The potential of benzo[ a]pyrene (BaP) quinones (diones) to stimulate NAD(P)H-dependent hydroxyl radical (·OH) production (2-keto-4-methiolbutyric acid (KMBA) oxidation) by digestive gland microsomes of M. edulis was studied using iron/EDTA as a promotor of the Haber-Weiss reaction (O2− + H2O2 = ·OH + OH− + O2). Stimulation of basal rates of KMBA oxidation was observed for all three diones. Stimulation was similar for the 1,6- and 3,6-diones, but much less for the 6,12-dione, and greater for the NADH than the NADPH-dependent reaction, viz. % increases of 78 to 333 (NADH) compared to 0 to 78 (NADPH). Maximal KMBA oxidation was obtained at dione concentrations of 12.5 to 50 μm. Inhibition of KMBA oxidation by Superoxide dismutase and catalase indicated the involvement of respectively Superoxide anion radical (O2−) and hydrogen peroxide (H2O2) in 1,6-dione stimulated NADPH-dependent · OH formation. The apparent Km values of xenobiotic-stimulated KMBA oxidation were lower for BaP-diones than for the model redox cycling quinone menadione (2-methyl-1,4-naphthoquinone), viz. in μm, 0.6 to 14.6 (NADH) and 4.4 to 28.5 (NADPH) compared to 315 to 457 (NADH) and 72 to 103 (NADPH). The results are consistent with metabolism of BaP to diones and resultant enhanced generation of oxyradicals being a potential mechanism of pollutant-mediated toxicity in molluscs. 相似文献

The modulation of the mutagenicity of 4-nitroquinoline N-oxide (4-NQO) by M. edulis digestive gland cytosolic and microsomal fractions was studied by monitoring the induction of the umu operon by 4-NQO in Salmonella typhimurium 1535/pSK1002. The mutagenicity of 4-NQO was increased by M. edulis cytosol and microsomes in dose-dependent manner with respect to protein and toxicant concentration. The mutagenic enhancement reflected increased NAD(P)H-dependent oxygen consumption by microsomes in the presence of 4-NQO and enhanced nitroreductase activity in cytosol. The former was stimulated by azide, indicating 4-NQO-enhanced oxyradical production by microsomes, and the latter was inhibitable by dicoumarol, indicating involvement of DT-diaphorase in nitroreduction by M. edulis cytosol. Neither dicoumarol nor allopurinol (inhibitors of DT-diaphorase and xanthine oxidase, respectively) affected the enhanced mutagenic expression of 4-NQO by cytosol. The results support the possibility of oxidative stress as a pollution-mediated mechanism of toxicity. 相似文献

The current benchmark approach for mathematical modelling of floodplain hydrologic regime consists of dynamically coupling one‐dimensional (1D) and two‐dimensional (2D) models for flow routing along the main channel and the floodplain, respectively. For large‐scale sites, floodplain inundation may spread over hundreds of square kilometres and may last for many months and even influence seasonal floods in following years. This paper aims at evaluating the effect of vertical water balance representation on modelling a large‐scale floodplain. The Pantanal wetland (140 000 km2; Brazil) is simulated using a 1D/2D coupled model approach, which also considers the representation of vertical water processes over the floodplain. Four scenarios are simulated: Baseline (the reference scenario), NoVertBal (in which the vertical water balance over floodplain is turned off) and ETp+1 and ETp?1 scenarios, characterized by artificially increasing or decreasing daily potential evapotranspiration (ETp) by 1 mm, respectively. The results showed that the effect of the vertical water processes scenarios on channel flow is directly dependent on the lateral exchange of water between the channel and floodplain in the upstream river reach. This influence is stronger when there is a gain of water from the floodplain to the channel. The inclusion of these vertical water processes into floodplain modelling was essential to represent the process of wetting and drying, this effect being more relevant for areas not directly connected to main channels, which is a characteristic of the Pantanal region. The conclusion that tektite falls were restricted in time to an earlier era in Earth history is supported by all known facts, in particular, by the results of critical examinations of various reported falls, including 2 australite falls described not long ago by the late Dr. E. S. Simpson. On the contrary, if the tektites originate as “lunar impactites,” as Dr. H. H. Nininger has recently suggested, it seems evident that they would have continued to fall onto the Earth right up to the present time. Furthermore, the probability that the distribution of “lunar impactite” falls over the Earth's surface would exhibit the alinement along 3 great circles, which is characteristic of all known tektite deposits, is vanishingly small. 相似文献

Sea ice has been suggested, based on simple models, to play an important role in past glacial–interglacial oscillations via the so-called “sea-ice switch” mechanism. An important requirement for this mechanism is that multiple sea-ice extents exist under the same land ice configuration. This hypothesis of multiple sea-ice extents is tested with a state-of-the-art ocean general circulation model coupled to an atmospheric energy–moisture-balance model. The model includes a dynamic-thermodynamic sea-ice module, has a realistic ocean configuration and bathymetry, and is forced by annual mean forcing. Several runs with two different land ice distributions represent present-day and cold-climate conditions. In each case the ocean model is initiated with both ice-free and fully ice-covered states. We find that the present-day runs converge approximately to the same sea-ice state for the northern hemisphere while for the southern hemisphere a difference in sea-ice extent of about three degrees in latitude between the different runs is observed. The cold climate runs lead to meridional sea-ice extents that are different by up to four degrees in latitude in both hemispheres. While approaching the final states, the model exhibits abrupt transitions from extended sea-ice states and weak meridional overturning circulation, to less extended sea ice and stronger meridional overturning circulation, and vice versa. These transitions are linked to temperature changes in the North Atlantic high-latitude deep water. Such abrupt changes may be associated with Dansgaard–Oeschger events, as proposed by previous studies. Although multiple sea ice states have been observed, the difference between these states is not large enough to provide a strong support for the sea-ice-switch mechanism. 相似文献

