We previously reported the characteristics of a ChgH-GFP transgenic medaka line that indicates estrogenic compound pollution in environmental water by the green fluorescence of their liver. Recently, we established four more lines. In this study, the characteristics of the five transgenic medaka lines were investigated. The intensity of reporter gene expression varied among transgenic lines and generally correlated well with the amount of integrated transgene in each line. Line-specific ectopic expression was also observed. However, the sensitivity to 17-beta estradiol did not differ among transgenic lines. Three transgenic lines are considered to be suitable as bio-indicators of estrogenic activity, due to the ease of observing green fluorescence in their livers. The transgenic lines can also detect the estrogenic activity of testosterone and 17-beta trenbolone at the nominal concentration of 30 and 100 microg/l, respectively.  相似文献

To comprehensively understand the Arctic and Antarctic upper atmosphere, it is often crucial to analyze various data that are obtained from many regions. Infrastructure that promotes such interdisciplinary studies on the upper atmosphere has been developed by a Japanese inter-university project called the Inter-university Upper atmosphere Global Observation Network (1UGONET). The objective of this paper is to describe the infrastructure and tools developed by IUGONET. We focus on the data analysis software. It is written in Interactive Data Language (IDL) and is a plug-in for the THEMIS Data Analysis Software suite (TDAS), which is a set of IDL libraries used to visualize and analyze satellite- and ground-based data. We present plots of upper atmospheric data provided by IUGONET as examples of applications, and verify the usefulness of the software in the study of polar science. We discuss IUGONET's new and unique developments, i.e., an executable file of TDAS that can run on the IDL Virtual Machine, IDL routines to retrieve metadata from the IUGONET database, and an archive of 3-D simulation data that uses the Common Data Format so that it can easily be used with TDAS.  相似文献

Laboratory measurements for compressional and shear wave velocities (Vp and Vs, respectively) and porosity were conducted with core samples from the Nobeoka Thrust Drilling Project (NOBELL) under controlled effective pressure (5–65 MPa at 5 MPa intervals) and wet conditions. Samples were classified according to deformation texture as phyllite, foliated cataclasite, or non‐foliated cataclasite. Measured values of Vp, Vs, and porosity are within a range of 5.17–5.57 km/s, 2.60–2.71 km/s, and 2.75–3.10 %, respectively, for phyllite; 4.89–5.23 km/s, 2.46–2.57 km/s, and 3.58–4.53 %, respectively, for foliated cataclasite; and 4.90–5.32 km/s, 2.51–2.63 km/s, and 3.79–4.60 %, respectively, for non‐foliated cataclasite, which are all consistent with the previous laboratory experiments conducted with outcrop samples under dry conditions. However, our results also indicate higher Vp and Vs and lower porosity than those measured by the previous studies that adopted the wire‐line logging methods. The variations in Vp, Vs, and porosity are controlled by deformation structure and are greater for phyllite and foliated cataclasite than for non‐foliated cataclasite.  相似文献

Intensive groundwater development in the urban area of the Nagaoka Plain, Japan, has induced changes in the pH and saturation index of calcite in groundwater. To account for these chemical changes, it is important to determine seasonal variations of recharge and the groundwater flow system in the aquifer. This study identified the sources and flow system of groundwater in this urban area by a comprehensive method using stable isotope data and a numerical groundwater model of the Nagaoka Plain. Stable isotope evidence shows that the groundwater is recharged by meteoric water originating from low‐elevation areas rather than the mountains surrounding the plain. The water table in the study area is drawn down during the winter and recovers in the other seasons. Numerical modeling shows that discharge occurs primarily along the Shinano River during the recovery period, whereas discharge is centered in urbanized areas during the drawdown period, when a conical depression of the water table stimulates recharge from the immediate area. These results are indications of a local groundwater flow system, with its recharge area between the Shinano River and the urban areas, which is governed by intensive seasonal groundwater extraction.  相似文献

Recent studies have shown that base-isolated objects with long fundamental natural periods are highly influenced by long-period earthquakes. These long-period waves result in large displacements for isolators, possibly leading to exceedance of the allowable displacement limits. Conventional isolation systems, in general, fail to resist such large displacements. This has prompted the need to modify conventional base isolation systems. The current work focuses on the development of an external device, comprising a unit of negative and positive springs, for improving the performance of conventional base isolation systems. This unit accelerates the change in the stiffness of the isolation system where the stiffness of the positive spring varies linearly in terms of the displacement response of the isolated objects. The target objects of the present study are small structures such as computer servers, sensitive instruments and machinery. Numerical studies show that the increase in the damping of the system and the slope of the linear function is effective in reducing the displacement response. An optimal range of damping values and slope, satisfying the stability condition and the allowable limits of both displacement and acceleration responses when the system is subjected to near-fault and long-period ground motions simultaneously, is proposed.

