculata only exhibit one peak in the prevailing periods of the northeastern monsoon. In addition, fluxes and percentages of Globigerinoides sacculifer and Globorotalia menardii as well as the fluxes of carbonate and total amount of planktonic foraminifera decrease gradually from 1993 to 1996, and those of Globigerina bulloides, Globigerinita glutinata and biogenic opal increase gradually from 1993 to 1996. The fluxes of carbonate and organic carbon in the upper trap are higher than those in the lower one. The study indicates that the seasonal and annual variations of the sediment fluxes and planktonic foraminiferal species are mainly controlled by the changes of surface primary productivity and hydrological conditions related to the East Asian monsoon. The lower carbonate and organic carbon fluxes in the lower trap are related to the dissolution.  相似文献

采用廉价的乳糖替代IPTG诱导重组别藻蓝蛋白(rAPC)在大肠杆菌(Escherichiacoli)JM109中表达,对诱导产物表达的培养基、培养条件、诱导剂的浓度和诱导时机进行了优化,将优化条件用于5L自控发酵罐进行高密度培养。结果表明,对rAPC表达的条件进行优化后,乳糖诱导rAPC的表达率可达26.8%,与IPTG的诱导结果接近;在高密度培养中,OD600最大达21.8。研究结果可为乳糖替代IPTG大规模生产rAPC奠定基础。  相似文献

Copper complexing ligand concentrations in the Daya Bay, Qingdao coast, Jiaozhou Bay, South China Sea and Huanghe Estuary waters were determined by the anodic stripping voltammetry technique. The distribution regularity and the relationship with other parameters were discussed. The results were as follows: Copper complexing ligand concentrations of the South China Sea were a little higher than those of other sea areas, and they were apparently higher than those of the ocean. Compared with the subsurface layer (SSL) in the sea surface microlayer copper complexing ligand concentrations showed an enrichment phenomenon, of which the mechanism is similar to dissolved organic matter. The metal complexing ligand concentration profiles of the South China Sea showed that the value in the sea surface was the highest, then it decreased with depth accruing, and a higher value appeared at the bottom. Copper complexing ligand concentrations were higher than those of cadmium and lead. Ligands in each sea area exhibited a complicated property. In short, the distribution regularity of copper complexing ligand concentrations in China' s coastal waters was consistent with that of other regions in the world. Meanwhile, the positive relationship between the copper complexing ligand concentrations and biological oxygen demand, chemical oxygen demand, dissolved organic carbon, and viscosity were found clearly.  相似文献
