Background

Reactive oxygen species (ROS) are vital regulators of many cellular functions in the body. The intracellular ROS concentration is highly regulated by a balance between pro-oxidants and anti-oxidants. A chronic excess of pro-oxidants leads to elevated ROS concentrations and inflammation, possibly initiating or enhancing disease onset. Mineral-induced generation of ROS, the role of minerals in upregulating cellular ROS, and their role in the development of several occupational diseases are now widely recognized. However, there is no standard protocol to determine changes in ROS production in cells after exposure to mineral dust or earth materials in general. In this study, a new method for determining the degree of cellular toxicity (i.e., cytotoxicity) of particles is described that will help bridge the gap in knowledge.

Results

By measuring the production of ROS and the viability of cells, an inflammatory stress response (ISR) indicator is defined. This approach normalizes the ROS upregulation with respect to the number of viable cells at the time of measurement. We conducted experiments on a series of minerals and soils that represent materials that are inert (i.e., glass beads, anatase, and a soil with low trace element content), moderately reactive (i.e., soil with high trace element content), and highly reactive (i.e., pyrite). Inert materials generated the lowest ISR, averaging 350% compared to the control. Acid washed pyrite produced the highest ISR (1,100 fold higher than the control). The measurements conducted as a function of time showed a complex response. Most materials showed an increase in ISR with particle loading.

Conclusions

The amount of cellularly generated ROS and cell viability combined provide a better understanding of particle-induced oxidative stress. The results indicate that some earth materials may solicit an initial burst of ROS, followed by a second phase in which cell viability decreases and ROS production increases, leading to a high ISR value. Hence, measurements conducted over a range of particle loading combined with multiple data measurements up to 24 hours can provide new insights in the possible effect of exposure to earth materials on human health.  相似文献

The abundant iron sulfide mineral pyrite has been shown to catalytically produce hydrogen peroxide (H2O2) and hydroxyl radical ( . OH) in slurries of oxygenated water. Understanding the formation and fate of these reactive oxygen species is important to biological and ecological systems as exposure can lead to deleterious health effects, but also environmental engineering during the optimization of remediation approaches for possible treatment of contaminated waste streams. This study presents the use of the amino acid phenylalanine (Phe) to monitor the kinetics of pyrite-induced . OH formation through rates of hydroxylation forming three isomers of tyrosine (Tyr) - ortho-, meta-, and para-Tyr. Results indicate that about 50% of the Phe loss results in Tyr formation, and that these products further react with . OH at rates comparable to Phe. The overall loss of Phe appeared to be pseudo first-order in [Phe] as a function of time, but for the first time it is shown that initial rates were much less than first-order as a function of initial substrate concentration, [Phe]o. These results can be rationalized by considering that the effective concentration of . OH in solution is lower at a higher level of reactant and that an increasing fraction of . OH is consumed by Phe-degradation products as a function of time. A simplified first-order model was created to describe Phe loss in pyrite slurries which incorporates the [Phe]o, a first-order dependence on pyrite surface area, the assumption that all Phe degradation products compete equally for the limited supply of highly reactive . OH, and a flux that is related to the release of H2O2 from the pyrite surface (a result of the incomplete reduction of oxygen at the pyrite surface). An empirically derived rate constant, K pyr , was introduced to describe a variable . OH-reactivity for different batches of pyrite. Both the simplified first-order kinetic model, and a more detailed numerical simulation, yielded results that compare well to the observed kinetic data describing the effects of variations in concentrations of both initial Phe and pyrite. This work supports the use of Phe as a useful probe to assess the formation of . OH in the presence of pyrite, and its possible utility for similar applications with other minerals.  相似文献

