β-mannanase is an enzyme that is commonly expressed in environmental bacteria. It degrades hemicellulose found in plant material and recycles nutrients back into the environment. Because this enzyme significantly contributes to biodegradation and has recently been applied in industry, we conducted a comparative analysis of bacterial isolates found in soil samples from Schirmacher Oasis, Antarctica, and Sabah, Malaysia that were capable of degrading mannan. A total of 9 bacterial isolates from Antarctica and 30 bacterial isolates from Malaysia exhibited β-mannanase activity. These bacteria were differentiated and clustered using their random amplified polymorphic DNA (RAPD) profiles, and the β-mannanase activity of these isolates was tested at different temperatures and pH. Five out of 9 Antarctica isolates and seven out of 30 Malaysian isolates were identified based on their 16S rDNA sequences. Identified bacterial isolates from Antarctica were: MP1 (Bacillus amyloliquefaciens), MP2 (Bacillus pumilus), MP5 (Bacilluspumilus), A40 (Arthrobacter sp.), and C27 (Arthrobacter oxydans). Identified bacterial isolates from Ma- laysia were: Y1 (Paenibacillus sp.), Y2 (Bacillus sp.), Y16 (Paenibacillus sp.), Y18 (Paenibacillus sp.), A7 (Paenibacillus sp.), B26 (Streptomyces sp.), and D4 (Paenibacillus amylolyticus). β-mannanases produced by the Antarctica bacterial isolates MP1 (Bacillus amyloliquefaciens) and A40 (Arthrobacter sp.) were active at 5℃ and 20℃, respectively, while the β-mannanase pro- duced by the bacterial isolate from Malaysia, A7 (Paenibacillus sp.), was active at 35 ℃. 相似文献

The Paleoproterozoic Jiao-Liao-Ji Belt lies in the Eastern Block of the North China Craton, with its southern segment extending across the Bohai Sea into the Jiaobei massif. High-pressure pelitic and mafic granulites have been recently recognized in the Paleoproterozoic Jingshan Group (Jiaobei massif). New SHRIMP U–Th–Pb geochronology combined with cathodoluminescence (CL) imaging of zircon has been applied to the determination of the timing of the metamorphism of the high-temperature and high-pressure granulites and associated gneisses and marbles. Metamorphic zircons in these high-pressure granulites, gneisses and marbles occur as either single grains or overgrowth (or recrystallization) rims surrounding and truncating oscillatory-zoned magmatic zircon cores. Metamorphic zircons are all characterized by nebulous zoning or being structureless, with high luminescence and relatively low Th/U values. Metamorphic zircons from two high-pressure mafic granulites yielded 207Pb/206Pb ages of 1956 ± 41 Ma and 1884 ± 24 Ma. One metamorphic zircon from a garnet–sillimanite gneiss also gave an apparent 207Pb/206Pb age of 1939 ± 15 Ma. These results are consistent with interval of ages of c. 1.93–1.90 Ga already obtained by previous studies for the North and South Liaohe Groups and the Laoling Group in the northern segment of the Jiao-Liao-Ji Belt. Metamorphic zircons from a high-pressure pelitic granulite and two pelitic gneisses yielded weighted mean 207Pb/206Pb ages of 1837 ± 8 Ma, 1821 ± 8 Ma and 1836 ± 8 Ma respectively. Two diopside–olivine–phlogopite marbles yielded weighted mean 207Pb/206Pb ages of 1817 ± 9 Ma and 1790 ± 6 Ma. These Paleoproterozoic metamorphic ages are largely in accordance with metamorphic ages of c. 1.85 Ga produced from the Ji'an Group in the northern segment of the Jiao-Liao-Ji Belt and c. 1.86–1.80 Ga obtained for the high-pressure pelitic granulites from the Jingshan Group in the southern segment. As this metamorphic event was coeval with the emplacement of A-type granites in the Jiao-Liao-Ji Belt and its adjacent areas, it is interpreted as having resulted from a post-orogenic or anorogenic extensional event. 相似文献

Mesoproterozoic (~ 1050 Ma; Stenian) zircon crystals from the Saranac Prospect, Bancroft, Ontario, contain up to ~ 1 wt.% U and ~ 0.15 wt.% Th and, correspondingly, they are for the most part extensively radiation-damaged (calculated total α-doses 2.3?35.3 × 1018/g). The crystals show textures of complex, intense chemical alteration that is attributed to multiple, low-T replacement events along fluid-controlled reaction fronts. Centers of crystals appear totally replaced; the primary zoning is virtually erased and the material has high porosity and numerous inclusions. Interior regions surrounding the central reworked areas still exhibit primary igneous-type zoning; in those regions the alteration emanates from fractures and then follows the more radiation-damaged growth zones. Altered areas are typically recognized by their high porosity, low BSE intensity, and deficient analytical totals. Those regions often have lost a significant fraction of their radiogenic Pb. They are in general somewhat depleted in Zr, Si, and U, and are notably enriched in Ca and Fe. Element maps reveal elevated concentrations of Al and Y within filled fractures. Our observations indicate that the fluid-driven ion exchange is mainly controlled by the accessibility of micro-areas with elevated levels of radiation damage to transporting fluids via “fast pathways”. Most importantly, there is apparent Zr?Si?U equilibrium between initially existing and newly formed zircon. The retention of U after the chemical replacement (94 ± 14% relative to the original U content in the respective zones) does not significantly fall below the retention of two major cations Zr (95 ± 4%) and Si (95 ± 2%). In spite of the partially extreme hydrothermal alteration overprinting, the original U zoning in the crystals is well preserved. These observations suggest that preferential chemical leaching of U from zircon is clearly not a general feature of this mineral. This in turn seems to question the general validity of hydrothermal experiments to low-T, fluid-driven alteration of zircon in geological environments. The observed apparent immobility of U may affect the interpretation of U?Pb discordance in zircon, and the performance assessment of this mineral as potential waste form for actinides. 相似文献