ABSTRACTThe spread of impervious surfaces in urban areas combined with the rise in the intensity of rainfall events as a result of climate change has led to dangerous increases in storm water flows. This paper discusses a new implementation of the fully distributed hydrological model Multi-Hydro (developed at École des Ponts ParisTech), when operating storage basins, and its ability to deal with high-resolution radar rainfall data. The peri-urban area of Massy (south of Paris, France) was selected as a case study for having six of these drainage facilities, used extensively in flood control. Two radar rainfall datasets with different spatiotemporal resolutions were used: Météo-France’s PANTHER rainfall product (C-band) and ENPC’s X-band DPSRI. The rainfall spatiotemporal variability was analysed statistically using Universal Multifractals (UM). Finally, to validate the application, the water level simulations were compared with local measurements in the Cora storage basin located next to the catchment’s single outlet. 相似文献

Abstract We evaluate a set of current measurements done in a section of the Strait of Juan de Fuca. The flow is of estuarine character, the upper layer flow usually being directed seaward. The RMS value of steady current exceeds its mean value appreciably in the upper layer; it also exceeds the mean near the bottom. The near‐surface currents do change their direction on occasions and can run landward for over five consecutive days, especially in the southern part of the channel. The lower layer flow (landward) and the upper layer flow (seaward) varied in magnitude from 90 to 160 thousand m3/sec, and their fluctuations were in phase; their difference, the net flow, is of the order of 5% to 10% of these flows and could not be calculated with any confidence. Tidal motion is barotropic in the section, with some deformation in the vertical caused by bottom friction and internal stresses. The M2 tide in the system Juan de Fuca‐Georgia Strait can be represented by a standing Kelvin wave influenced by friction. The K1 tide can also be represented by a standing Kelvin wave if some leakage is allowed in the northern end of Georgia Strait. Topographic effects mask the Coriolis influence on the intensity of tidal currents. 相似文献

This study presents the distribution of dissolved inorganic carbon (DIC) along the Strait of Gibraltar, its tidal-induced variability, as well as the inorganic carbon exchange between the Atlantic Ocean and Mediterranean Sea. During November 2003, water column samples were collected at nine stations to measure total alkalinity (TA), pH, and dissolved oxygen (DO) for the spatial characterization of the carbonate system. At the same time, anchored samplings were carried out, above the Camarinal Sill and in the Eastern Section of the Strait, in order to assess the tidal mixing effects for oxygen and DIC distribution on the water column. Three distinct water masses can be discerned in this area: the Surface Atlantic Water (SAW), the Mediterranean Water (MW), and the less abundant North Atlantic Central Water (NACW). The observations show an increase in the DIC and a decrease in oxygen concentration with depth, related to the different physico-chemical features of each water mass. The results show the high time-dependence of the vertical distribution of DIC with the interface oscillation, affected by the intense mixing processes taking place in the Strait. Intense mixing episodes over the Camarinal Sill are responsible for an increase in the DIC concentrations in the upper layer of the Eastern Section of the Strait. Higher DIC concentrations in the Mediterranean than in the Atlantic waters are responsible for a net DIC transport of 1.47×1012 mol C yr−1 to the Atlantic Ocean. The monitoring of bedload flux under flash flood conditions has been successfully achieved since 1992 using slot samplers in the semiarid Nahal Eshtemoa. In the present study, a surrogate bedload monitoring technique - the Japanese plate microphone - has been deployed and calibrated against data from the slot samplers. Since a slot sampler has a sensitivity threshold that becomes especially important when transport rates are low, different averaging periods should be considered for high and low fluxes. In order to overcome the deficiencies of time-based aggregation used hitherto, we have developed a new method involving mass aggregation and commensurably variable intervals, thereby enabling a more accurate analysis and optimizing the bedload sampler's capabilities. The data derived with this new method has then been utilized to calibrate the Japanese plate microphone. The Eshtemoa is an ephemeral gravel bed channel with a high proportion of fine gravel (< 0.02 m); for these conditions, acoustic sensors have not been calibrated as yet. Two multiple linear regression models incorporating the effect of median bedload grain size on pulse rate have been established to predict bedload flux and cumulative transported bedload mass. The coefficients in these models are statistically significant. Good predictions are obtained for bedload flux (adj. r2 = 0.83) and for cumulative bedload mass (adj. r2 = 0.98) during flood recession. Overall, the multiple linear regression models, used in conjunction with the mass aggregation method of estimating bedload flux, suggest that field calibration of acoustic devices is feasible under these conditions for ca. 90% of the duration of bedload transport. © 2020 John Wiley & Sons, Ltd. 相似文献
