Metallothionein was quantified in sea bass Dicentrarchus labrax intraperitoneally (i.p.) injected with different Cu, Cd and Hg doses (50-250 microg kg(-1) wet wt) after 48 h exposure. A distinct peak with 16.8 min retention time was obtained by reversed-phase high performance liquid chromatography coupled to fluorescence detection (RP-HPLC-FD) with the three metals. Total metallothionein levels assayed in unheated liver extracts by RP-HPLC-FD were significantly higher (1.3-1.95-fold) than those obtained by the well-established spectrophotometric method. In the RP-HPLC-FD method, metallothionein increased linearly with Cu and Hg doses, being saturated beyond 100 mug kg(-1) Cd. Maximum induction was obtained at 100 microg kg(-1) Cd (5.3-fold), and 250 microg kg(-1) Cu or Hg (8- and 5.1-fold, respectively). Previous field and laboratory studies have identified the alga Lobophora variegata as a good candidate for biomonitoring metal contamination in the New Caledonia lagoon which is subjected to intensive and extensive metal inputs from land-based mining activities. The aim of this work was to further assess the bioindicative potential of this species by investigating, in the field, its bioaccumulation capacity for local key contaminants, i.e. Ag, As, Cd, Co, Cr, Cu, Mn, Ni and Zn. Algae from clean and contaminated sites were cross-transplanted for a period of three months in order to determine the in situ uptake and depuration kinetics of the nine elements. Results indicate that algae transplanted to the contaminated site displayed a significant linear increase in concentration with time for Ag, As, Cd, Co, Cr, Cu, Mn and Ni. In contrast, algae transplanted to the clean site did not show major depuration of these elements, except for Co. Overall, L. variegata showed a rapid temporal response in metal uptake, especially for the elements intensively released into the coastal environment of New Caledonia (viz., Co, Cr, Mn and Ni). This species appears therefore as an excellent bioindicator species of metal contamination in this area. Our results also provide background information necessary for using L. variegata under in situ experimental conditions so as to provide better quantitative information on ambient metal contamination levels. The wide distribution of L. variegata in tropical areas further enhances its potential as a bioindicator species of metal contamination in other tropical coastal environments. 相似文献

Human fecal contamination of coral reefs is a major cause of concern. Conventional methods used to monitor microbial water quality cannot be used to discriminate between different fecal pollution sources. Fecal coliforms, enterococci, and human-specific Bacteroides (HF183, HF134), general Bacteroides-Prevotella (GB32), and Clostridium coccoides group (CP) 16S rDNA PCR assays were used to test for the presence of non-point source fecal contamination across the southwestern Puerto Rico shelf. Inshore waters were highly turbid, consistently receiving fecal pollution from variable sources, and showing the highest frequency of positive molecular marker signals. Signals were also detected at offshore waters in compliance with existing microbiological quality regulations. Phylogenetic analysis showed that most isolates were of human fecal origin. The geographic extent of non-point source fecal pollution was large and impacted extensive coral reef systems. This could have deleterious long-term impacts on public health, local fisheries and in tourism potential if not adequately addressed. 相似文献

Concentrations of PCBs and their toxic potential were examined in subcutaneous fat of eight albatross and one petrel species collected from the North Pacific and the Southern Oceans. Among all the species analyzed, high PCB levels were found in adult male blackfooted albatross from the North Pacific with the mean value of 92 microg/g wet weight. No significant gender difference in PCB accumulation was observed (P>0.1). The mean PCB levels in Southern Oceanic birds were 1 or 2 orders of magnitude lower than those from the North Pacific albatrosses. A regional-specific accumulation of non-ortho coplanar congeners were observed, most birds from the Southern Ocean had higher IUPAC 169 levels while IUPAC 126 concentrations were higher in those from the North Pacific. The estimated toxic equivalents for black-footed and Laysan albatrosses from the North Pacific were in the same range of some fish-eating birds, which were highly contaminated by PCBs. The correlation between ratio of IUPAC 169/126 concentration and total PCBs concentration indicated the possibility of induction in cytochrome P450 activities in North Pacific albatrosses (P<0.01). The calculated hazard indices indicated that black-footed and Laysan albatrosses inhabiting in the North Pacific had similar threshold levels which were known to cause toxic effects in some populations of fish-eating birds. 相似文献

A simple apparatus was designed to effectively isolate magnetotactic bacteria from soils or sediments based on their magnetotaxis. Through a series of processes including sample incubation, MTB harvesting, isolation, purification and identification, several strains of bacteria were isolated from the samples successfully. By Transmission Electron Microscopy (TEM) and Energy-Dispersive X-ray Analysis (EDXA), these bacteria were certificated to be magnetotactic bacteria. The phylogenetic relationship between the isolated magnetic strains and some known magnetotactic bacteria was inferred by the construction of phylogenetic tree based on 16SrDNA sequences. This apparatus has been proven to have the advantages of being inexpensive, simple to assemble, easy to perform and highly efficient to isolate novel magnetotactic bacteria. The research indicated that the combined approach of harvesting MTB by home-made apparatus and the method of plate colony isolation could purify and isolate magnetotactic bacteria effectively. 相似文献

The oxidation state of the Earth's surface is one of the most obvious indications of the effect of life on this planet. The surface of Mars is highly oxidized, as evidenced by its red color, but the connection to life is less apparent. Two possibilities can be considered. First, the oxidant may be photochemically produced in the atmosphere. In this case the fundamental source of O2 is the loss of H2 to space and the oxidant produced is H2O2. This oxidant would accumulate on the surface and thereby destroy any organic material and other reductants to some depth. Recent models suggest that diffusion limits this depth to a few meters. An alternative source of oxygen is biological oxygen production followed by sequestration of organic material in sediments--as on the Earth. In this case, the net oxidation of the surface was determined billions of years ago when Mars was a more habitable planet and oxidative conditions could persist to great depths, over 100 m. Below this must be a compensating layer of biogenic organic material. Insight into the nature of past sources of oxidation on Mars will require searching for organics in the Martian subsurface and sediments. 相似文献

Plastic pellets found stranded on beaches are hydrophobic organic materials and thus, they are a favourable medium for persistent organic pollutants to absorb to. In the present study, plastic pellets are used to determine the diffuse pollution of selected Greek beaches. Samples of pellets were taken from these beaches and were analyzed for PCBs, DDTs, HCHs, and PAHs. The observed differences among pellets from various sampling sites are related to the pollution occurring at each site. Plastic pellets collected in Saronikos Gulf beaches demonstrate much higher pollutant loading than the ones collected in a remote island or close to an agricultural area. Based on data collected in this study and the International Pellet Watch program, pollution in Saronikos Gulf, Greece, is comparable to other heavily industrialized places of the world. The present study demonstrates the potential of pellet watch to be utilized as a detailed-scale monitoring tool within a single country. 相似文献
