Existing methods for analyzing methylsulfonyl-PCBs in animal tissues require multiple laborious clean-up steps prior to quantitation. In this paper, we report a new, efficient method for the isolation and determination of methylsulfonyl-PCB metabolites in the blubber, liver, kidney, lung, and milk from mammals of marine origin using: solvent extraction, GPC fractionation, and clean-up on an adsorption chromatography column prior to analysis by gas chromatography-mass spectrometry. The method is rapid and can be completed in about half the analysis time required by most other reported methods. The method exhibits excellent analyte recoveries (89-95%), and good reproducibilities with CV's ranging from 3-12% depending on analyte concentration. Detection limits are approximately 1 ng/g lipid. The method is further validated by comparing its quantitative results to that of an independent reference analytical method for several different marine mammal blubber samples. Finally, the method is applied to profile the distribution of methylsulfones among the various tissue types studied for a single bottle-nose dolphin (Tursiops truncatus) sample. In addition, the extent of their transplacental transfer from mother to unborn fetus for several pilot whale (Globicephala melaena) mother-fetus pairs is examined. 相似文献

Aquitards protect underlying aquifers from contaminants and limit recharge to those aquifers. Understanding the mechanisms and quantity of ground water flow across aquitards to underlying aquifers is essential for ground water planning and assessment. We present results of laboratory testing for shale hydraulic conductivities, a methodology for determining the vertical hydraulic conductivity (K(v)) of aquitards at regional scales and demonstrate the importance of discrete flow pathways across aquitards. A regional shale aquitard in southeastern Wisconsin, the Maquoketa Formation, was studied to define the role that an aquitard plays in a regional ground water flow system. Calibration of a regional ground water flow model for southeastern Wisconsin using both predevelopment steady-state and transient targets suggested that the regional K(v) of the Maquoketa Formation is 1.8 x 10(-11) m/s. The core-scale measurements of the K(v) of the Maquoketa Formation range from 1.8 x 10(-14) to 4.1 x 10(-12) m/s. Flow through some additional pathways in the shale, potential fractures or open boreholes, can explain the apparent increase of the regional-scale K(v). Based on well logs, erosional windows or high-conductivity zones seem unlikely pathways. Fractures cutting through the entire thickness of the shale spaced 5 km apart with an aperture of 50 microns could provide enough flow across the aquitard to match that provided by an equivalent bulk K(v) of 1.8 x 10(-11) m/s. In a similar fashion, only 50 wells of 0.1 m radius open to aquifers above and below the shale and evenly spaced 10 km apart across southeastern Wisconsin can match the model K(v). 相似文献

As part of a multinational workshop on marine environmental quality, sediments were collected from seven sites in Vancouver Harbour and analyzed for polynuclear aromatic hydrocarbons (PAHs) and organochlorines (OCs), and for trace metals. English sole were collected from five sites, and muscle was analyzed for trace metals and liver for OCs. As expected, sediment PAH and OC concentrations and tissue OC concentrations were higher at sites east of the First Narrows, compared to the outer harbor and reference sites. Sediment PAH concentrations east of the First Narrows were similar to concentrations at moderately contaminated sites in Puget Sound, south of Vancouver Harbour. In contrast, concentrations of OCs in sediments and tissue were low to moderate, even at relatively contaminated sites within Vancouver Harbour. Although several trace metals in sediments were higher than in contaminated sediments from Puget Sound, trace metals measured in fish muscle were lower. 相似文献

Estimation of mass discharge has become an increasingly valuable analysis technique at sites with contaminated groundwater plumes. We propose a simple plume magnitude classification system based on mass discharge comprised of 10 separate magnitude categories, such as a "Mag 7 plume." This system can be a useful tool for scientists, engineers, regulators, and stakeholders to better communicate site conceptual models, prioritize sites, evaluate plumes both spatially and temporally, and determine potential impacts. 相似文献

Measurements of dissolved N2, O2, Ar, CO2, and CH4 were made in perennially ice-covered Lake Hoare. Results confirm previous reports that O2 concentrations in the upper water column exceed atmospheric equilibrium and that N2 and Ar are supersaturated throughout the water column. The mean supersaturation of N2 was found to be 2.0 (+/- 0.37) and Ar was 3.8 (+/- 1.1). The ratios of N2/Ar (20.3 +/- 13.8), and O2/Ar (22.5 +/- 4.0) at the ice-water interface are consistent with those previously measured, suggesting that bubble formation is the main process for removing gas from the lake. However, the saturations of N2 and Ar greatly exceed those previously predicted for degassing by bubble formation only at the ice-water interface. The data support the hypothesis that removal of gas by bubbles occurs in the water column to a depth of 11 m in Lake Hoare. CO2 concentration increases from near zero at the ice-water interface to 80-100 times saturation at and below the chemocline at c. 28 m. There is considerable variability in the gas concentrations throughout the water column; samples separated in depth by one metre may vary by more than 50% in gas content. It is likely that this phenomenon results from the lack of turbulent mixing in the water column. Methane (c. 2 micrograms l-1) was detected below the chemocline and immediately above the sediment/water interface at a depth of 30 m. Samples from lakes Vanda, Joyce, and Miers, also show supersaturations of O2, N2, and Ar at levels similar to levels found in Lake Hoare. 相似文献