The flow, turbulence, and longitudinal dispersion in wave-current flow through submerged vegetation are experimentally examined.
Laboratory experiments are carried out by superimposing progressive waves on a steady flow through simulated submerged vegetation.
The resultant wave-current-vegetation interaction shows strong interface shear with increase in the velocity due to the wave-induced
drift. The increase in turbulence in the region of vegetation is found to be about twice higher than in the no-wave case due
to the additional mixing by wave motions. Solute experiments are conducted to quantify the wave-current-vegetation longitudinal
dispersion coefficient (WCVLDC) by the routing method and by defining length and velocity scales for the wave-current-vegetation
flow. An empirical expression for the WCVLDC is proposed. Although the increase in vertical diffusivity is observed as compared
with bare-bed channels, the shear effect is stronger, which increases the value of the WCVLDC. The study can be a guideline
to understand the combined hydrodynamics of waves, current, and vegetation and quantify the longitudinal dispersion therein.

Magnetic susceptibility (MS) of the Quaternary long-term mid-latitude Maros fluvial fan (Pannonian Basin) was recorded to understand the stratigraphical features of source-proximal fluvial depositional settings. Three fully cored 500-m-deep boreholes were sampled at 0.5-m intervals; low-field and frequency dependent MS were measured, and complementary hysteresis and SEM-EDAX investigations were performed on selected samples. Logged susceptibility data were also used to log correlations established by a simultaneous comparison of wireline log and laboratory measurements. Time-series analyses of the susceptibility records reveal a ~41-ka and ~100-ka cyclicity. Towards the source-distal sections the intensity of the ~41-ka cycles decreases, while that of the ~100-ka cycles remains strong. Stratigraphical and spectral similarities were observed between the Maros fluvial fan and Chinese loess records; however, based on complementary magnetic data, the magnetic phase of the Maros Fan sections is related to the detrital magnetite that originates from the catchment during early postglacial permafrost degradations. The amplification of the ~41-ka cycles can be attributed to the very high susceptibility values in source-proximal settings and to the special stratigraphical feature of the distributive fluvial systems. This comprises the increased avulsion frequency on the fluvial fans in ‘glacial recession periods’, in concert with the ‘early postglacial’ occurrence of the permafrost-related magnetite originating from the catchment. As a local phenomenon, this is significant since it records the obliquity-driven variations in permafrost development in a catchment. However, fluvial and alluvial fans are widespread depositional landforms within the Eurasian mountain range and were possibly the same during the Quaternary deglaciations. Thus, obliquity-driven magnetic susceptibility variations in source-proximal fan deposits attached or adjacent to regions of loess deposition should also be considered when scanning for potential source material of aeolian deposits. 相似文献

Mangroves commonly found along tropical and subtropical coastlines are susceptible to oil pollution. In December 2000, around 500 1 m tall Kandelia candel saplings at the age of 3–5 years old located at the foreshore region of Sheung Pak Nai swamp, Hong Kong SAR, were found to be damaged by oil pollution. More than 80% of the saplings were either dead or washed away and leaving less than 5% healthy saplings with dense green leaves. Elevated concentrations of light n-alkanes (ranging from n-C14 to n-C20), pristane and phytane were recorded in surface sediments collected in December 2000. The ratio between light and total n-alkanes was 0.4. The total petroleum hydrocarbons (60–80 μg g−1 TPH) and unresolved complex mixtures (60–70 μg g−1 UCM) were higher than the background values of other mangrove sediments in Hong Kong, which were 40 and 20 μg g−1, respectively. In certain root zone sediments, TPH concentrations were above 1000 μg g−1. These results suggest that surface sediments in Sheung Pak Nai were contaminated by petroleum oil, most likely by illegal discharge of fuel oil which occurred between 1998 and 2002. One year later, in December 2001, unhealthy saplings had recovered and re-grown. The concentrations of TPH and UCM in sediments declined to around 40 μg g−1, pristane and phytane dropped by 80%, and the ratio of light to total n-alkanes was 0.15, suggesting that residual oil in sediments was weathered leading to a remarkable recovery of the unhealthy saplings. 相似文献

The temporal responses of peroxidase (POD), superoxide dismutase (SOD), total polyphenols (TP), extractable condensed tannins (ECT) to different levels of lead (Pb) or manganese (Mn) stress, as well as the metal accumulation, in seedlings of Kandelia obovata were investigated. Both stress time and stress intensity had significant effects with significant accumulation of Pb and Mn in roots at Day 1. Pb and Mn showed no significant effects on root or leaf TP and ECT at Day 1. Prolonged exposure to metals caused significant drops of root TP and ECT but increased in leaves at Days 7 and 49. POD activities decreased in both roots and leaves at Day 1, while POD and SOD both increased under moderate levels of Pb and Mn at Day 7. The present study showed that antioxidative enzyme activities were more sensitive indicators to Pb or Mn stresses than phenolic compounds. 相似文献
