Decapterus maruadsi is a commercially important species in China, but has been heavily exploited in some areas. There is a growing need to develop microsatellites promoting its genetic research for the adequate management of this fishery resources. The recently developed specific-locus amplified fragment sequencing (SLAF-seq) is an efficient and high-resolution method for genome-wide microsatellite markers discovery. In this study, 28 905 microsatellites (mono- to hexa-nucleotide repeats) were identified using SLAF-seq technology, of which di-nucleotide was the most frequent (13 590, 47.02%), followed by mono-nucleotide (8 138, 28.15%), tri-nucleotide (5 727, 19.81%), tetra-nucleotide (1 104, 3.82%), pentanucleotide (234, 0.81%), and hexa-nucleotide (112, 0.39%). One hundred and thirty-two microsatellite loci (di- and tri-nucleotide) were randomly selected for amplification and polymorphism, of which 49 were highly polymorphic and well-resolved. The average number of alleles per locus was 13.63, ranging from 4 to 25, and allele sizes varied between 110 bp and 309 bp. The observed heterozygosity ( Ho ) and expected heterozygosity ( He ) ranged from 0.233 to 1.000 and from 0.374 to 0.959, with mean values of 0.738 and 0.836, respectively. The polymorphism information content (PIC) ranged from 0.341 to 0.941 (mean=0.806). However, 12 loci deviated from Hardy-Weinberg equilibrium. Furthermore, transferability tests were also successful in validating the utility of the developed markers in five phylogenetically related species of family Carangidae. A total of 48 microsatellite markers were successfully cross-amplified in Decapterus macarellus, Decapterus macrosoma, Decapterus kurroides, Trachurus japonicus, and Selaroides leptolepis. The present microsatellites provided the first known set of microsatellite DNA markers for D. maruadsi, D. macarellus, D. kurroides, and D. macrosoma, and would be useful for further population genetic and molecular phylogeny studies as well as help with the fisheries management formulation and implementation of the understudied species.
Lidar (laser scanning) technology has been proven as a prominent technique for the acquisition of high-density and accurate topographic information. Because of systematic errors in the lidar measurements (drifts in the position and orientation information and biases in the mirror angles and ranges) and/or in the parameters relating the system components (mounting parameters), adjacent lidar strips may exhibit discrepancies. Although position and orientation drifts can have a more significant impact, these errors and their impact do not come as a surprise if the quality of the GPS/INS integration process is carefully examined. Therefore, the mounting errors are singled out in this work. The ideal solution for improving the compatibility of neighbouring strips in the presence of errors in the mounting parameters is the implementation of a rigorous calibration procedure. However, such a calibration requires the original observations, which may not be usually available. In this paper, a strip adjustment procedure to improve the compatibility between parallel lidar strips with moderate flight dynamics (for example, acquired by a fixed-wing aircraft) over an area with moderately varying elevation is proposed. The proposed method is similar to the photogrammetric block adjustment of independent models. Instead of point features, planar patches and linear features, which are represented by sets of non-conjugate points, are used for the strip adjustment. The feasibility and the performance of the proposed procedure together with its impact on subsequent activities are illustrated using experimental results from real data. 相似文献
Estuaries are elementary geochemical fronts where river water and seawater mix. Within this mixing zone, iron and other non-conservative
elements can undergo complex reactions to form new solid phases. In order to understand authigenic iron oxide formation in
the Yangtze River Estuary, two onsite water-mixing sets of experiments were conducted, one by mixing variable amounts of unfiltered
Yangtze River water with filtered East China Sea water of different salinity (set 1), the other by mixing variable amounts
of filtered Yangtze River water with filtered East China Sea water of different salinity (set 2). In set 2, the minerals newly
formed in the course of mixing were investigated by means of a scanning electron microscope fitted with an energy-dispersive
X-ray analytical system. It was found that ferrihydrite and lepidocrocite were formed in these mixing experiments, coexisting
in nearly equal amounts. These iron oxides appear as aggregated particles with a large grain-size range of several microns
to more than 100 μm. The electrolytic properties of seawater played an important role in the formation of these authigenic
iron oxides. Kaolinite and organic aggregates were also found in the experimentally mixed pre-filtered waters. Amounts of
newly formed suspended matter (set 2) were one to three orders of magnitude lower than those of total suspended matter (TSM)
(set 1). This implies that newly formed minerals represent only a very small proportion of TSM in the estuarine mixing zone

Cold seeps are pervasive along the continental margin worldwide,and are recognized as hotspots for elemental cycling pathway on Earth.In this study,analyses of pore water geochemical compositions of one-400 cm piston core(S3) and the application of a mass balance model are conducted to assess methane-associated biogeochemical reactions and uncover the relationship of methane in shallow sediment with gas hydrate reservoir at the Makran accretionary wedge off Pakistan.The results revealed that approximately 77% of sulfate is consumed by the predominant biogeochemical process of anaerobic oxidation of methane.However,the estimated sulfate-methane interface depth is-400 cm below sea floor with the methane diffusive flux of 0.039 mol/(m~2·a),suggesting the activity of methane seepage.Based on the δ~(13)C_(DIC) mass balance model combined with the contribution proportion of different dissolved inorganic carbon sources,this study calculated the δ~(13)C of the exogenous methane to be-57.9‰,indicating that the exogenous methane may be a mixture source,including thermogenic and biogenic methane.The study of pore water geochemistry at Makran accretionary wedge off Pakistan may have considerable implications for understanding the specific details on the dynamics of methane in cold seeps and provide important evidence for the potential occurrence of subsurface gas hydrate in this area. 相似文献
This paper theoretically investigates the high-order harmonic generation cutoff extension using intense few-cycle linearly chirped laser pulses. It shows that the cutoff of the harmonic can be extended remarkably by optimising the chirping parameters. The time-frequency characteristics of high-order harmonics with different chirping parameters are analysed by means of wavelet transform of the dipole acceleration. It also gives out the classical three-step model pictures of electron. By superposing a properly selected range of the harmonic spectrum, it obtains an isolated 65as pulse. 相似文献