This study suggests a novel approach to the retrieval of soil surface parameters using a single-acquisition single-configuration synthetic-aperture radar (SAR) system. Soil surface parameters such as soil moisture and surface roughness are key elements for many environmental studies, including Earth surface water cycles, energy exchange, agriculture, and geology. Remote sensing techniques, especially SAR data, are commonly used to retrieve such soil surface parameters over large areas. Several backscattering models have been proposed for soil surface parameters retrieval from SAR data. However, commonly, these backscattering models require multi configuration SAR data, including multi-polarization, multi-frequency, and multi-incidence angle. Here we propose a methodology that employs single-acquisition single-configuration SAR data for the retrieval of soil surface parameters. The originality is to use single-acquisition single-configuration SAR data to retrieve the soil surface parameters using an optimization approach by the genetic algorithm (GA); we have used the modified Dubois model (MDM) in HH polarization as the backscattering model. Three HH polarization and C band data sets from Quebec (Radarsat-1), Ontario (SIR-C), and Oklahoma (AIRSAR) were analyzed. The retrieved values of soil moisture and soil surface roughness were then compared to ground truth measurements with corresponding parameters. We employed diverse criteria, including the mean absolute error (MAE), the root mean square error (RMSE), the coefficient of performance (CP), and the correlation coefficient to investigate the performance of the proposed methodology. This analysis suggests the capability of the GA for the retrieval of soil surface parameters. Based on our findings, this method presents a viable alternative approach to the retrieval of soil surface parameters when only single-acquisition single-configuration SAR data is available.  相似文献

The rock engineering classification system is based on six parameters defined by Bieniawski [5], who employed parallel sets of linguistic and numerical criteria that were acknowledged to influence the behaviour of rock masses and the stability of rock structures. Consequently, experts frequently relate rock joints and discontinuities as well as ground water conditions in linguistic terms, with rough calculations. Recently, intelligence system approaches such as artificial neural network (ANN) and neuro-fuzzy methods have been used successfully for time series modelling. Using neuro-fuzzy approaches, which enable the information that is stored in trained networks to be expressed in the form of a fuzzy rule base, would help to overcome this issue. This paper presents the results of a study of the application of neuro-fuzzy methods to predict rock mass rating. We note that the proposed weights technique was applied in this process. We show that neuro-fuzzy methods give better predictions than conventional modelling approaches.  相似文献

The occurrence of infections with Trichinella sp. in polar bears (Ursus maritimus) from northeastern Greenland has been studied by examination of muscle samples, mainly diaphragm, from 38 animals shot during the period 1983-1987. Trichinella larvae were demonstrated in 12 bears (32%) with an average of 9.2 larvae/g muscle tissue. No bears younger than three years old were infected. The prevalence of Trichinella among bears of the age group 3-4 years was 25% and 53% among older animals.  相似文献

Enhancing of the efficient tissue culture protocol for somatic embryos would facilitate the engineered breeding plants program. In this report, we describe the reproducible protocol of Malaysian rice (Oryza sativa L.) cultivar MR219 through somatic embryogenesis. Effect of a wide spectrum of exogenesis materials was assessed in three phases, namely callogenesis, proliferation and regeneration. Initially, rice seeds were subjected under various auxin treatments. Secondly, the effect of different concentrations of 2,4-D on callus induction was evaluated. In the next step, the efficiency of different explants was identified. Subsequently, the effects of different auxins, cytokinins, l-proline, casein hydrolysate and potassium metasilicate concentrations on the callus proliferation and regeneration were considered. For the callogenesis phase, 2 mg L?1of 2,4-D and roots were chosen as the best auxin and explant. In the callus proliferation stage, the highest efficiency was observed at week eight in the MS media supplemented with 2 mg L?1 of 2,4-D, 2 mg L?1 of kinetin, 50 mg L?1 of l-proline, 100 mg L?1 of casein hydrolysate and 30 mg L?1 of potassium metasilicate. In the last phase of the research, the MS media added with 3 mg L?1 of kinetin, 30 mg L?1of potassium metasilicate and 2 mg L?1 of NAA were selected. Meanwhile, to promote the roots of regenerated explants, 0.4 mg L?1 of IBA has shown potential as an appropriate activator.  相似文献

A hybrid dimensional analysis fuzzy inference system approach was introduced to predict blast-induced flyrock in surface mining, by integrating a dimensional analysis procedure and Mamdani’s fuzzy inference system. In the dimensional analysis, the blast-induced flyrock was considered as a function of the most effective parameters. Hence, a number of dimensionless products resulted and were used as input and output parameters of Mamdani’s fuzzy inference system. The capability of the hybrid approach was determined by comparing its results with the real measurement of flyrock in the case of a copper mine, based on a number of 320 in situ blasting datasets. Predictions by the system were close to the real measurements. Sensitivity analysis of the hybrid dimensional analysis fuzzy inference system showed that the most effective dimensionless products on flyrock distance were spacing, the multiplication of rock mass rating and hole length, and the subtraction of burden and hole length multiplication and stemming length.  相似文献
