cDNA clones for glutathione S-transferases A (GST-A) and A1 (GST-A1) from plaice (Pleuronectes platessa) were expressed as N-terminally 6XHis tagged proteins in Escherichia coli and purified to homogeneity from Ni-NTA silica. GST-A was an efficient catalyst for conjugation of unsaturated alkenals derived from peroxidation of polyunsaturated fatty acids with the highest activity observed with trans-non-2-enal (8 micromol min(-1) mg(-1)). GST-A1 was a very efficient Se-independent glutathione peroxidase with an activity towards cumene hydroperoxide of 25 micromol min(-1) mg(-1). Although the enzymes exhibited moderately high activities towards the model substrate 1-chloro-2,4-dinitrobenzene (CDNB) they exhibited little or no activity towards other common prototypical xenobiotic substrates. Together with data for ontogeny, tissue distribution and inducibility of these enzymes, we contend that a primary function of these enzymes is protection from the harmful effects of lipid peroxidation products generated naturally or exacerbated by xenobiotic exposure.  相似文献

The Acid-Producing Potential (APP) and the Acid-Consuming Capacity (ACC) are introduced as basic parameters for long-term pollution assessment of mud disposal. They can be obtained by a four-step sequential leaching technique. The concentration of Ca extracted by the first Na-acetate step permits calculating the ACC. Both the Fe- and S-fractions deliberated by subsequent leaching steps are used to calculate the APP. When the APP of a sample is substacted from its ACC, a negative value indicates a potential increase of the bioavailability of the toxic metal load upon disposal of this mud in oxic environments.  相似文献

The spin up and relaxation of an autumn upwelling event on the Beaufort slope is investigated using a combination of oceanic and atmospheric data and numerical models. The event occurred in November 2002 and was driven by an Aleutian low storm. The wind field was strongly influenced by the pack-ice distribution, resulting in enhanced winds over the open water of the Chukchi Sea. Flow distortion due to the Brooks mountain range was also evident. Mooring observations east of Barrow Canyon show that the Beaufort shelfbreak jet reversed to the west under strong easterly winds, followed by upwelling of Atlantic Water onto the shelf. After the winds subsided a deep eastward jet of Atlantic Water developed, centered at 250 m depth. An idealized numerical model reproduces these results and suggests that the oceanic response to the local winds is modulated by a propagating signal from the western edge of the storm. The disparity in wave speeds between the sea surface height signal—traveling at the fast barotropic shelf wave speed—versus the interior density signal—traveling at the slow baroclinic wave speed—leads to the deep eastward jet. The broad-scale response to the storm over the Chukchi Sea is investigated using a regional numerical model. The strong gradient in windspeed at the ice edge results in convergence of the offshore Ekman transport, leading to the establishment of an anti-cyclonic gyre in the northern Chukchi Sea. Accordingly, the Chukchi shelfbreak jet accelerates to the east into the wind during the storm, and no upwelling occurs west of Barrow Canyon. Hence the storm response is fundamentally different on the Beaufort slope (upwelling) versus the Chukchi slope (no upwelling). The regional numerical model results are supported by additional mooring data in the Chukchi Sea.  相似文献

A small, inexpensive, and easily deployable meteorological buoy is described. Buoy motion is greatly reduced by appropriate ballast techniques; vector averaging further removes buoy motion effects from wind data. Data is transmitted to the GOES satellite and is retrieved by telephone. Measurements are vector-averaged wind components, wind speed, wind direction, water temperature, air temperature, and compass direction. Data from two field trials are discussed. Speed comparisons averaged 0.2 m sec−1 with a standard deviation of 0.6 m sec−1. Direction comparisons were different due to local topography, but they indicate a probable accuracy of ±5°.  相似文献

To assess whether life existed on Mars, it is crucial to identify geochemical biosignatures that are relevant to specific Martian environments. In this paper, thermochemical modeling was used to investigate fluid chemistries and secondary minerals that would have evolved biotically over geological time scales in Martian fluvio-lacustrine and evaporitic settings, and that could be used as potential inorganic biosignatures for life detection on Mars. Modeling was performed using fluid and rock chemistries relevant to Gale crater aqueous environments. Potential inorganic biosignatures were identified investigating alteration deposits found at the surface of a simulant exposed to short-term bio-mediated weathering and comparing experimental and modeling results. In a fluvio-lacustrine setting (water/rock of 2000–278), models suggest that less complex mineral assemblages form during biotic basalt dissolution and subsequent brine evaporation compared to what would happen in an abiotic system. Mainly nontronite, kaolinite, and quartz form under biotic conditions, whereas celadonite, talc, and goethite would also precipitate abiotically. Quartz, sepiolite, and gypsum would precipitate from the evaporation of fluids evolved biotically, whereas nontronite, talc, zeolite, and gypsum would form in an abiotic evaporitic environment. These results could be used to distinguish products of abiotic and biotic processes, aiding the interpretation of data from Mars exploration missions.  相似文献

Increasing concentrations of atmospheric CO2 and other greenhouse gases are expected to contribute to a global warming. This paper examines the potential implications of a climatic change corresponding to a doubling of atmospheric concentrations of CO2 on crop production opportunities throughout Ontario, a major food producing region in Canada. The climate is projected to become warmer and drier, but the extent of these shifts are expected to vary from region to region within Ontario. The effect of this altered climate on crop yields and the area of land capable of supporting specific crops varies according to region, soil quality and crop type. Most notable are the enhanced opportunities for grains and oilseeds in the northern regions, and the diminished production prospects for most crops in the most southerly parts of Ontario.  相似文献

A period of minimum solar activity in April and May 1996 was used to analyze full-disk CaK spectroheliograms with the aim of deriving the intensity thresholds of the quiet network and the plages by applying the so-called inflexion point method. The average network intensity threshold is found to be influenced mainly by the seeing, whereas the average plage threshold shows an increasing trend from disk center towards the solar limb.
