Release into the environment of metals by two vascular salt marsh plants

Metals in contaminated salt marshes are mainly locked in the anaerobic layer of sediments, where they are tightly bound as sulfides and organic complexes. Vascular plants survive in saturated soils in part by pumping O2 into their root zones, changing their microenvironment to an oxic one. This, along with chelating exudates, mobilizes metals, allowing uptake by the roots. We compared the common reed Phragmites australis and cordgrass Spartina alterniflora in lab and field studies for ways in which they handle trace metals. Both plants store most of their metal burden in their roots, but some is transported to aboveground tissues. Spartina leaves contain approximately 2-3 x more Cr, Pb, and Hg than Phragmites leaves, but equivalent Cu and Zn. Furthermore, Spartina leaves have salt glands, so leaf excretion of all metals is twice that of Phragmites. In-depth studies with Hg indicate that Hg excretion correlates with Na release but not with transpiration, which is 2.2 x higher in Phragmites; and that more Hg accumulates in early-appearing leaves than in upper (i.e. later) leaves in both species. Spartina thus makes more metals available to salt marsh ecosystems than Phragmites by direct excretion and via dead leaves which will enter the food web as detritus.     

Evaluation of acetylcholinesterase activity in several zooplanktonic crustaceans

Zooplankton are an essential component of the marine and estuarine food chains. The ecotoxicological risk to zooplanktonic communities, estimated through the use of the biomarkers, can be used as an early warning signal of a potential alteration of the ecosystem health. The aim of this project was to evaluate the potential use of several biomarkers (esterases, mixed function oxidases, porphyrins) in zooplanktonic organisms. The acetylcholinesterase activity (AChE) was determined in homogenates of whole organisms of 10 different zooplanktonic crustaceans. Mean activity of AChE was 10.05 micromol/min/g for Acartia margalefi; 3.30 for Acartia latisetosa; 79.70 for Siriella clausi; 49.97 for Diamysis bahirensis; 7.48 for Siriella armata; 14.20 for Mysidopsis gibbosa; 4.49 for Euphausia crystallorophias; 1.66 for Euphausia superba; 2.74 for Streetsia challengeri; 13.26 for Meganycthiphanes norvegica. The species moreover showed a linear increase in enzyme activity in relation to the increase in sample concentration. The key result of this study concerns the different AChE activity basal values of the crustaceans' different species.     

An aromatase inhibitor and tamoxifen decrease plasma levels of o,p'-DDT and its metabolites in Nile tilapia (Oreochromis niloticus)

The objective of this study was to determine if tamoxifen or an aromatase inhibitor (4-hydroxyandrostenedione; 4-OHA) affected plasma concentrations of o,p' -DDT and its metabolites, o,p'-DDD and o,p'-DDE, in mature tilapia. Male and female tilapia were injected 6 times intraperitoneally with o,p'-DDT (40 mg/kg) alone or in combination with 4-OHA (2 mg/kg) over a 12 day period. An additional group of male fish was injected with tamoxifen (5 mg/kg) plus o,p'-DDT. At the end of the treatment period, plasma samples were extracted and analyzed by GC/ECD. Females injected only with o,p'-DDT had significantly higher levels of o,p'-DDT compared with males. Interestingly, females and males treated concomitantly with o,p'-DDT and 4-OHA or tamoxifen had significantly lower concentrations of plasma o,p'-DDT (about 50%) compared with fish treated with only o,p'-DDT. These initial results suggest that an interaction between endocrine-active compounds occurs in vivo in tilapia and may involve alterations in metabolism of o,p'-DDT.

Development of two novel CYP-antibodies and their use in a PCB exposure experiment with Mytilus edulis

In an attempt to learn more about the cytochrome P450 (CYP) system of mussels, we used protein databases and alignment software to extract highly conserved CYP sequences. From these alignments synthetic peptides were produced and used for rabbit immunisation, which yielded polyclonal antibodies against the CYP families 2 and 4. The antibodies were evaluated with Western Blot and ELISA assays, using digestive gland microsomal samples from the mussel Mytilus edulis. Western Blots revealed immunoreactions for both antibodies. The anti-CYP2 sequence rendered one major immunopositive protein of approximately 49 kDa size, and weak signals for proteins of approximately 41 and 56 kDa size. The anti-CYP4 sequence rendered two major bands of approximately 56 and 59 kDa size, and also a weak immunoreaction with a protein of approximately 43 kDa size. ELISA rendered only weak signals even with a 1:50 dilution of IgG-purified serum. A 10-day exposure to Aroclor 1254 did not appear to affect any of the immunopositive proteins, while total PCBs in soft bodies increased from 14-40 ng/g DW in controls to 373-638 ng/g DW in exposed mussels.     

Use of suppressive subtractive hybridization and cDNA arrays to discover patterns of altered gene expression in the liver of dihydrotestosterone and 11-ketotestosterone exposed adult male largemouth bass (Micropterus salmoides)

In this study male largemouth bass (LMB) were exposed to the naturally occurring androgens, dihydrotestosterone (DHT) or 11-ketotestosterone (11-KT) in order to identify genes that are differentially regulated by these steroid hormones. Using subtractive hybridization on livers of fish treated with DHT against vehicle control, many novel LMB genes were cloned. These genes were added to our gene library and arrayed. Six genes were up-regulated and five were down-regulated by both androgens. But, each androgen also regulated specific genes. One gene that was identified as a potential androgen marker was spermidine-spermine-N(1)-acetyltransferase that was up-regulated by both androgens. Determining which genes are responsive to natural androgens will help to identify biochemical pathways that are impacted.

双刺镰状水蚤Drepanopus bispinosus为南极沿海伯尔顿盐湖(Burton lake)中的越冬种类之一,其雌性性成熟大约于每年的初冬季节,在随后8个多月的生活史中,经历了一个盐度从33(5月份)到39(11月份)的环境变化.本文报道了该水蚤雌性成体对环境盐度适应的季节差异,动物在冬季的耐低盐能力较夏季强,从冬季到夏季,动物对盐度的适应范围明显地向高盐度方向偏移,这与环境盐度对动物的驯化有关.在低盐水中,动物的呼吸率增高,且夏季大于冬季,因而进一步证实了环境盐度的变化对动物盐度适应性的影响.     

Susceptibility to genetic damage and cell types in Mediterranean mussels

Micronucleus (MN) frequency is generally accepted as a marker of chromosomal damage and has been studied in a variety of cells and species. In previous work, we detected significant dose-related MN increases in the epithelial-like gill cells and agranular haemocytes of Mytilus galloprovincialis treated with benzo[a]pyrene, a well-known mutagenic pollutant. In addition, we have studied micronuclei and other nuclear abnormalities in mussels collected from the Venice lagoon (Italy). Frequency changes, possibly related to genotoxic/toxic stress, in both granular and micronucleated cells from gills and haemolymph, were detected. Environmental data suggest the effect of genotoxic pollutants and the importance of cell replication in the interpretation of micronucleus frequencies.