We report several biogeochemical parameters (dissolved inorganic carbon (DIC), total alkalinity (TA), dissolved oxygen (DO), phosphate (PO4), nitrate + nitrite (NO3 + NO2), silicate (Si(OH)4)) in a region off Otaru coast in Hokkaido, Japan on a “weekly” basis during the period of April 2002–May 2003. To better understand the long-term temporal variations of the main factors affecting CO2 flux in this coastal region and its role as a sink/source of atmospheric CO2, we constructed an algorithm of DIC and TA using other hydrographic properties. We estimated the CO2 flux across the air–sea interface by using the classical bulk method. During 1998–2003 in our study region, the estimated fCO2sea ranged about 185–335 μatm. The maximum of fCO2sea in the summer was primarily due to the change of water temperature. The minimum of fCO2sea in the early spring can be explained not only by the change of water temperature but also the change of nutrients and chlorophyll-a. To clarify the factors affecting fCO2sea (water temperature, salinity, and biological activity), we carried out a sensitivity analysis of these effects on the variation of fCO2sea. In spring, the biological effect had the largest effect for the minimum of fCO2sea (40%). In summer, the water temperature effect had the largest effect for the maximum of fCO2sea (25%). In fall, the water temperature effect had the largest effect for the minimum of fCO2sea (53%). In winter, the biological effect had the largest effect for the minimum of fCO2sea (35%).We found that our study region was a sink region of CO2 throughout a year (−0.78 mol/m2/yr). Furthermore, we estimated that the increase of fCO2sea was about 0.56 μatm/yr under equilibrium with the atmospheric CO2 content for the period 1998–2003, with the temporal changes in the variables (T, S, PO4) on fCO2sea, thus as the maximum trend of each variable on fCO2sea was 0.22 μatm/yr, and the trend of residual fCO2 including gas exchange was 0.34 μatm/yr. This result suggests that interaction among variables would affect gas exchange between air and sea effects on fCO2sea. We conclude that this study region as a representative coastal region of marginal seas of the North Pacific is special because it was measured, but there is no particular significance in comparison to any other area.  相似文献

The temporal evolution of a thin phytoplankton layer was observed by field measurements using a research vessel and mooring instruments in the Yatsushiro Sea, a semi-enclosed narrow embayment in Japan, in early August 2013. The subsurface chlorophyll maximum developed into a thin layer within 2 days just below the pycnocline at around 10-m depth, where turbulent mixing (the dissipation rate of turbulent kinetic energy) was weak (low). The layer persisted for 1.5 to 2 days and declined after irradiance drastically decreased at the sea surface. At the peak period, the layer thickness, which is defined as the full-width at half-maximum of the peak in chlorophyll a concentration, ranged from 0.6 to 1.4 m, and the maximum concentration reached 42.3 mg m?3. The horizontal extent of the layer was approximately 10 km along the longitudinal axis of the bay. The phytoplankton population characterized by the layer was dominated by a chain-forming centric diatom, Chaetoceros spp. The formation mechanisms of the thin diatom layer were investigated using the observed data and a vertical one-dimensional model that includes physical and biological processes. The results suggest that the development of the thin layer was caused by in situ growth and aggregation due to nutrient-dependent sinking of the species under weak turbulence. The study highlights that continuous multidisciplinary observations and understanding species-specific physiological responses to environmental variations are necessary to elucidate drastically fluctuating phytoplankton dynamics in a coastal water.  相似文献

We estimated gas exchange rates in Kabira Reef at Ishigaki Island, southwest Japan, using a mass balance calculation with dual “biological” tracers: dissolved inorganic carbon (DIC) and dissolved oxygen (DO). The nighttime results allowed us to obtain reasonable gas transfer velocity k w values, all of which exceeded those obtained in wind-dominant studies. The difference is likely due to the contribution of turbulence generated by the interaction between the current and bottom topography. The k w obtained during high tides is consistent with that reported by Raymond and Cole (2001), whereas k w during low tides is significantly higher, which seems to be caused by enhanced friction with the bottom of the reef and/or bubble-induced gas transfer by wave breaking at the reef crest.  相似文献

On 26 May 1983 the rivers of the Japanese Islands were invaded by a tsunami from the Sea of Japan. Water levels in five large rivers were measured and were highest midway towards the uppermost point of inundation. The level at the uppermost point was approximately the same as that at the river mouths. As a result of resonance, a standing wave develops in the rivers. The period of the invading tsunami was found to be 80 min. This continental shelf oscillation resulted in a long-wave propagation of the tsunami.The water levels showed another peak in the neighbourhood of estuarine inflows. The period of this wave was estimated to be 20 min; its formation was attributed to generation near the tsunami source.The cross-section and longitudinal profiles of the rivers, together with water flow, complicated the observed water level profiles.  相似文献
